1Big Earth Data:a new challenge and opportunity for Digital Earth’s development显示文摘Digital Earth has seen great progress during the last 19 years.When it entered into the era of big data,Digital Earth developed into a new stage,namely one characterized by‘Big Earth Data’,confronting new challenges and opportunities.In this paper we give an overview of the development of Digital Earth by summarizing research achievements and marking the milestones of Digital Earth’s development.Then,the opportunities and challenges that Big Earth Data faces are discussed.As a data-intensive scientific research approach,Big Earth Data provides a new vision and methodology to Earth sciences,and the paper identifies the advantages of Big Earth Data to scientific research,especially in knowledge discovery and global change research.We believe that Big Earth Data will advance and promote the development of Digital Earth.Huadong Guo Zhen Liu Hao Jiang Changlin Wang Jie Liu Dong Liang 2017International Journal of Digital Earth2017,10,1:5
2Arterial relaxation is coupled to inhibition of mitochondrial fission in arterial smooth muscle cells:comparison of vasorelaxant effects of verapamil and phentolamine显示文摘Mitochondria are morphologically dynamic organelles which undergo fission and fusion processes. Our previous study found that arterial constriction was always accompanied by increased mitochondrial fission in smooth muscle cells, whereas inhibition of mitochondrial fission in smooth muscle cells was associated with arterial relaxation. Here, we used the typical vasorelaxants, verapamil and phentolamine, to further confirm the coupling between arterial constriction and mitochondrial fission in rat aorta. Results showed that phentolamine but not verapamil induced vasorelaxation in phenylephrine(PE)-induced rat thoracic aorta constriction. Verapamil, but not phentolamine, induced vasorelaxation in high K^+(KPSS)-induced rat thoracic aorta constriction. Pre-treatment with phentolamine prevented PEbut not KPSS-induced aorta constriction and pre-treatment with verapamil prevented both PE-and KPSSinduced aorta constriction. Transmission electron microscopy(TEM) results showed that verapamil but not phentolamine inhibited KPSS-induced excessive mitochondrial fission in aortic smooth muscle cells,and verapamil prevented both PE-and KPSS-induced excessive mitochondrial fission in aortic smoothmuscle cells. Verapamil inhibited KPSS-induced excessive mitochondrial fission in cultured vascular smooth muscle cells(A10). 3Integrated transcriptome and metabolite profiling highlights the role of benzoxazinoids in wheat resistance against Fusarium crown rot显示文摘Fusarium crown rot(FCR), caused by Fusarium spp., is a chronic and severe plant disease worldwide. In the last years, the incidence and severity of FCR in China has increased to the point that it is now considered a threat to local wheat crops. In this study, for the first time, the metabolites and transcripts responsive to FCR infection in the partial resistant wheat cultivar 04 Zhong 36(04 z36) and susceptible cultivar Xinmai 26(XM) were investigated and compared at 20 and 25 days post inoculation(dpi). A total of 443 metabolites were detected, of which 102 were significantly changed because of pathogen colonization.Most of these 102 metabolites belonged to the flavonoid, phenolic acid, amino acid and derivative classes.Some metabolites, such as proline betaine, lauric acid, ribitol, and arabitol, were stably induced by Fusarium pseudograminearum(Fp) infection at two time points and may have important roles in FCR resistance. In line with the reduced seedling height of 04 z36 and XM plants, RNA-seq analysis revealed that FCR infection significantly affected the photosynthesis activities in two cultivars. Furthermore, 15 jasmonate ZIM-domain genes(JAZ) in the significantly enriched ‘regulation of jasmonic acid mediated signaling pathway’ in 04 z36 were down-regulated. The down-regulation of these JAZ genes in 04 z36 may cause a strong activation of the jasmonate signaling pathway. Based on combined data from gene expression and metabolite profiles, two metabolites, benzoxazolin-2-one(BOA) and 6-methoxy-benzoxazolin-2-one(MBOA), involved in the benzoxazinoid-biosynthesis pathway, were tested for their effects on FCR resistance. Both BOA and MBOA significantly reduced fungal growth in vitro and in vivo, and, thus, a higher content of BOA and MBOA in 04 z36 may contribute to FCR resistance. 