3Identification and Typing of Respiratory Adenoviruses in Guangzhou, Southern China Using a Rapid and Simple Method显示文摘Human adenoviruses (HAdVs), especially HAdV-B3, -E4 and -B7, are associated with Acute Respiratory Disease in Chinese children, and occasionally in adults. In order to establish and document the profiles of the respiratory adenovirus pathogen among children in Guangzhou, Southern China, a rapid, simple and practical method for identification and typing of respiratory adenoviruses was developed and evaluated. One pair of universal PCR primers was designed according to the conserved region of the hexon gene, which can detect not only HAdV-B3, -E4 and -B7, but also HAdV-B14, -F40 and -F41, with a specific 300bp PCR product. Three pairs of type-specific PCR primers were also designed according to the hypervariable regions of the hexon gene to type HAdV-B3, -E4 and -B7 by three independent PCR reactions, making it easy to optimize the PCR conditions. By using this method, one hundred throat swab specimens collected during Oct 2010 to Dec 2011 and suspected of being positive for adenoviral infection were identified and typed for adenoviruses. Of these samples, fifty-five were adenovirus-positive. The most common HAdV type was HAdV-B3, identified in 92.7% of samples, which is not only consistent with the data reported in 2004-2006, but also consistent with the recent report in Hangzhou, eastern China, indicating that HAdV-B3 has been circulating in Guangzhou, and maybe in eastern China, for many years. 6Improving fatigue performance of Ti-6Al-4V alloy via ultrasonic surface rolling process显示文摘The effect of a gradient nanostructured(GNS) surface layer obtained by ultrasonic surface rolling process(USRP) on the fatigue behavior of Ti-6Al-4V alloy has been studied in this paper. Microstructure, surface topography, surface roughness and residual stress measurements were performed to characterize the surface under different conditions. Rotating bending fatigue tests were carried out to evaluate the fatigue behavior of different treatments. The results present a remarkable fatigue performance enhancement for the Ti-6Al-4V alloy with a GNS surface layer obtained by application of USRP with respect to the untreated condition, notwithstanding its considerable surface roughness due to severe ultrasonic impacts and extrusions. Mechanical surface polishing treatment further enhances the beneficial effects of USRP on the fatigue performance. The significantly improved fatigue performance can mainly be ascribed to the compressive residual stress. 8Structural Evolvement of Heating Treatment of Mg/AI-LDH and Preparation of Mineral Mesoporous Materials显示文摘Although hydrotalcite, or layered double hydroxides (LDHs), is not a common mineral, it is an important material that can be easily synthesized in laboratory. In this study, structural evolvement and BET surface area changes of heat treated Mg/Al-LDH is evaluated by XRD, TEM and N2-BET analyses. The results indicate that the magnesium-aluminum LDH with carbonate as interlayer anion, periclase-like oxides was formed at temperatures of 400-800℃. Meanwhile, 2-3 nanometer mesoporous were formed during decomposition of LDH. However, the heat treated samples still preserve the morphology of the original LDH plates. Periclase-like formed from LDH heat treatment may re-hydrolyze and recover the structure of LDH. However, crystallinity of the recovered LDH is lower than that of the original LDH. This heat treatment will result in formation of (Mg, Al)-oxide nano-crystals and nanopores among the nano-crystals. When heating temperature exceeds 1000, the periclase-like (Mg, Al)-oxide is transformed into a composite with periclase (MgO) and spinel phases. The periclase can be re-hydrolyzed and dissolved in HCl solution. After acid treatment, the sample with a high surface area is composed of spinel nano-crystals and nanopores among them. 10Piperlongumine Inhibits Zika Virus Replication In vitro and Promotes Up-Regulation of HO-1 Expression, Suggesting An Implication of Oxidative Stress显示文摘Owing to the widespread distribution of mosquitoes capable of transmitting Zika virus, lack of clinical vaccines and treatments, and poor immunity of populations to new infectious diseases, Zika virus has become a global public health concern. Recent studies have found that Zika virus can continuously infect human brain microvascular endothelial cells.These cells are the primary components of the blood–brain barrier of the cerebral cortex, and further infection of brain tissue may cause severe damage such as encephalitis and fetal pituitary disease. The present study found that a biologically active base, piperlongumine(PL), inhibited Zika virus replication in human brain microvascular endothelial cells, Vero cells, and human umbilical vein endothelial cells. PL also significantly increased heme oxygenase-1(HO-1) gene expression, while silencing HO-1 expression and using the reactive oxygen species scavenger, N-acetylcysteine, attenuated the inhibitory effect of PL on Zika virus replication. These results suggest that PL induces oxidative stress in cells by increasing reactive oxygen species. This, in turn, induces an increase in HO-1 expression, thereby inhibiting Zika virus replication. 