| 2 | Characterization of a disease susceptibility locus for exploring an efficient way to improve rice resistance against bacterial blight显示文摘Bacterial blight caused by Xanthomonas oryzae pv.oryzae(Xoo) is the most harmful bacterial disease of rice worldwide.Previously,we characterized major disease resistance(MR) gene xa25,which confers race-specific resistance to Xoo strain PXO339.The xa25 is a recessive allele of the SWEET13 locus,but SWEET13's interaction with PXO339 and how efficiently using this locus for rice breeding still need to be defined.Here we show that the SWEET 13 allele from rice Zhenshan 97 is a susceptibility gene to PXO339.Using this allele's promoter to regulate xa25 resulted in disease,suggesting that the promoter is a key determinant in SWEET13 caused disease in Zhanshan 97 after PXO339 infection.PXO339 transcriptionally induces SWEET13 to cause disease.Partial suppressing SWEET13 expression leads to a high level of resistance to PXO339.Thus,the transcriptionally suppressed SWEET13 functions as xa25 in resistance to PXO339.Hybrid rice is widely grown in many countries.However,recessive MR genes have not been efficiently used for disease resistance breeding in hybrid rice production for both parents of the hybrid have to carry the same recessive gene.However,the suppressed SWEET 13 functions dominantly,which will have advantage to improve the resistance of hybrid rice to xa25-incomptible Xoo. | Qi Cheng Weihua Mao Wenya Xie Qinsong Liu Jianbo Cao Meng Yuan Qinglu Zhang Xianghua Li Shiping Wang | 2017 | Science China(Life Sciences)2017,60,3: | 4 |
| 3 | miR395-regulated sulfate metabolism exploits pathogen sensitivity to sulfate to boost immunity in rice显示文摘MicroRNAs (miRNAs) play important roles in plant physiological activities. However, their roles and molecular mechanisms in boosting plant immunity, especially through the modulation of macronutrient metabolism in response to pathogens, are largely unknown. Here, we report that an evolutionarily conserved miRNA, miR395, promotes resistance to Xanthomonas oryzae pv. oryzae (Xoo) and X. oryzae pv. oryzicola (Xoc), two destructive bacterial pathogens, by regulating sulfate accumulation and distribution in rice. Specifically, miR395 targets and suppresses the expression of the ATP sulfurylase gene OsAPS1, which functions in sulfate assimilation, and two sulfate transporter genes, OsSULTR2;1 and OsSULTR2;2, which function in sulfate translocation, to promote sulfate accumulation, resulting in broad-spectrum resistance to bacterial pathogens in miR395-overexpressing plants. Genetic analysis revealed that miR395-triggered resistance is involved in both pathogen-associated molecular pattern-triggered immunity and R gene-mediated resistance. Moreover, we found that accumulated sulfate but not S-metabolites inhibits proliferation of pathogenic bacteria, revealing a sulfate-mediated antibacterial defense mechanism that differs from sulfur-induced resistance. Furthermore, compared with other bacteria, Xoo and Xoc, which lack the sulfate transporter CysZ, are sensitive to high levels of extracellular sulfate. Accordingly, miR395-regulated sulfate accumulation impaired the virulence of Xoo and Xoc by decreasing extracellular polysaccharide production and biofilm formation. | 7 | Investigation of ion-atom collision dynamics through imaging techniques显示文摘The principle and technique details of recoil ion momentum imaging are discussed and summarized. The recoil ion momentum spectroscopy built at the Institute of Modern Physics (Lanzhou) is presented. The first results obtained at the setup are analyzed. For 30 keV He2+ on He collision, it is found that the capture of single electron occurs dominantly into the first excited states, and the related scattering angle results show that the ground state capture occurs at large impact parameters, while the capture into excited states occurs at small impact parameters. The results manifest the collision dynamics for the sub-femto-second process can be studied through the techniques uniquely. | 8 | Extraordinary characteristics for one-dimensional parity-time-symmetric periodic ring optical waveguide networks显示文摘In this paper, we design a one-dimensional(1D) parity-time-symmetric periodic ring optical waveguide network(PTSPROWN) and investigate its extraordinary optical characteristics. It is found that quite different from traditional vacuum/dielectric optical waveguide networks, 1D PTSPROWN cannot produce a photonic ordinary propagation mode, but can generate simultaneously two kinds of photonic nonpropagation modes: attenuation propagation mode and gain