| 1 | 转录辅激活因子和乙酰基转移酶p300在心脏成纤维细胞衰老相关纤维化中的作用显示文摘目的:探讨转录辅激活因子和乙酰基转移酶p300在心脏成纤维细胞(CFs)衰老相关纤维化中的作用。方法:不同月龄的C57BL/6小鼠处死后取左心房组织或原代分离的CFs,Western blot检测p300蛋白,纤维化相关因子I型胶原蛋白α1链(Col1A1)、基质金属蛋白酶2(MMP-2)和转化生长因子β(TGF-β),以及衰老相关蛋白p53和p21的表达。将Cre酶转染来自p300+/+和p300 flox/+小鼠的原代培养CFs,筛选建立稳定敲除p300的CFs,PCR检测细胞基因型,Western blot检测沉默效率;衰老相关β-半乳糖苷酶(SA-β-Gal)染色观察敲除p300对CFs衰老率的影响,细胞计数观察细胞增殖能力。结果:随着月龄增加,小鼠左心房组织p300、Col1A1、MMP-2、TGF-β、p53和p21表达增加(P<0.05)。随着小鼠CFs传代代数增加,SA-β-G染色显示细胞衰老率增加,同时p300、Col1A1、MMP-2、TGF-β、p53和p21蛋白表达明显增加(P<0.05)。与野生型组相比,敲除p300的CFs随着代数的增加,未见衰老细胞,且细胞增殖能力明显强于对照组(P<0.05)。结论:心房组织中,p300可能参与CFs衰老相关纤维化。 | 高小燕 李昕 韦鉴钦 Nanette Hahr BISHOPRIC 王钊煜 周慧珊 饶芳 邓春玉 | 2019 | 中国病理生理杂志2019，35，12： | 2 |
