| 1 | 变性梯度凝胶电泳(DGGE)在微生物生态学中的应用显示文摘由于从环境样品中分离和培养细菌的困难 ,分子生物学方法已发展用来描述和鉴定微生物群落。近年来基于 DNA方法的群落分析得到了迅速的发展 ,如 PCR扩增技术 ,克隆文库法 ,荧光原位杂交法 ,限制性酶切片段长度多态性法 ,变性和温度梯度凝胶电泳法。DGGE已广泛用于分析自然环境中细菌、蓝细菌 ,古菌、微微型真核生物、真核生物和病毒群落的生物多样性。这一技术能够提供群落中优势种类信息和同时分析多个样品。具有可重复和容易操作等特点 ,适合于调查种群的时空变化 ,并且可通过对切下的带进行序列分析或与特异性探针杂交分析鉴定群落成员。DGGE分析微生物群落的一般步骤如下 :一是核酸的提取 ,二是 1 6Sr RNA,1 8S r RNA或功能基因如可容性甲烷加单氧酶羟化酶基因 ( mmo X)和氨加单氧酶 α-亚单位基因 ( amo A)片段的扩增 ,三是通过DGGE分析 PCR产物。DGGE使用具有化学变性剂梯度的聚丙烯酰胺凝胶 ,该凝胶能够有区别的解链PCR扩增产物。由 PCR产生的不同的 DNA片段长度相同但核苷酸序列不同。因此不同的双链 DNA片段由于沿着化学梯度的不同解链行为将在凝胶的不同位置上停止迁移 [5] 。 DNA解链行为的不同导致一个凝胶带图案 ,该图案是微生物群落中主要种类的一个轮廓。 DGGE使用所有生物中保守的基因? | 马悦欣 Carola Holmstrm Jeremy Webb Staffan Kjelleberg | 2003 | 生态学报2003,23,8: | 100 |

| 6 | Gastrointestinal neuromuscular apparatus: An underestimated target of gut microbiota显示文摘Over the last few years, the importance of the resident intestinal microbiota in the pathogenesis of several gastrointestinal diseases has been largely investigated. Growing evidence suggest that microbiota can influence gastrointestinal motility. The current working hypothesis is that dysbiosis-driven mucosal alterations induce the production of several inflammatory/immune mediators which affect gut neuro-muscular functions. Besides these indirect mucosal-mediated effects, the present review highlights that recent evidence suggests that microbiota can directly affect enteric nerves and smooth muscle cells functions through its metabolic products or bacterial molecular components translocated from the intestinal lumen. Tolllike receptors, the bacterial recognition receptors, are expressed both on enteric nerves and smooth muscle and are emerging as potential mediators between microbiota and the enteric neuromuscular apparatus. | 8 | Expression of HER2 and bradykinin B_1 receptors in precursor lesions of gallbladder carcinoma显示文摘AIM:To determine the expression of HER2 and bradykinin B 1 receptors(B 1 R) in the two pathogenic models of gallbladder cancer:the metaplasia-dysplasia-carcinoma and the adenoma-carcinoma pathways.METHODS:Receptor proteins were visualized by immunohistochemistry on 5-μm sections of paraffin-embedded tissue.Expression of both receptors was studied in biopsy samples from 92 patients(6 males and 86 females;age ranging from 28 to 86 years,mean 56 years).High HER2 expression in specimens was additionally investigated by fluorescence in situ hybridiza-tion.Cell proliferation in each sample was assessed by using the Ki-67 proliferation marker.RESULTS:HER2 receptor protein was absent in adenomas and in normal gallbladder epithelium.On the contrary,there was intense staining for HER2 on the basolateral membrane of epithelial cells of intestinal metaplasia(22/24;91.7%) and carcinoma in situ(9/10;90%),the lesions that displayed a significantly high proliferation index.Protein up-regulation of HER2 in the epithelium with metaplasia or carcinoma in situ was not accompanied by HER2 gene amplification.A similar result was observed in invasive carcinomas(0/12).The B 1 R distribution pattern mirrored that of HER2 except that B 1 R was additionally observed in the adenomas.The B 1 R appeared either as cytoplasmic dots or labeling on the apical cell membrane of the cells composing the epithelia with intestinal metaplasia(24/24;100%) and carcinoma in situ(10/10;100%) and in the epithelial cells of adenomas.In contrast,both HER2(4/12;33%) and B 1 R(1/12;8.3%) showed a low expression in invasive gallbladder carcinomas.CONCLUSION:The up-regulation of HER2 and B1R in precursor lesions of gallbladder carcinoma suggests cross-talk between these two receptors that may be of importance in the modulation of cell proliferation in gallbladder carcinogenesis. | Cesar Toledo Carola E Matus Ximena Barraza Pamela Arroyo Pamela Ehrenfeld Carlos D Figueroa Kanti D Bhoola Maeva del Pozo Maria T Poblete | 2012 | World Journal of Gastroenterology2012,18,11: | 6 |

