| 1 | Alterations in metastatic properties of hepatocellular carcinoma cell following H-ras oncogene transfection显示文摘AIM To demonstrate the relationship betweenH-ras oncogene and hepatocellular carcinoma(HCC) metastasis.METHODS Activated H-ras oncogene wastransfected into SMMC 7721, a cell line derivedfrom human HCC, by calcium phosphatetransfection method. Some metastasis-relatedparameters were detected in vitro, includingadhesion assay, migration assay, expression ofcollagenase ⅣV (c ⅣV ase) and epidermal growthfactor receptor (EGFR).RESULTS The abilities of H-ras-transfected cellclones in adhesion to laminin (LN) or fibronectin(FN), migration, c Ⅳ ase secretion increasedmarkedly, and the expression of EGFR elevatedmoderately. | 4 | Some open problems in granular matter mechanics显示文摘Granular matter is a large assemblage of solid particles, which is fundamentally different from any other type of matters, such as solid and liquid. Most models presented for granular matter are phenomenological and are only suitable for solving engineering problems. Many fundamental mechanical problems remain open. By analyzing characteristics of internal state structure, we propose that granular matter is intrinsically multiscale, i.e. microscale of particle size, mesoscale of force chain, and macroscale of the bulk of granular matter. The correlations among difference scales would be crucial. The mesoscale force chain network is determined by both particle properties and macroscopic boundary conditions. The evolution of the force the chain network contributes to macroscopic mechanical properties of granular matter. In addition, we discuss the drawbacks in simplifying contact forces in the current models, and the difficulties in analyzing the interaction of interstitial fluid in wet granular matter. | 5 | The integration of methanogenesis with denitrification and anaerobic ammonium oxidation in an expanded granular sludge bed reactor显示文摘The integration of methanogenesis with denitrification and anaerobic ammonium oxidation(ANAMMOX) was studied in an expanded granular sludge bed(EGSB) reactor in this work. Experimental results from the continuous treatment of wastewater with nitrite and ammonium, which lasted for 107 days, demonstrated that wastewater with high nitrite and ammonium could be anaerobically treated in an expanded granular sludge bed reactor. More than 91% to 97% of COD were removed at up to about 3.9 g COD/(L·d) of COD volumetric loading rate. More than 97% to 100% of nitrite was denitrified at up to about 0.8 g NO -2-N/(L·d), which is 16 times higher than that in a conventional activated sludge system with nitrification/denitrification(0.05 gN/(L·d)). No dissimilatory reduction of nitrite to ammonium occurred in the process. However, maximum of about 40% ammonium was found to be lost. Batch tests of 15 days with sludge from the reactor showed that 100% of nitrite was denitrified completely, and about 3% of ammonium was removed when only ammonium (34.3 mg/L) and nitrite(34.3 mg/L) were added into the sludge suspension medium. Furthermore, about 15% of ammonium amounts were lost with organic COD addition. It suggested that the methanogenesis in the system could enhance ANAMMOX because of intermediate hydrogen produced during methanogenesis. | ZHANG Dai-jun 1,2 (1. | 6 | Dissimilatory Fe(Ⅲ) reduction characteristics of paddy soil extract cultures treated with glucose or fatty acids显示文摘Dissimilatory Fe(Ⅲ) reduction is a universal process with irreplaceable biological and environmental importance in anoxic environments. Our knowledge about Fe(Ⅲ) reduction predominantly comes from pure cultures of dissimilatory Fe(Ⅲ) reducing bacteria (DFRB). The objective of this study was to compare the effects of glucose and a selection of short organic acids (citrate, succinate, pyruvate, propionate, acetate, and formate) on Fe(Ⅲ) reduction via the anaerobic culture of three paddy soil solutions with Fe(OH)3 as the sole electron acceptor. The results showed significant differences in Fe(Ⅲ) reduction among the three paddy soil solutions and substrate types. Bacteria from the Sichuan paddy soil responded quickly to substrate supply and showed higher Fe(Ⅲ) reducing activity than the other two soil types. Bacteria in the Jiangxi paddy soil culture solution could not use propionate as a source of electrons for Fe(Ⅲ) reduction. Similarly, bacteria in the Jilin paddy soil culture could not