| 1 | Substrate stiffness modulates bone marrow-derived macrophage polarization through NF-κB signaling pathway显示文摘The stiffness of the extracellular matrix(ECM)plays an important role in regulating the cellular programming.However,the mechanical characteristics of ECM affecting cell differentiation are still under investigated.Herein,we aimed to study the effect of ECM substrate stiffness on macrophage polarization.We prepared polyacrylamide hydrogels with different substrate stiffness,respectively.After the hydrogels were confirmed to have a good biocompatibility,the bone marrow-derived macrophages(BMMs)from mice were incubated on the hydrogels.With simulated by the low substrate stiffness,BMMs displayed an enhanced expression of CD86 on the cell surface and production of reactive oxygen species(ROS)in cells,and secreted more IL-1βand TNF-αin the supernatant.On the contrary,stressed by the medium stiffness,BMMs expressed more CD206,produced less ROS,and secreted more IL-4 and TGF-β.In vivo study by delivered the hydrogels subcutaneously in mice,more CD68+CD86+cells around the hydrogels with the low substrate stiffness were observed while more CD68+CD206+cells near by the middle stiffness hydrogels.In addition,the expressions of NIK,phosphorylated p65(pi-p65)and phosphorylated IκB(pi-IκB)were significantly increased after stimulation with low stiffness in BMMs.Taken together,these findings demonstrated that substrate stiffness could affect macrophages polarization.Low substrate stiffness promoted BMMs to shift to classically activated macrophages(M1)and the middle one to alternatively activated macrophages(M2),through modulating ROS-initiated NF-κB pathway.Therefore,we anticipated ECM-based substrate stiffness with immune modulation would be under consideration in the clinical applications if necessary. | Mimi Chen Yu Zhang Pinghui Zhou Xingzhi Liu Huan Zhao Xichao Zhou Qiaoli Gu Bin Li Xuesong Zhu Qin Shi | 2020 | Bioactive Materials2020,5,4: | 7 |
| 2 | Archean-Paleoproterozoic Lithospheric Mantle at the Northern Margin of the North China Craton Represented by Tectonically Exhumed Peridotites显示文摘Tectonically emplaced peridotites from North Hebei Province, North China Craton, have retained an original harzburgite mineral assemblage of olivine(54%–58%) + orthopyroxene(40%–46%)+minor clinopyroxene(<1%)+spinel. Samples with boninite-like chemical compositions also coexist with these peridotites. The spinels within the peridotites have high-Al end-members with Al_2O_3 content of 30 wt % –50 wt %, typical of mantle spinels. When compared with experimentally determined melt extraction trajectories, the harzburgites display a high degree of melting and enrichment of SiO_2, which is typical of cratonic mantle peridotites. The peridotites display variably enriched light rare earth elements(REEs), relatively depleted middle REEs and weakly fractionated heavy REEs, which suggest a melt extraction of over 25% in the spinel stability field. The occurrence of arc-and SSZ-type chromian spinels in the peridotites suggests that melt extraction and metasomatism occurred mostly in a subduction-related setting. This is also supported by the geochemical data of the coexisting boninite-like samples. The peridotites have ^(187)Os/^(188)Os ratios ranging from 0.113–0.122, which is typical of cratonic lithospheric mantle. These ^(187)Os/^(188)Os ratios yield model melt extraction ages(TRD) ranging from 981 Ma to 2054 Ma, which may represent the minimum estimation of the melt extraction age. The Al_2O_3-^(187)Os/^(188)Os-proxy isochron ages of 2.4 Ga–2.7 Ga suggest a mantle melt depletion age between the Late Achaean and Early Paleoproterozoic. | 3 | Activation of Piezo1 contributes to matrix stiffness-induced angiogenesis in hepatocellular carcinoma显示文摘Background:Despite integrin being highlighted as a stiffness-sensor molecule in matrix stiffness-driven angiogenesis,other stiffness-sensor molecules and their mechanosensory pathways related to angiogenesis in hepatocellular carcinoma(HCC)remain obscure.Here,we explored the interplay between Piezo1 and integrinβ1 in the mechanosensory pathway and their effects on HCC angiogenesis to better understand matrix stiffness-induced angiogenesis.Methods:The role of Piezo1 in matrix stiffness-induced angiogenesis was investigated using orthotopic liver cancer SD rat models with high liver stiffness background,and its clinical significance was evaluated in human HCC tissues.Matrix stiffness-mediated Piezo1 upregulation and activation were assayed using an in vitro fibronectin(FN)-coated cell culture system with different stiffness,Western blotting and Ca^(2+)probe.The effects of shPiezo1-conditioned medium(CM)on angiogenesis were examined by tube formation assay,wound healing assay and angiogenesis array.The underlying mechanism by which Piezo1 participated in matrix stiffness-induced angiogenesis was analyzed by microRNA quantitative real-time polymerase chain reaction(qRT-PCR),matrix stiffness measurement,dual-luciferase reporter assay,ubiquitination assay and co-immunoprecipitation.Results:Increased matrix stiffness significantly upregulated Piezo1 expression at both cellular and tissue levels,and high expression of Piezo1 indicated an unfavorable prognosis.High matrix stiffness also noticeably enhanced the activation level of Piezo1,similar to its expression level.Piezo1 knockdown significantly suppressed tumor growth,angiogenesis,and lung metastasis of HCC rat models with