| 10 | Partial external biliary diversion in bile salt export pump deficiency: Association between outcome and mutation显示文摘AIM To investigate the relation of two different mutations to the outcome of partial external biliary diversion(PEBD)in severe bile salt export pump(BSEP) deficiency.METHODS Mutations in the gene encoding BSEP leading to severe BSEP deficiency in two unrelated patients were identified by genomic sequencing. Native liver biopsies and transiently transfected human embryonic kidney(HEK) 293 cells expressing either wild-type or mutated BSEP were subjected to immunofluorescence analysis to assess BSEP transporter localization. Bile acid profiles of patient and control bile samples were generated by ultra-performance liquid chromatographytandem mass spectrometry. Wild-type and mutant BSEP transport of [~3H]-labeled taurocholate(TC) and taurochenodeoxycholate(TCDC) was assessed by vesicular transport assays.RESULTS A girl(at 2 mo) presented with pruritus, jaundice and elevated serum bile salts(BS). PEBD stabilized liver function and prevented liver transplantation. She was heterozygous for the BSEP deletion p.T919 del and the nonsense mutation p.R1235 X. At the age of 17 years relative amounts of conjugated BS in her bile were normal, while total BS were less than 3% as compared to controls. An unrelated boy(age 1.5 years) presenting with severe pruritus and elevated serum BS was heterozygous for the same nonsense and another missense mutation, p.G1032 R. PEBD failed to alleviate pruritus, eventually necessitating liver transplantation. BS concentration in bile was about 5% of controls. BS were mainly unconjugated with an unusual low amount of chenodeoxycholate derivatives(< 5%). The patients' native liver biopsies showed canalicular BSEP expression. Both BSEP p.T919 del and p.G1032 R were localized in the plasma membrane in HEK293 cells. In vitro transport assays showed drastic reduction of transport by both mutations. | 11 | Proton pump inhibitors and dysbiosis: Current knowledge and aspects to be clarified显示文摘Proton pump inhibitors (PPIs) are common medications within the practice of gastroenterology. These drugs, which act through the irreversible inhibition of the hydrogen/potassium pump (H+/K+-ATPase pump) in the gastric parietal cells, are used in the treatment of several acid-related disorders. PPIs are generally well tolerated but, through the long-term reduction of gastric acid secretion, can increase the risk of an imbalance in gut microbiota composition (i.e., dysbiosis). The gut microbiota is a complex ecosystem in which microbes coexist and interact with the human host. Indeed, the resident gut bacteria are needed for multiple vital functions, such as nutrient and drug metabolism, the production of energy, defense against pathogens, the modulation of the immune system and support of the integrity of the gut mucosal barrier. The bacteria are collected in communities that vary in density and composition within each segment of the gastrointestinal (GI) tract. Therefore, every change in the gut ecosystem has been connected to an increased susceptibility or exacerbation of various GI disorders. | 13 | Dietary inclusion of a partially defatted black soldier fly(Hermetia illucens)larva meal in low fishmeal-based diets for rainbow trout(Oncorhynchus mykiss)显示文摘Background:Recent investigations highlighted that Hermetia illucens(BSF)larva meal can be a valuable alternative protein source for aquafeed production.In this study,in substitution of fishmeal,we used increasing inclusion levels of a partially defatted BSF larva meal in low fishmeal-based diets for rainbow trout(Oncorhynchus mykiss Walbaum)and we evaluated the related implications in terms of growth performance,physical characteristics,proximate and fatty acid(FA)compositions