7Adoptive transfer of Pfkfb3-disrupted hematopoietic cells to wild-type mice exacerbates diet-induced hepatic steatosis and inflammation显示文摘Background and objectives:Hepatic steatosis and inflammation are key characteristics of non-alcoholic fatty liver disease(NAFLD).However,whether and how hepatic steatosis and liver inflammation are differentially regulated remains to be elucidated.Considering that disruption of 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3(Pfkfb3/iPfk2)dissociates fat deposition and inflammation,the present study examined a role for Pfkfb3/iPfk2 in hematopoietic cells in regulating hepatic steatosis and inflammation in mice.Methods:Pfkfb3-disrupted(Pfkfb3^(+/-))mice and wild-type(WT)littermates were fed a high-fat diet(HFD)and examined for NAFLD phenotype.Also,bone marrow cells isolated from Pfkfb3^(+/-)mice andWT mice were differentiated into macrophages for analysis of macrophage activation status and for bone marrow transplantation(BMT)to generate chimeric(WT/BMT-Pfkfb3^(+/-))mice in which Pfkfb3 was disrupted only in hematopoietic cells and control chimeric(WT/BMT-WT)mice.The latter were also fed an HFD and examined for NAFLD phenotype.In vitro,hepatocytes were co-cultured with bone marrowderived macrophages and examined for hepatocyte fat deposition and proinflammatory responses.Results:After the feeding period,HFD-fed Pfkfb3^(+/-)mice displayed increased severity of liver inflammation in the absence of hepatic steatosis compared with HFD-fed WT mice.When inflammatory activation was analyzed,Pfkfb3^(+/-)macrophages revealed increased proinflammatory activation and decreased anti-proinflammatory activation.When NAFLD phenotype was analyzed in the chimeric mice,WT/BMT-Pfkfb3^(+/-) mice displayed increases in the severity of HFD-induced hepatic steatosis and inflammation compared with WT/BMT-WT mice.At the cellular level,hepatocytes co-cultured with Pfkfb3^(+/-) macrophages revealed increased fat deposition and proinflammatory responses compared with hepatocytes co-cultured with WT macrophages.Conclusions:Pfkfb3 disruption only in hematopoietic cells exacerbates HFD-induced hepatic steatosis and inflammation whereas the Pfkfb3/iPfk2 in nonhematopoietic cells appeared to be needed for HFD feeding to induce hepatic steatosis.As such,the Pfkfb3/iPfk2 plays a unique role in regulating NAFLD pathophysiology.Xin Guo Bilian Zhu Hang Xu Honggui Li Boxiong Jiang Yina Wang Benrong Zheng Shannon Glaser Gianfranco Alpini Chaodong Wu 2020Liver Research2020,4,3:1

13Advertisement vs.Monetary Subsidy:Which is Better for Remanufacturing?显示文摘The government subsidy plays an important role in the recycling and remanufacturing process of a closed-loop supply chain.This paper focuses on the impacts of different patterns of government subsidy on the chain members'profits and the environment.Furthermore,we develop three models in a supply chain including a manufacturer,a retailer and a collector,namely(1)the government invests public service advertisement to stimulate the remanufacturing process(Model AS);(2)the government offers only monetary subsidy to the collector(Model MS);(3)the government allocates the subsidy into two parts,namely both advertisement and monetary subsidy(Model HS).We find that,no matter which subsidy pattern the government adopts,both the manufacturer and the collector are beneficiaries of the government subsidy,and that such subsidizing activity is definitely conducive to the environmental sustainability.Taking into account the chain members,profits and the environmental impact,the optimal hybrid subsidy scheme(Model HS)is more beneficial than that in both Model MS and Model AS,which can be attributed to the combination of the incentives provided by the public service advertisement and the price attraction by the monetary subsidy.We also find that,for single advertisement and monetary subsidy,the amount of government subsidy will decide their merits and demerits.The direct monetary subsidy in Model MS will have a greater influence regarding to the supply quantity of used products than the mere advertisement subsidy in Model AS when the subsidy is relatively large.Yanting Huang Benrong Zheng Zongjun Wang 2020Journal of Systems Science and Systems Engineering2020,29,3:0
14Managing Sales Channel Selection for a Manufacturer in the Presence of Remanufacturing显示文摘Motivated by the fact that the product remanufacturing operations are increasingly performed as firms,competitive advantage and may also play an important role in the choice of channel structure,we construct game-theoretical models to examine the manufacturer's optimal sales channel strategy in a closed-loop supply chain(CLSC),in which the manufacturer is responsible for used product recycling and remanufacturing and the retailer operates a traditional retail channel.We show that the manufacturer's optimal sales channel selection depends on the customers7 acceptance of the direct channel and the remanufacturing efficiency.Specifically,in the centralized system,the manufacturer would prefer the dual-channel strategy rather than either the exclusive direct or retail channel,and becomes more willing to introduce a direct channel as the remanufacturing cost savings increase.However,in a decentralized system,there exists a Pareto improvement zone where both the manufacturer and the retailer are better off in the dual-channel format,and the increasing remanufacturing efficiency spills over to the retailer via a lower wholesale price and thus a higher retail demand.Moreover,we extend the study to the retailer-collecting mode and demonstrate that the main results of the original model remain robust.Benrong Zheng Niu Yu Jie Chu 2021Journal of Systems Science and Systems Engineering2021,30,5:0