high liver stiffness background.shPiezo1-CM from HCC cells attenuated tube formation and migration abilities of vascular endothelial cells remarkably,and analysis of differentially expressed pro-angiogenic factors revealed that Piezo1 promoted the expression and secretion of vascular endothelial growth factor(VEGF),CXC chemokine ligand 16(CXCL16)and insulin-like growth factor binding protein 2(IGFBP2).Matrix stiffness-caused Piezo1 upregulation/activation restrained hypoxia inducible factor-1α(HIF-1α)ubiquitination,subsequently enhanced the expression of downstream pro-angiogenic factors to accelerate HCC angiogenesis.Besides,collagen 1(COL1)-reinforced tissue stiffening resulted in more expression of Piezo1 via miR-625-5p.Conclusions:This study unravels a new mechanism by which the inte-grinβ1/Piezo1 activation/Ca2+influx/HIF-1αubiquitination/VEGF,CXCL16 and IGFBP2 pathway participates in matrix stiffness-driven HCC angiogenesis.Simultaneously,a positive feedback regulation loop as stiff matrix/integrinβ1/miR-625-5p/Piezo1 and COL1/stiffer matrix mediates matrix stiffness-caused Piezo1 upregulation. | Miao Li Xi Zhang Mimi Wang Yaohui Wang Jiali Qian Xiaoxia Xing Zhiming Wang Yang You Kun Guo Jie Chen Dongmei Gao Yan Zhao Lan Zhang Rongxin Chen Jiefeng Cui Zhenggang Ren | 2022 | Cancer Communications2022,42,11: | 3 |

| 5 | GSH-induced chemotaxis nanomotors for cancer treatment by ferroptosis strategy显示文摘Overexpression of glutathione(GSH) in tumor cells greatly inhibits the therapy effect of traditional ferroptosis inducers;thus,control of the GSH level is an important way to improve the efficacy of ferroptosis.Herein,a kind of nanomotor based on metal organic framework material NH_(2)-MIL-101 is constructed,in which polyethylene glycol(PEG) and glutathione hydrolase γ-glutamyltransferase(GGT) are asymmetrically modified to obtain mPEG@MIL-101@GGT nanomotors(PMG NMs).The nanomotor proposed in this article can be induced by overexpressing GSH in tumors to form chemotactic effects through the specific affinity between enzymes and substrates.Results indicate that the tail structure provided by PEG and the affinity between GGT and GSH can enable the stable chemotaxis behavior of nanomotors in a complex environment,thus enriching and penetrating deeply at the tumor site.In addition,after loading the ferroptosis inducer Erastin,the system shows a highly effective induction effect of tumor ferroptosis.Erastin in the system can effectively inhibit the synthesis of GSH,and PMG NMS can react with GSH through Fe^(3+)and GGT to promote GSH depletion.The produced Fe^(2+)can generate excessive reactive oxygen species through Fenton reaction,which further promotes the death of tumor cells.Meantime,the chemotaxis behavior of the nanomotors based on the endogenous biochemical reaction of GGT-catalyzed GSH hydrolysis can endow nanomotors with the enhanced delivery and penetration ability in tumors,thus collaboratively enhancing the ferroptosis effect.This strategy designed according to the physiological characteristics of tumors has good biosafety and treatment effect,providing new perspectives for micro/nanomotor and tumor treatment. | Zhiyong Liu Ting Li Nan Li Yujing Wang Lin Chen Xueting Tang Mimi Wan Chun Mao | 2022 | Science China Chemistry2022,65,5: | 1 |

| 14 | Spatial Autocorrelation Analysis of Genetic Structure of Zelkova schneideriana in Mailing Town,Guangxi显示文摘We analyzed the fine-scale spatial genetic structure of the individuals of Zelkova schneideriana,which were classified by age using the spatial autocorrelation method,to quantify spatial patterns of genetic variation within the population and to explore potential mechanisms that determine genetic variation in population. The spatial autocorrelation coefficient( r) at 13 distance classes was determined on the basis of both geographical distance and genetic distance matrix which was derived from co-dominant SSR data using Gen Al Ex software. The results showed that all the individuals of Z. schneideriana exhibited significantly positive spatial genetic structure at distance less than 40 m( the X-intercept was 53. 568),indicating that the average length of the smallest genetic patch for the same genotype clustering of the Z. schneideriana Mailing population was about 50 m. Limited seed dispersal is the main factor that leads to the spatial genetic variation within populations. The individuals in age Class II showed significantly positive spatial genetic structure at distance less than 30 m( the X-intercept was 47. 882),while the individuals in age Class I and age Class III showed no significant spatial genetic structure in any of the spatial distance classes. Due to the infiltration and heterogeneity of GBM, the obstruction of the blood-brain barrier(BBB) and the unique immunosuppressive mechanism, it is hard to achieve significant effects of GBM treatment. Here, a kind of chemotactic nanomotor that loaded with glucose oxidase(GOx) and carboxylated cisplatin(Pt(IV)) prodrug on the L-arginine-derived polymer is proposed. The nanomotors are driven by catalysis of glucose decomposition and the positive chemotaxis towards the GBM microenvironment where inducible nitric oxide synthase and reactive oxygen species are highly expressed. This facilitates the BBB crossing and GBM targeting of the nanomotors. In addition, the released nitric oxide(NO) during propulsion as well as the loaded GOx and Pt(IV) can exert combined NO/starvation/chemotherapy.