of the fillets,gut and liver histology,and diet digestibility.In a 131-day trial,576 fish(100.1±9.29 g)were randomly allotted to 24 tanks(24 fish/tank,4 replicates/treatment).Six experimental diets were produced to have partial replacement of fishmeal(0,10%,20%,30%,40%and 50%)by increasing levels of BSF meal(0%[BSF0],3%[BSF3],6%[BSF6],9%[BSF9],12%[BSF12]and 15%[BSF15]on as fed basis,respectively).Results:No differences were observed among the treatments for all the considered growth performance parameters.The viscero and hepato-somatic indexes showed significant differences among the treatments,with the highest values observed in the BSF15 group.No differences were recorded in terms of fillet’s physical characteristics,dry matter(DM),crude protein(CP)and ether extract(EE)contents.Total saturated and monounsaturated FA increased,while polyunsaturated FA(particularly n-3 FA)decreased while increasing the HI meal inclusion in the diet.Histopathology of liver and gut was not affected,whereas,in posterior gut,villi were higher in BSF6 and BSF9 compared to BSF3 fish.The apparent digestibility of DM,CP,EE and gross energy did not vary among the treatments.Conclusions:These results suggest that a partially defatted BSF meal could be included up to 15%in low fishmeal-based diets for rainbow trout with no adverse effects on growth performance,fillet’s physical characteristics,gut and liver health,and diet digestibility.On the contrary,the fillet FA composition worsened while increasing the level of BSF meal in the diet. | Christian Caimi Ilaria Biasato Giulia Chemello Sara Bellezza Oddon Carola Lussiana Vanda Maria Malfatto Maria Teresa Capucchio Elena Colombino Achille Schiavone Francesco Gai Angela Trocino Alberto Brugiapaglia Manuela Renna Laura Gasco | 2021 | Journal of Animal Science and Biotechnology2021,12,4: | 3 |

| 15 | Recurrence rates after endoscopic resection of large colorectal polyps:A systematic review and meta-analysis显示文摘BACKGROUND Complete polyp resection is the main goal of endoscopic removal of large colonic polyps.Resection techniques have evolved in recent years and endoscopic submucosal dissection(ESD),endoscopic mucosal resection(EMR)with margin ablation,cold snare polypectomy(CSP),cold EMR,and underwater EMR have been introduced.Yet,efficacy of these techniques with regard to local recurrence rates(LRRs)vs traditional hot snare polypectomy and standard EMR remains unclear.AIM To analyze LRR of large colonic polyps in a systematic review and meta-analysis.METHODS MEDLINE,EMBASE,EBM Reviews,and CINAHL were searched for prospective studies reporting LRR or incomplete resection rate(IRR)after colonic polypectomy of polyps≥10 mm,published between January 2011 and July 2021.Primary outcome was LRR for polyps≥10 mm.RESULTS Six thousand nine hundred and twenty-eight publications were identified,of which 34 prospective studies were included.LRR for polyps≥10 mm at up to 12 mo’follow-up was 11.0%(95%CI,7.1%-14.8%;15 studies;4904 polyps).ESD(1.7%;95%CI,0%-3.4%;3 studies,221 polyps)and endoscopic mucosal resection with margin ablation(3.3%;95%CI,2.2%-4.5%;2 studies,947 polyps)significantly reduced LRR vs standard EMR without(15.2%;95%CI,12.5%-18.0%;4 studies,650 polyps)or with unsystematic margin ablation(16.5%;95%CI,15.2%-17.8%;6 studies,3031 polyps).CONCLUSION LRR is significantly lower after ESD or EMR with routine margin ablation;thus,these techniques should be considered standard for endoscopic removal of large colorectal polyps.Other techniques,such as CSP,cold EMR,and underwater EMR require further evaluation in prospective studies before their routine implementation in clinical practice can be recommended. | Carola Rotermund Roupen Djinbachian Mahsa Taghiakbari Markus D Enderle Axel Eickhoff Daniel von Renteln | 2022 | World Journal of Gastroenterology2022,28,29: | 3 |
