1Influence of mild hypothermia on vascular endothelial growth factor and infarct volume in brain tissues after cerebral ischemia in rats显示文摘BACKGROUND: It has been demonstrated that mild hypothermia has obvious protective effect on both whole and local cerebral ischemia. However, the definite mechanism is still unclear for the brain protection of mild hypothermia on cerebral edema, inhibiting inflammatory reaction, stabilizing blood brain barrier, etc. OBJECTIVE: To investigate the effect of mild hypothermia on the expression of vascular endothelial growth factor and the infarct volume after cerebral ischemia in rats, and analyze the brain protective mechanism of mild hypothermia. DESIGN: A randomized grouping and controlled animal trial. SETTING: Department of Neurology, People's Hospital of Yunyang Medical College. MATERIALS: Twenty adult male SD rats of clean degree, weighing (250±30) g, were provided by the animal experimental center, School of Medicine, Wuhan University. The kits for SP immunohistochemistry were purchased from Beijing Zhongshan Golden Bridge Biotechnology Co., Ltd. METHODS: The experiments were carried out in the laboratory of Department of Neurology, Renmen Hospital of Wuhan University from May to July 2005. ① The 20 rats were divided randomly into normal temperature group (n =10) and mild hypothermia group (n =10). Models of permanent middle cerebral artery occlusion were established with modified nylon suture embolization. The rats were assessed with the Longa standards: 0 point for without nerve dysfunction; 1 for mild neurological deficit (fore claws could no extend completely); 2 for moderate neurological deficit (circling towards the affected side); 3 for severe neurological deficit (tilting towards the affected side); 4 for coma and unconscious; 1-3 points represented that models were successfully established. The rats of the normal temperature group were fed at room temperature, and those in the mild hypothermia group were induced by hypothermia from 2 hours postoperatively, and the rectal temperature was kept at 34-35 ℃ for 72 hours. ② Measurement of infarct volume: All the rats were anesthetized by intraperitoneal injection overdose sodium pentobarbital 7 days postoperatively, and then the heads were cut down to harvest brain. The brain tissues were placed into -20 ℃ refrigerator for 20 minutes, coronal sections of 2 mm were prepared. The infarct sites were not stained, whereas normal brain tissues were stained as red. The infarct volumes were calculated by using MPLAS-500 multimedia color pathological image&word analytical system. ③ Counting positive cells of vascular endothelial growth factor protein: The brains were harvested by cutting heads, then coronal sections of 2 mm were prepared. Routine dehydration, hyalinization, wax immersion and embedding were performed, then the detected with SP immunohistochemistry, the kits were purchased from Beijing Zhongshan Golden Bridge Biotechnology Co., Ltd. The cells whose cytoplasm was yellow-brown were positive ones, a single sample as a unit, peri-ischemic site and ischemic core were selected, and the corresponding sites in controlateral hemisphere were taken as controls. Five visual fields were selected from each site to be observed under microscope, the cells were counted, and the average number of positive cells was calculated in each group. The numbers of positive cells were determined with the image analytical apparatus. MAIN OUTCOME MEASURES: Number of the positive cells of vascular endothelial growth factor protein; Infarct volume of rat brain tissue. RESULTS: All the 20 rats were involved in the analysis of results. ① Number of positive cells of vascular endothelial growth factor protein in brain tissue: It was obviously lower in the mild hypothermia group than in the normal temperature group [(24.02±5.05), (36.07±2.69) cells/high power visual field, P < 0.01]. ② Comparison of infarct volume of brain tissue: After MCAO, it was obviously smaller in the mild hypothermia group than in the normal temperature group [(153.25±23.14), (253.45±36.21) mm3, P < 0.01]. CONCLUSION: Mild hypothermia can inhibit the expression of vascular endothelial growth factor and decrease the volume of cerebral infarction. The inhibition of mild hypothermia on the expression of vascular endothelial growth factor may be one of the brain protective mechanisms.Fei Ye1, Gangming Xi1, Biyong Qin1, Shifeng Wang1, Chengyan Li2 1Department of Neurology, People’s Hospital of Yunyang Medical College, Shiyan 442000, Hubei Province, China 2Department of Neurology, Renmin Hospital of Wuhan University, Wuhan 430060, Hubei Province, China 2006Neural Regeneration Research2006,1,9:2
