1Effect of intravenous transplantation of bone marrow mesenchymal stem cells on neurotransmitters and synapsins in rats with spinal cord injury显示文摘Bone marrow mesenchymal stem cells were isolated, purified and cultured in vitro by Percoll density gradient centrifugation combined with the cell adherence method. Passages 3-5 bone marrow mesenchymal stem cells were transplanted into rats with traumatic spinal cord injury via the caudal vein. Basso-Beattie-Bresnahan scores indicate that neurological function of experimental rats was significantly improved over transplantation time (1-5 weeks). Expressions of choline acetyltransferase, glutamic acid decarboxylase and synapsins in the damaged spinal cord of rats was significantly increased after transplantation, determined by immunofluorescence staining and laser confocal scanning microscopy. Bone marrow mesenchymal stem cells that had migrated into the damaged area of rats in the experimental group began to express choline acetyltransferase, glutamic acid decarboxylase and synapsins, 3 weeks after transplantation. The Basso-Beattie- Bresnahan scores positively correlated with expression of choline acetyltransferase and synapsins. Experimental findings indicate that intravenously transplanted bone marrow mesenchymal stem cells traverse into the damaged spinal cord of rats, promote expression of choline acetyltransferase, glutamic acid decarboxylase and synapsins, and improve nerve function in rats with spinal cord injury.Shaoqiang Chen Bilian Wu Jianhua Lin 2012Neural Regeneration Research2012,7,19:5

3Soluble CD163 and CD163 Expression on Monocytes Associated with Chronic Hepatitis B Inflammation and HBsAg Loss显示文摘Background and Aims:Monocyte/macrophage-associat-ed CD163 is an indicator of the severity of liver inflam-mation and cirrhosis,but the difference of soluble CD163(sCD163)levels in chronic hepatitis B(CHB)patients and hepatitis B surface antigen(HBsAg)-loss patients is un-clear.Herein,we aimed to compare the sCD163 levels in CHB patients and HBsAg-loss patients with or without an-tiviral treatment.Methods:sCD163 and CD163 expres-sion on monocytes were compared among four groups,healthy subjects,treatment-naïve CHB patients,sponta-neous HBsAg-loss patients,and treatment-related HBsAg-loss patients.The correlation between sCD163 levels and clinical parameters in CHB patients was analyzed.A group of 80 patients with hepatitis B virus(HBV)infection and liver biopsy were recruited.Results:sCD163 levels were higher in the CHB group than in the other three groups.sCD163 levels were higher in treatment-related HBsAg-loss patients than in spontaneous HBsAg-loss patients.sCD163 levels were negatively correlated with hepatitis B e-antigen(HBeAg)and HBsAg levels in HBeAg-positive patients.Liv-er biopsy results further demonstrated that sCD163 levels were elevated in CHB patients with substantial inflamma-tion(A≥2)or fibrosis(F≥2).The sCD163 model was more sensitive in predicting inflammation than other noninvasive models.Its levels were higher in patients with normal ala-nine aminotransferase levels and significant inflammation(A≥2)than in patients with no or mild inflammation.Con-clusions:sCD163 and CD163 expression on monocytes were associated with CHB inflammation and HBsAg loss,and may be used as markers to predict HBV-specific im-mune activation.Peilin Xie Bilian Yao Dao Huang Yongyan Chen Qiming Gong Xinxin Zhang 2022Journal of Clinical and Translational Hepatology2022,10,6:2

10HDL quality features revealed by proteome–lipidome connectivity are associated with atherosclerotic disease显示文摘Lipoprotein,especially high-density lipoprotein(HDL),particles are composed of multiple heterogeneous subgroups containing various proteins and lipids.The molecular distribution among these subgroups is closely related to cardiovascular disease(CVD).Here,we established high-resolution proteomics and lipidomics(HiPL)methods to depict the molecular profiles across lipoprotein(Lipo-HiPL)and HDL(HDL-HiPL)subgroups by optimizing the resolution of anion-exchange chromatography and comprehensive quantification of proteins and lipids on the omics level.Furthermore,based on the Pearson correlation coefficient analysis of molecular profiles across high-resolution subgroups,we achieved the relationship of proteome–lipidome connectivity(PLC)for lipoprotein and HDL particles.By application of these methods to high-fat,high-cholesterol diet-fed rabbits and acute coronary syndrome(ACS)patients,we uncovered the delicate dynamics of the molecular profile and reconstruction of lipoprotein and HDL particles.Of note,the PLC features revealed by the HDL-HiPL method discriminated ACS from healthy individuals better than direct proteome and lipidome quantification or PLC features revealed by the Lipo-HiPL method,suggesting their potential in ACS diagnosis.Together,we established HiPL methods to trace the dynamics of the molecular profile and PLC of lipoprotein and even HDL during the development of CVD.Dandan Wang Bilian Yu Qingrun Li Yanhong Guo Tomonari Koike Yui Koike Qingqing Wu Jifeng Zhang Ling Mao Xiaoyu Tang Liang Sun Xu Lin Jiarui Wu Y.Eugene Chen Daoquan Peng Rong Zeng 2022Journal of Molecular Cell Biology2022,14,3:0
11Cancer cell employs a microenvironmental neural signal transactivating nucleus-mitochondria coordination to acquire stemness显示文摘Cancer cell receives extracellular signal inputs to obtain a stem-like status,yet how tumor microenvironmental(TME)neural signals steer cancer stemness to establish the hierarchical tumor architectures remains elusive.Here,a pan-cancer transcriptomic screening for 10852 samples of 33 TCGA cancer types reveals that cAMP-responsive element(CRE)transcription factors are convergent activators for cancer stemness.Deconvolution of transcriptomic profiles,specification of neural markers and illustration of norepinephrine dynamics uncover a bond between TME neural signals and cancer-cell CRE activity.Specifically,neural signal norepinephrine potentiates the stemness of proximal cancer cells by activating cAMP-CRE axis,where ATF1 serves as a conserved hub.Upon activation by norepinephrine,ATF1 potentiates cancer stemness by coordinated trans-activation of both nuclear pluripotency factors MYC/NANOG and mitochondrial biogenesis regulators NRF1/TFAM,thereby orchestrating nuclear reprograming and mitochondrial rejuvenating.Accordingly,single-cell transcriptomes confirm the coordinated activation of nuclear pluripotency with mitochondrial biogenesis in cancer stem-like cells.These findings elucidate that cancer cell acquires stemness via a norepinephrine-ATF1 driven nucleus-mitochondria collaborated program,suggesting a spatialized stemness acquisition by hijacking microenvironmental neural signals.Bin He Rui Gao Shasha Lv Ailin Chen Junxiu Huang Luoxuan Wang Yunxiu Feng Jiesi Feng Bing Liu Jie Lei Bing Deng Bin He Bai Cui Fei Peng Min Yan Zifeng Wang Eric W-F Lam Bilian Jin Zhiming Shao Yulong Li Jianwei Jiao Xi Wang Quentin Liu 2023Signal Transduction and Targeted Therapy2023,8,8:0
