| 1 | Epigenetic regulation: methylation of histone and non-histone proteins显示文摘Histone methylation is believed to play important roles in epigenetic memory in various biological processes. However, questions like whether the methylation marks themselves are faithfully transmit- ted into daughter cells and through what mechanisms are currently under active investigation. Previ- ously, methylation was considered to be irreversible, but the recent discovery of histone lysine de- methylases revealed a dynamic nature of histone methylation regulation on four of the main sites of methylation on histone H3 and H4 tails (H3K4, H3K9, H3K27 and H3K36). Even so, it is still unclear whether demethylases specific for the remaining two sites, H3K79 and H4K20, exist. Furthermore, be- sides histone proteins, the lysine methylation and demethylation also occur on non-histone proteins, which are probably subjected to similar regulation as histones. | 3 | Intrinsic control of axon regeneration显示文摘Spinal cord injury disrupts the connections between the brain and spinal cord, often resulting in the loss of sensory and motor function below the lesion site. The most important reason for such permanent functional deficits is the failure of injured axons to regenerate after injury. In principle, the functional recovery could be achieved by two forms of axonal regrowth: the regeneration of lesioned axons which will reconnect with their original targets and the sprouting of spared axons that form new circuits and compensate for the lost function. Our recent studies reveal the activity of the mammalian target of rapamycin (mTOR) pathway, a major regulator of new protein synthesis, as a critical determinant of axon regrowth in the adult retinal ganglion neurons[1]. | 4 | Inducible resistance to Fas-mediated apoptosis in B cells显示文摘Apoptosis produced in B cells through Fas (APO-1, CD95) triggering is regulated by signals derived from other surface receptors: CD40 engagement produces upregulation of Fas expres. sion and marked susceptibility to Fas-induced cell death, whereas antigen receptor engagement, or IL-4R engagement, inhibits Fas killing and in so doing induces a state of Fas-resistance, even in otherwise sensitive, CD40-stimulated targets. Surfaceim. munoglobulin and IL-4R utilize at least partially distinct pathways to produce Fas-resistance that differentially depend on PKC and STAT6, respectively. Further, surface immunoglob- 'lin signaling for inducible Fas-resistance bypasses Bib, requires NF-GB, and entails new macromolecular synthesis. Terminal effectors of B cell Fas-resistance include the known anti-apoptotic gene products, Bcl-xL and FLIP, and a novel anti-apoptotic gene that encodes FAIM (Fas Apoptosis Inhibitory Molecule). faim was identified by differential display and exists in two alternatively spliced forms; maim-S is broadly expressed, but faim-L expression is tissue-specific. The FAIM sequence is highly evolu tionarily conserved, suggesting an important role for this molecule throughout phylogeny. Inducible resistance to Fas killing is hypothesized to protect foreign antigen-specific B cells during potentially hazardous interactions with FasL-bearing T cells, whereas autoreactive B cells fail to become Fas-resistant and are deleted via Fas-dependent cytotoxicity. | 8 | FINE PARTICULATE AIR POLLUTION: EPIDEMIOLOGIC BASIS FOR AIR QUALITY STANDARDS显示文摘In proposing a new ambient air quality standard for fine particulate air pollution in 1997, the U.S. Environmental Protection Agency was responding to a wealth of epidemiologic evidence. At the same time, it was recognized that there was little supporting evidence from toxicologic or controlled human exposure studies. The epidemiologic findings were questioned as only statistical associations without a plausible biological mechanism or an identified causal agent in the complex mix of pollutants characterized as fine particles. In response to these weaknesses in the scientific evidence, a series of research initiatives have been undertaken in exposure assessment, controlled animal and human exposures, and assessments of free-living populations. Key papers have been subjected to comprehensive review and replication. It is now four to seven years since the seminal papers suggesting adverse health effects of fine particles were published. Have these results held up over time? Have they been confirmed by other investigators in other populations? Do we have insights into the biologic mechanisms? | 10 | Arginine: New and Exciting Developments for an“Old”Amino Acid显示文摘Arginine (2-amino-5-guanidino pentanoic acid) was shown in 1895 by Hedin to be present in the proteins of horn. Metabolic nitrogen balance studies, conducted in 1957 by Rose in human adults and in 1959 by Snyderman and coworkers in young infants, revealed that a dietary source of this amino acid was not an obligatory requirement for growth and maintenance of nitrogen homeostasis in healthy individuals. Hence, It was initially classified as a non-essential (dispensable) amino acid and, perhaps, for reasons of this classification arginine did not receivc the earlier atteniion it now deserves, in relation to an understanding of the nutritional biochemistry and physiology of its metabolism in humans subjects. However, there is currently a considerable interest in the cellular and tissue functions, as well as clinical, therapeutic significance, of arginine. In this paper we review the multiple functions of arginlne, including its role in the L-arginine-nitric oxide pathway,cellular regeneration, immune function, protein synthesis and protein breakdown. We then consider some in vivo aspects of the physiology of arginine metabolism, which varies greatly among eukaryotes, with particular reference to humans. Against this background, studies of arginine in the nutrition of humans under various pathophysiological conditions are reviewed briefly. | 16 | Simulation studies of high-latitude magnetospheric boundary dynamics显示文摘Magnetic reconnection at the high-latitude magnetopause is studied using 2.5-dimensional Hall-MHD simulation. Concentric flow vortices and magnetic islands appear when both Hall effect and sheared flow are considered. Plasma mixing across the magnetopause occurs in the presence of the flow vortices. Reconnected structure generated in the vicinity of the subsolar point changes its geometry with increasing flow shear while moving to high latitudes. In the presence of flow shear, with the Hall-MHD reconnection a higher reconnection rate than with the traditional MHD is obtained. The out-of-plane components of flow and magnetic field produced by the Hall current are redistributed under the action of the flow shear, which makes the plasma transport across the boundaries more complicated.