4Review of the development of digital earth research during 1998–2015 based on a bibliometric analysis显示文摘Since Al Gore created the vision for Digital Earth in 1998,a wide range of research in this field has been published in journals.However,little attention has been paid to bibliometric analysis of the literature on Digital Earth.This study uses a bibliometric analysis methodology to study the publications related to Digital Earth in the Science Citation Index database and Social Science Citation Index database(via the Web of Science online services)during the period from 1998 to 2015.In this paper,we developed a novel keyword set for‘Digital Earth’.Using this keyword set,11,061 scientific articles from 23 subject categories were retrieved.Based on the searched articles,we analyzed the spatiotemporal characteristics of publication outputs,the subject categories and the major journals.Then,authors’performance,affiliations,cooperation,and funding institutes were evaluated.Finally,keywords were examined.Through keyword clustering,research hotspots in the field of Digital Earth were detected.We assume that the results coincide well with the position of Digital Earth research in the context of big data.Zhen Liu Changlin Wang Qiao Lin Yang Liu 2017International Journal of Digital Earth2017,10,7:0
5Adenovirus-mediated hypoxia-inducible factor-1 alpha gene transfer induces angiogenesis and neurogenesis following cerebral ischemia in rats显示文摘BACKGROUND: Hypoxia-inducible factor-1 (HIF-1) accumulates under conditions of hypoxia. HIF-1α target genes have pleiotropic effects on neurogenesis, neuroprotection and angiogenesis in the brain. OBJECTIVE: To investigate whether a recombinant adenovirus carrying HIF-1α can increase the expres- sion of HIF-1α in vivo and thus promote angiogenesis and neurogenesis in a rat model of focal cerebral ischemia. DESIGN, TIME AND SETTING: The randomized, controlled experiment was performed at the Department of Neurobiology, Third Military Medical University of Chinese PLA from September 2006 to October 2007. MATERIALS: 68 healthy adult male Sprague-Dawley (SD) rats, weighing 230-250 g, were used. HIF-1α antibody was purchased from Wuhan Boster Company. Vascular endothelial growth factor (VEGF) antibody was purchased from Santa Cruz Biotech Company. METHODS: All 68 rats were induced with a transient middle cerebral artery occlusion (MCAO), according to the method of intra-luminal vascular occlusion. 54 rats, in which MCAO was successfully induced, were randomly divided into adenovirus (Ad) group and recombinant adenovirus with HIF-1αgene (Ad-HIF-1α) group (27 rats for each group). Rats were injected with 10 μL Ad (Ad group) or Ad-HIF-1α (Ad-HIF-1α group) into the lateral ventricle, 1 day after MCAO induction. MAIN OUTCOME MEASURES: Reverse transcription polymerase chain reaction was used to measure the expression of HIF-1α and of VEGF. Immunohistochemistry was used to detect the localization of HIF-1α, VEGF and factor Ⅷ in ischemic penumbra. Rat newborn nerve cells were labeled with 5-bromodeoxyuridine (BrdU) after ischemia. BrdU/neurofilament 200 (NF200) and BrdU/glial fibrillary acidic protein (GFAP) double labeled immunofluorescent histochemistry was used to identify the differentiation of newborn cells. Neurological function was evaluated using the modified neurological severity score (NSS). RESULTS: Compared with Ad, Ad-HIF-1αenhanced the expression of HIF-1αand VEGF (P < 0.01). The numbers of factor VIII, BrdU, BrdU/NF200 and BrdU/GFAP positive cells were increased significantly (P < 0.01) in the Ad-HIF-1α group compared to the Ad group. Levels of HIF-1α and VEGF mRNA in the Ad-HIF-1α group were enhanced compared with those in the Ad group. NSS scores of the Ad-HIF-1α group were superior to those of the Ad group at days 7, 14, 21, and 28 after MCAO (P < 0.05).