propagation mode. It creates neither passband nor stopband and possesses no photonic band structure. | 17 | The Effect of the Cyp19a1 Gene Methylation Modification on Temperature-dependent Sex Determination of Reeves' Turtle(Mauremys reevesii)显示文摘Temperature-dependent sex determination(TSD) is a type of environmental sex determination in which the sex of the embryos depends on the ambient temperature; however,the molecular mechanisms governing this process remain unknown.Aromatase,encoded by the cyp19a1 gene,which converts androgens into estrogens in animals,was considered to be the key gene for TSD.In this study,the 5'-flanking region of the cyp19a1 gene in Reeves' turtle(Mauremys reevesii) was cloned,and the promoter region was identified using the luciferase reporter assay.Then the eggs of Reeves' turtle were incubated at different temperatures(26°C: male-biased temperature; 29°C: non-sex-biased temperature and 32°C: female-biased temperature).During the thermosensitive period,the adrenal kidney gonad complexes(AKG) were sampled.DNA methylation analysis of the AKG samples showed that the promoter region of the cyp19a1 gene was significantly de-methylated in the female-biased temperature regime(P<0.01).Quantitative analysis of the cyp19a1 gene and estrogen by q PCR and Elisa assay showed that the expression level of the cyp19a1 gene and estrogen content were both upregulated significantly at the female-biased temperature(P<0.01).These results indicated that the de-methylation response of the cyp19a1 gene to incubation temperature,especially at the female-biased temperature,could lead to temperature-specific expression of aromatase and increased estrogen levels,which may further determine gonadal development in Reeves' turtle.These findings provide insights into the genetic mechanisms underlying TSD. | Wenxiu RU Liushuai HUA Yufeng WEI Weiye LI Dainan CAO Yan GE Hong CHEN Xianyong LAN Shiping GONG | 2017 | Asian Herpetological Research2017,18,3: | 1 |

| 19 | Electrochemical performance of NiCl_(2) with Br-free molten salt electrolyte in high power thermal batteries显示文摘NiCl_(2) with high theoretical voltage and thermal decomposition temperature attracts much attention as cathode material for thermal batteries with the requirement of high power density, high energy density and long work time. Unfortunately, the practical utilization of thermal batteries with NiCl_(2) cathode is limited by their poor electrochemical performance under large current, even with the conventional Li F-Li Cl-Li Br all-lithium molten salt electrolyte which proposes ultrahigh lithium ion conductivity. In this work, an unexpected ionic exchange reaction between NiCl_(2) and Li Br in Li F-Li Cl-Li Br was found, which would be the main reason for the poor electrochemical behavior of thermal batteries with NiCl_(2) cathode and Li F-Li Cl-Li Br molten salt. On this basis, Li F-Li Cl-Li_(2)SO_(4), another all-lithium molten salt free of Li Br, was investigated as the new electrolyte for NiCl_(2) cathode. | 20 | Effects of Acute Temperature Stress on mRNA Expression of Transferrin in the Yellow Pond Turtle Mauremys mutica显示文摘The yellow pond turtle Mauremys mutica is widely cultured using both greenhouse-reared and outdoor pond-reared models.Individuals from the two models often show different tolerances to dramatic temperature changes caused by extreme weather events.However,the mechanism underlying the difference is unclear.In this study,we found that for greenhouse-reared turtles(GRTs),the expression levels of an immune-related gene for transferrin were significantly different(P<0.05)between the control group and the acute cold stress(ACS)group for most time points(3 h,6 h and 48 h),while at two time points(6 h and 12 h)there was a significant difference(P<0.05)between the control group and the acute heat stress(AHS)group.However,for the outdoor pond-reared turtles(OPTs),we found the opposite pattern:the ACS group showed no significant difference(P>0.05)from the control group for all time points(3 h,6 h,12 h,24 h and 48 h),whereas two time points(12 h and 24 h)were significantly different(P<0.05)for the AHS group.Our results indicate that ACS may influence the immunity of GRTs and have no influence on OPTs,whereas AHS may largely affect the immunity of OPTs and have little influence on GRTs.The findings provide insights into the mechanism underlying the different morbidity and mortality rates of turtles from different culture models after extreme weather events. | Yufeng WEI Yangchun GAO Dainan CAO Yan GE Haitao SHI Shiping GONG | 2020 | Asian Herpetological Research2020,11,2: | 0 |