use succinate effectively. Pyruvate was readily used by cultures from all three paddy soil solutions, thus indicating that there were some similarities in substrate utilization by bacteria for Fe(Ⅲ) reduction. Effect of S-ODNson SMMC-7721 human hepatoma cell growth invitro was determined using microculturetetrazolium assay. In vivo antitumor activitiesof S-ODNs were monitored by measuring tumorweight differences in treated and control micebearing SMMC-7721 xenografts. Induction of cellapoptosis was evaluated by fluorescence-activated cell sorter (FACS) analysis.RESULTS Antisense S-ODN treatment led toreduced AFP gene expression. Specificantisense S-ODNs, but not control S-ODNs,inhibited the growth of heaptoma cells in vitro.In vivo. only antisense S-ODNs exhibitedobvious antitumor activities. FACS analysisrevealed that the growth inhibition by antisenseS. | 11 | Metabonomic phenotype and identification of “heart blood stasis obstruction pattern” and “qi and yin deficiency pattern” of myocardial ischemia rat models显示文摘The traditional Chinese medicine concepts of 'Xinxueyuzuzheng (heart blood stasis obstruction pattern)' and 'Qiyinliangxuzheng (qi and yin deficiency pattern)' for myocardial ischemia rat models were constructed in the present study. Endogenous metabolites in rat plasma were analyzed using the GC/TOF-MS-based metabonomic method. Significant metabolic differences were observed between the control and two model groups, and the three groups were distinguished clearly by pattern recognition. Compared with those of the control, the levels of hydroxyproline, threonic acid, glutamine and citric acid were strikingly up or down-regulated in model rats. The metabolites contributing most to the classification between the two 'pattern' rats were identified, such as valine, serine, threonine, ornithine, hydroxyproline, lysine, 2-hydroxybutanoic acid, 3-hydroxybutanoic acid, galactofuranose and inositol. These compounds were indicated as the potential biomarkers. The results suggested that the two 'patterns' are involved in dysfunction in oxidative stress, energy metabolism and amino acid metabolism. | 12 | Virulence and potential pathogenicity of coccoid Helicobacter pylori induced by antibiotics显示文摘AIM To explore the virulence and the potential pathogenicity of coccoid Helicobacter pylori (H. pylori) transformed from spiral form by exposure to antibiotic.METHODS Three strains of H. pylori, isolated from gastric biopsy specimens of confirmed peptic ulcer, were converted from spiral into coccoid from by exposure to metronidazole.Both spiral and coccoid form of H. pylori were tested for the urease activity, the adherence to Hep-2 cells and the vacuolating cytotoxicity to Hela cells, and the differences of the protein were analysed by SDS-PAGE and Western blot,The mutation of the genes including ureA, ureB,hpaA; vacA and cagA, related with virulence,was detected by means of PCR and PCR-SSCP.RESULTS In the coccoid H. pylori, the urease activity, the adherence to Hep-2 cells and the vacuolating cytotoxicity to Hela cells alldecreased. In strain F44, the rate and index of adherence reduced from 70.0% ± 5.3% to 33% ±5.1% and from 2.6 ±0.4 to 0.96 ±0.3 (P<0.01),respectively. The invasion of coccoid H. pylori into Hep-2 cell could be seen under electronmicroscope. SDS-PAGE showed that the content of the protein with the molecular weight over Mr74 000 decreased, and the hybriditional signal in band Mr 125 000 weakened, while the band Mr 110000 and Mr63000 strengthened in coccoid H. pylori as shown in Western blot. | 13 | Mycoplasma infections and different human carcinomas显示文摘AIM To explore relationships between human carcinomas and mycoplasma infection.METHODS Monoclonal antibody PD4, which specifically recognizes a distinct protein from mycoplasma hyorhinis, was used to detect mycoplasma infection in different paraffinembedded carcinoma tissues with immunohistochemistry. PCR was applied to amplify the mycoplasma DNA from the positive samples for confirming immunohistochemistry.RESULTS Fifty of 90 cases (56%) of gastric carcinoma were positive for mycoplasma hyorhinis. In other gastric diseases, the mycoplasma infection ratio was 28% (18/49) in chronic superficial gastritis, 30% (14/ 46) in gastric ulcer and 37% (18/ 49) in intestinal metaplasia. The difference is significant with gastric cancer (X2=12.06, P<0.05). In colon carcinoma, the mycoplasma infection ratio