1Homing and migration of mesenchymal stromal cells: How to improve the efficacy of cell therapy?显示文摘Mesenchymal stromal cells(MSCs) are currently being investigated for use in a wide variety of clinical applications. For most of these applications, systemic delivery of the cells is preferred. However, this requires the homing and migration of MSCs to a target tissue. Although MSC hominghas been described, this process does not appear to be highly efficacious because only a few cells reach the target tissue and remain there after systemic administration. This has been ascribed to low expression levels of homing molecules, the loss of expression of such molecules during expansion, and the heterogeneity of MSCs in cultures and MSC culture protocols. To overcome these limitations, different methods to improve the homing capacity of MSCs have been examined. Here, we review the current understanding of MSC homing, with a particular focus on homing to bone marrow. In addition, we summarize the strategies that have been developed to improve this process. A better understanding of MSC biology, MSC migration and homing mechanisms will allow us to prepare MSCs with optimal homing capacities. 2Cryopreservation for delayed circulating tumor cell isolation is a valid strategy for prognostic association of circulating tumor cells in gastroesophageal cancer显示文摘AIM To demonstrate the feasibility of cryopreservation of peripheral blood mononuclear cells(PBMCs) for prognostic circulating tumor cell(CTC) detection in gastroesophageal cancer.METHODS Using 7.5 m L blood samples collected in EDTA tubes from patients with gastroesopheagal adenocarcinoma, CTCs were isolated by epithelial cell adhesion molecule based immunomagnetic capture using the Iso Flux platform. Paired specimens taken during the same blood draw(n = 15) were used to compare number of CTCs isolated from fresh and cryopreserved PBMCs. Blood samples were processed within 24 h to recover the PBMC fraction, with PBMCs used for fresh analysis immediately processed for CTC isolation. Cryopreservation of PBMCs lasted from 2 wk to 25.2 mo(median 14.6 mo). CTCs isolated from pre-treatment cryopreserved PBMCs(n = 43) were examined for associations with clinicopathological variables and survival outcomes.RESULTS While there was a significant trend to a decrease in CTC numbers associated with cryopreserved specimens(mean number of CTCs 34.4 vs 51.5, P = 0.04), this was predominately in samples with a total CTC count of > 50, with low CTC count samples less affected(P = 0.06). There was no significant association between the duration of cryopreservation and number of CTCs. In cryopreserved PBMCs from patient samples prior to treatment, a high CTC count(> 17) was associated with poorer overall survival(OS)(n = 43, HR = 4.4, 95%CI: 1.7-11.7, P = 0.0013). In multivariate analysis, after controlling for sex, age, stage, ECOG performance status, and primary tumor location, a high CTC count remained significantly associated with a poorer OS(HR = 3.7, 95%CI: 1.2-12.4, P = 0.03). 5Mapping summer soybean and corn with remote sensing on Google Earth Engine cloud computing in Parana state – Brazil显示文摘Brazilian farming influences directly the worldwide economy.Thus,fast and reliable information on areas sown with the main crops is essential for planning logistics and public or private commodity market policies.Recent farming practices have embraced remote sensing to provide fast and reliable information on commodity dynamics.Medium-to-low resolution free orbital images,such as those from Landsat 8 and Sentinel 2,have been used for crop mapping;however,satellite image processing requires high computing power,especially when monitoring vast areas.Therefore,cloud data processing has been the only feasible option to deal with a large amount of orbital data and its processing and analysis.Thus,our goal was to develop a method to map the two main crops(soybeans and corn)in Paraná,one of the major Brazilian state producers.Landsat-8,Sentinel-2,SRTM+,and field data from 2016 to 2018 were used with the Simple Non-Iterative Clustering segmentation method and the Continuous Naive Bayes classifier,to identify cropped areas.A minimum global accuracy of 90%was found for both crops.Comparison with field data showed correlations of 0.96 and agreement coefficients no lower than 0.86.This ensures mapping quality when using Sentinel and/or Landsat imagery on the GEE platform.Alex Paludo Willyan Ronaldo Becker Jonathan Richetti Laíza Cavalcante De Albuquerque Silva Jerry Adriani Johann 2020International Journal of Digital Earth2020,13,12:2
6Fatigue in children and adolescents with inflammatory bowel disease显示文摘AIM To identify factors other than active disease and anemia that contribute to fatigue in pediatric inflammatory bowel disease(IBD).METHODS We performed an electronic search in Medline and EMBASE from their inception to May 2017 using the search term 'fatigue' or the related keywords 'physical impairment' and 'inflammatory bowel disease' with the filter 'child'(age 0-18 years). Cross-sectional and case-control studies were included. We restricted our search to studies published in English. We used the PRISMA checklist and flow diagram. Duplicate articles were manually deleted in End Note. To identify further relevant studies, we checked the reference lists of the selected articles.RESULTS We identified 149 papers, of which 19 were retrieved for full text review. Eleven studies were subsequently excluded because fatigue was not evaluated as an outcome measure. Eight papers focused on the desired topic and were discussed in the final analysis. A lack of uniformity of outcome measures made the pooling of data impossible. In all but one study, questionnaires were used to evaluate fatigue. In the remaining study, an accelerometer was used to measure daily activities, sleeping time and their relationships with fatigue in a more quantifiable manner. Adolescents with IBD are significantly more fatigued than healthy controls. In addition to active disease, increased anxiety or depression and disturbed family relationships were frequently reported predictors of fatigue.