2Validation of different personalized risk models of chemotherapy-induced nausea and vomiting:results of a randomized,double-blind,phase III trial of fosaprepitant for cancer patients treated with high-dose cisplatin显示文摘Background:Highly emetogenic chemotherapy induces emesis in cancer patients without prophylaxis. The purpose of this study was to evaluate the efficacy and safety of a fosaprepitant-based triple antiemetic regimen for the prevention of chemotherapy-induced nausea and vomiting(CINV)in patients with solid malignant tumors, determine risk factors and externally validate different personalized risk models for CINV.Methods:This phase III trial was designed to test the non-inferiority of fosaprepitant toward aprepitant in cancer patients who were to receive the first cycle of single-day cisplatin chemotherapy. The primary endpoint was complete response(CR)during the overall phase(OP)with a non-inferiority margin of 10.0%.Logistic regression modelswere used to assess the risk factors ofCRand no nausea. To validate the personalized risk models, the accuracy of the risk scoring systems was determined by measuring the specificity, sensitivity and area under the receiver operating characteristic(ROC)curve(AUC),while the predictive accuracy of the nomogram was measured using concordance index(C-index).Results:A total of 720 patients were randomly assigned.CR during the OP in the fosaprepitant group was not inferior to that in the aprepitant group(78.1%vs.77.7%,P=0.765)with a between-group difference of 0.4%(95%CI,-5.7%to 6.6%).Female sex, higher cisplatin dose(≥70 mg/m2),no history of drinking and larger body surface area(BSA)were significantly associated with nausea. The AUC for the acute and delayed CINV risk indexes was 0.68(95%CI:0.66-0.71)and 0.66(95%CI:0.61-0.70),respectively,and the C-index for nomogram CINV prediction was 0.59(95%CI,0.54-0.64).Using appropriate cutoff points, the three models could stratify patients with high-or low-risk CINV.No nausea and CR rate were significantly higher in the low-risk group than in the high-risk group(P<0.001).Conclusions:Fosaprepitant-based triple prophylaxis demonstrated non-inferior control for preventing CINV in patients treated with cisplatin-base chemotherapy.Female cancer patients without a history of alcohol consumption, with larger BSA and received high-dose cisplatin might be more vulnerable to CINV.Three personalized prediction models were well-validated and could be used to optimize antiemetic therapy for individual patients.Yuanyuan Zhao Bing Zhao Gang Chen Yinlan Chen Zijun Liao Haiming Zhang Weineng Feng Yinyin Li Heng Weng Weidong Li Yuefen Zhou Biyong Ren Yanda Lu Jianhua Chen Zhenteng Liu Zhenzhong Su Wenliang Wang Li Zhang 2023Cancer Communications2023,43,2:2
3MR扩散加权成像结合动态增强扫描诊断胰腺内分泌肿瘤的价值(英文)显示文摘Objective: The aim of our study was to analysis the pictures of conventional magnetic resonance imaging(MR), diffusion-weighted imaging(DWI) and dynamic enhanced magnetic resonance imaging(DEMRI) of pancreatic endocrine tumors(PETs), and evaluate diagnostic value of MR, DWI and DEMRI for diagnosing PETs. Methods: DWI and DEMRI scanning toward 13 patients with PETs being confirmed by surgical pathology before surgery on the basis of conventional MR scanning were carried out, and MR findings was analyzed retrospectively. Results: Of 13 patients with PETs there was 11 cases with single lesion, 2 with multiple, and had 15 lesions altogether, of which there were 3 lesions in pancreatic head, 1 in its neck, 2 in its body, 4 in its body and tail, 5 in its tail. MR findings:(1) T1 WI signal was low or slightly lower(9/15), and equal ones(5/15);(2) T2 WI showed high or slightly higher signal(10/15), and equal ones(5/15);(3) T1 WI with fat suppression: the signal was low(11/15), mixed signal(2/15), and equal ones(2/15);(4) DWI: normal pancreatic tissue exhibited homogeneous intermediate signal, all 15 lesions were high or slightly higher signal, the measured ADC values of tissue of PETs was(1.124 ± 0.252) × 10–3 mm2/s, and the ADC value of normal pancreatic tissue(1.873 ± 0.157) × 10–3 mm2/s;(5) Enhanced(M3D/LAVA) scanning: among 13 patients with PETs there were 12 pancreatic lesions with significantly enhanced signals in the arterial phase in all 15, and significantly higher than normal pancreatic tissue, and two slight enhancement was slightly higher signal; and 1 no enhancement. Enhanced pattern: homogeneous enhancement were 6 lesions, and the heterogeneous 4, and the edge ring 5. Conclusion: MR and DWI combining with DEMRI help qualitative diagnosis of pancreatic endocrine tumors.Xingrong Hu Xiannian Cui Jun Chen Biyong Tan 2014The Chinese-German Journal of Clinical Oncology2014,13,1:1

7Effects of nitrite on phosphate uptake in anaerobic-oxic process显示文摘An anaerobic-oxic(A/O)biological phosphorus removal reactor was operated to study the effect of nitrite on phosphate uptake. The phosphorus uptake profile was determined under different operating conditions. The results indicated that in addition to oxygen and nitrate(DPB_(Na),nitrate denitrifying phosphorus removal),to some extent, nitrite could also serve as an electron acceptor to achieve nitrite denitrifying phosphorus removal(DPB_(Ni)).The quantity and rate of phosphorus uptake of DPBNi,however, were evidently lower than that of DPBNa.The experiment results revealed that nitrite would bring toxic action to phosphate-accumulating organisms(PAOs)when NO_(2)^(−)-Ni93.7 mg/L.The nitrite existing in the anoxic reactor made no difference to the quantity and rate of denitrifying phosphorus removal, but it could reduce the consumption of nitrate. Moreover, the data showed that the aerobic phosphate uptake of DPBNi was lower than that of anaerobic phosphorus-released sludge in a traditional A/O process. However, there was not much difference between these two kinds of sludge in terms of the total phosphorus uptake quantity and the effluent quality.LI Jie XIONG Biyong ZHANG Shude YANG Hong ZHANG Jie 2007Frontiers of Environmental Science & Engineering2007,1,1:1