was 55.1% (32/58), but it was 20.9% (10/49) in adenomarous polyp (X2=13.46, P<0.005).Gastric and colon cancers with high differentiation had a higher mycoplasma infection ratio than those with low differentiation (P< 0.05). Mycoplasma infection in esophageal cancer, lung cancer, breast cancer and glioma was 50.9% (27/53), 52.6% (31/ 59), 39.7%(25/63) and 41% (38/91), respectively. The mycoplasma DNA was successfully amplified with the DNA extracted from the cancer tissues that were positive for mycoplasma infection (detected with antibody PD4).CONCLUSION There was high correlation between mycoplasma infection and different cancers, which suggests the possibility of an association between the two. | 14 | Clostridium difficile infections in China显示文摘Clostridium difficile (C. difficile) infection has become one of the major hospital-associated infections in West-ern countries in the last two decades. However, there is limited information on the status of C. difficile infection inChinese healthcare settings. Given the large and increasing elderly population and the well-recognized problem ofover-prescribing of broad spectrum antibiotics in China, it is critical to understand the epidemiology and potentialrisk factors that may contribute to C. difficile infection in China. A literature review of available published studies,including those in Chinese language-based journals, was conducted. A review of the currently available literaturesuggested the presence of C. difficile infections in China, but also suggested that these infections were not particu-larly endemic. This finding should lead to better designed and greatly expanded studies to provide a more reliableepidemiologically-based conclusion on the actual status of C. difficile infection in China, including the identifica-tion of any associated risk factors. | 16 | Research progress on electrical signals in higher plants显示文摘This review introduces the characteristics of electrical signals in higher plants and their corresponding physiological significance, and describes in detail the impact of environmental factors (e.g. light and temperature) on the electrical potential of the plants. Also, we evaluate the measurement techniques used for electrical signals in plants, including intracellular measurement, extracellular measurement, measurement of the ion channel based on the patch-clamp technique and on the non-invasive microelectrode vibrating probe technique. We also give a brief review of the applications of these methods for investigating electrical signals in plants. The ionic mechanism of electrical activity in plants is then discussed in terms of environmental response in higher plants, and this is used to provide a theoretical basis for quantitative description of the electrical signals in plants. | 20 | Current progress in epigenetic research for hepato-carcinomagenesis显示文摘Hepatocellular carcinoma is the main type of primary liver cancer,and also one of the most malignant tumors.At present,the pathogenesis mechanisms of liver cancer are not entirely clear.It has been shown that inactivation of tumor suppressor genes and activation of oncogenes play a significant role in carcinogenesis,caused by the genetic and epigenetic aberrance.In the past,people generally thought that genetic mutation is a key event of tumor pathogenesis,and somatic mutation of tumor suppressor genes is in particular closely associated with oncogenesis.With deeper understanding of tumors in recent years,increasing evidence has shown that epigenetic silencing of those genes,as a result of aberrant hypermethylation of CpG islands in promoters and histone modification,is essential to carcinogenesis and metastasis.The term epigenetics refers to heritable changes in gene expression caused by regulation mechanisms,other than changes in the underlying DNA sequence.Specific epigenetic processes include DNA methylation,genome imprinting,chromotin remodeling,histone modification and microRNA regulations.This paper reviews recent epigenetics research progress in the hepatocellular carcinoma study,and tries to depict the relationships between hepatocellular carcinomagenesis and DNA methylation as well as microRNA regulation. | HUANG Jian National Human Genome Research Center at Shanghai-Ministry of Science and Technology to build a healthy and disease Ge-nomics Laboratory,Shanghai 201203,China | 2009 | Science China(Life Sciences)2009,52,1: | 9 |
