| 2 | Resveratrol improves cognition and reduces oxidative stress in rats with vascular dementia显示文摘Resveratrol possesses beneficial biological effects,which include anti-oxidant,anti-inflammatory and anti-carcinogenic properties.Recently,resveratrol has been shown to exhibit neuroprotective effects in models of Parkinson's disease,cerebral ischemia and Alzheimer's disease.However,its effects on vascular dementia remain unclear.The present study established a rat model of vascular dementia using permanent bilateral common carotid artery occlusion.At 8-12 weeks after model induction,rats were intragastrically administered 25 mg/kg resveratrol daily.Our results found that resveratrol shortened the escape latency and escape distances in the Morris water maze,and prolonged the time spent percentage and swimming distance percentage in the target quadrant during the probe test,indicating that resveratrol improved learning and memory ability in vascular dementia rats.Further experiments found that resveratrol decreased malonyldialdehyde levels,and increased superoxide dismutase activity and glutathione levels in the hippocampus and cerebral cortex of vascular dementia rats.These results confirmed that the neuroprotective effects of resveratrol on vascular dementia were associated with its anti-oxidant properties. | Xingrong Ma Zhikun Sun Yanru Liu Yanjie Jia Boai Zhang Jiewen Zhang | 2013 | Neural Regeneration Research2013,8,22: | 7 |
| 3 | Retrovirus-mediated delivery of an IL-4 receptor antagonist inhibits allergic responses in a murine model of asthma显示文摘This work reports the investigation of the effect of airway IL-4RA gene transfer by a recombinant retroviral vector on airway inflammation and airway responsiveness in asthmatic mice. The retrovirus-mediated delivery of IL-4RA to the airways of mice inhibited elevations of airway responsiveness and the development of allergic inflammation in asthmatic mice, and regulated the Th1/Th2 balance in OVA-sensitized and -challenged mouse models. | 4 | Fasudil hydrochloride differentiates bone marrow mesenchymal stem cells into neurons via notch signaling显示文摘BACKGROUND: Notch signaling regulates bone marrow mesenchymal stem cell (MSC) proliferation, differentiation, and apoptosis, Notch signaling and Rho kinase signaling exhibit a crosstalk phenomenon with JAK/STAT, and both participate in the neuronal dendritic spine development. Inhibition of RhoA/Rho kinase signaling may regulate MSC differentiation into neuronal-like cells. OBJECTIVE: To investigate the effect of Notch1 signaling on the differentiation of rat MSCs into neurons induced by fasudil hydrochloride (C14H17N3O2S·HCl), a Rho kinase inhibitor, through a siRNA approach. DESIGN, TIME AND SETTING: An in vitro cytological experiment was performed in the Cell Laboratory of Henan Academy of Medical and Pharmaceutical Sciences between December 2007 and May 2009. MATERIALS: MSCs were obtained from Wistar rat femoral bone, fasudil hydrochloride was provided by Tianjin Chase Sun Pharmaceutical Co., Ltd. Rn-notch1-siRNa, negative control siRNA (Cy3 label) and Rn-MAPK1 control siRNA were provided by QIAGEN, Coloqne, German. METHODS: The cultured MSCs were divided into non-transfected, transfected group (transfected with Rn-Notch1-siRNA), positive control (transfected with Rn-MAPK-1 control siRNA), and negative control (transfected with negative control siRNA) groups. Fasudil hydrochloride was applied to induce MSCs to differentiate into neurons. MAIN OUTCOME MEASURES: The fluorescence expression by the transfected MSCs was observed under an inverted fluorescence microscope; the expression of Notch1 mRNA, Hes1 mRNA, and MAPK1 mRNA in MSCs was detected by reverse transcription polymerase chain reaction; the expression of Notch1 protein, nestin, neurofilament M, and glial fibrillary acidic protein was detected by immunocytochemistry. The viability of MSCs was detected by tetrazolium bromide assay. RESULTS: MSC fluorescence increased following a 72-hour siRNA transfection, with transfection efficiencies of up to (0.91 ± 0.04); the Notch1 mRNA and Hes1 mRNA expressed by transfected MSCs was significantly decreased (P < 0.05) compared with non-transfected cells. Fasudil hydrochloride induced MSCs to differentiate into neurons with greater efficiency in the transfected group (P < 0.05). BrdU-positive cells at day 7 post-transplantation, as well as nestin- and neuron specific enolase-positive cells at day 14 were increased compared with those of the single neural stem cell transplantation group. In addition, the proportion of neuronal differentiation was enhanced. The genetically modified cell-transplanted rats exhibited enhanced performance in correctly crossing a Y-maze and climbing an angled slope compared with those of the single neural stem cell transplantation group. | 14 | Effects of complete Freund's adjuvant on immunohistochemical distribution of IL-1β and IL-1R I in neurons and glia cells of dorsal root ganglion显示文摘Aim: To investigate the effects of complete Freund's adjuvant (CFA) on inflammatory hyperalgesia and morphological change of the coexsistence of interleukin-1 beta (IL-1β) and type I IL-1 receptor (IL-1RI) in neurons and glia cells of rat dorsal root ganglion (DRG). Methods: The pain-related parameters and the expression of IL-1RI and IL-1β positive neurons and glia cells of DRG in normal saline (NS) and adjuvant-induced arthritic (AA) group were examined with pain behavior assessment methods and immunohistochemical assay, respectively. Results: Five hours, 1 d, and 2 d after intra-articular injection of 50 uL CFA, tactile hyperalgesia induced by CFA was observed in the foot flexion and extension scores of the ipsilateral hindpaw of AA group. Three days after injection, the distribution of IL-1RI/IL-1β double-stained coexisted neurons and glia cells were observed in ipsilateral DRG of both groups. The number of IL-1β positive neurons, IL-1RI positive neurons, IL-1β/IL-1RI double-stained neurons, and IL-1RI positive glia cells in ipsilateral DRG of the AA group were higher than that of NS group (P<0.05 or P<0.01). Conclusion: The coexsistence of IL- 1β and IL- 1RI in neurons and nonneuronal cells suggests an as yet unknown autocrine and/or paracrine function of IL-1β in the DRG.