11Desmoglein 2(DSG2) Is A Receptor of Human Adenovirus Type 55 Causing Adult Severe Community-Acquired Pneumonia显示文摘Human adenovirus type 55(HAdV-B55) is a re-emergent acute respiratory disease pathogen that causes adult communityacquired pneumonia(CAP). Previous studies have shown that the receptor of HAdV-B14, which genome is highly similar with HAdV-B55, is human Desmoglein 2(DSG2). However, whether the receptor of HAdV-B55 is DSG2 is undetermined because there are three amino acid mutations in the fiber gene between HAdV-B14 and HAdV-B55. Here, firstly we found the 3T3 cells, a mouse embryo fibroblast rodent cell line which does not express human DSG2, were able to be infected by HAdV-B55 after transfected with pcDNA3.1-DSG2, while normal 3T3 cells were still unsusceptible to HAdV-B55 infection. Next, A549 cells with h DSG2 knock-down by siRNA were hard to be infected by HAdV-B3/-B14/-B55, while the control siRNA group was still able to be infected by all these types of HAdVs. Finally, immunofluorescence confocal microscopy indicated visually that Cy3-conjugated HAdV-B55 viruses entered A549 cells by binding to DSG2 protein.Therefore, DSG2 is a major receptor of HAdV-B55 causing adult CAP. 14Reconstruction of linkage maps in the distorted segregation populations of backcross, doubled haploid and recombinant inbred lines显示文摘Non-Mendelian segregation of markers, known as distorted segregation, is a common biological phenomenon. Although segregation distortion affects the estimation of map distances and the results of quantitative trait loci (QTL) mapping, the effects of distorted markers are often ignored in the construction of linkage maps and in QTL mapping. Recently, we have developed a multipoint method via a Hidden Markov chain method to reconstruct linkage maps in an F2 population that corrects for bias of map distances between distorted markers. In this article, the method is extended to cover backcross, doubled haploid and recombinant inbred line (RIL) populations. The results from simulated experiments show that: (1) the degree that two linked segregation distortion loci (SDL) affect the estimation of map distances increases as SDL heritability and interval length between adjacent markers increase, whereas sample size has little effect on the bias; (2) two linked SDL result in the underesti- mation of linkage distances for most cases, overestimation for an additive model with opposite additive effects, and unbiased estimation for an epistatic model with negative additive-by-additive effects; (3) the proposed method can obtain the unbiased estimation of linkage distance. This new method was applied to a rice RIL population with severely distorted segregation to reconstruct the linkage maps, and a bootstrap method was used to obtain 95% confidence intervals of map distances. 18AGE-induced neuronal cell death is enhanced in G2019S LRRK2 mutation with increased RAGE expression显示文摘Background:Leucine-rich repeat kinase 2(LRRK2)mutations represent the most common genetic cause of sporadic and familial Parkinson’s disease(PD).Especially,LRRK2 G2019S missense mutation has been identified as the most prevalent genetic cause in the late-onset PD.Advanced glycation end products(AGEs)are produced in high amounts in diabetes and diverse aging-related disorders,such as cardiovascular disease,renal disease,and neurological disease.AGEs trigger intracellular signaling pathway associated with oxidative stress and inflammation as well as cell death.RAGE,receptor of AGEs,is activated by interaction with AGEs and mediates AGE-induced cytotoxicity.Whether AGE and RAGE are involved in the pathogenesis of mutant LRRK2 is unknown.Methods:Using cell lines transfected with mutant LRRK2 as well as primary neuronal cultures derived from LRRK2 wild-type(WT)and G2019S transgenic mice,we compared the impact of AGE treatment on the survival of control and mutant cells by immunostaining.We also examined the levels of RAGE proteins in the brains of transgenic mice and PD patients by western blots.Results:We show that LRRK2 G2019S mutant-expressing neurons were more sensitive to AGE-induced cell death compared to controls.Furthermore,we found that the levels of RAGE proteins were upregulated in LRRK2 G2019S mutant cells.Conclusions:These data suggest that enhanced AGE-RAGE interaction contributes to LRRK2 G2019S mutation-mediated progressive neuronal loss in PD.Hyun Jin Cho Chengsong Xie Huaibin Cai 2018Translational Neurodegeneration2018,7,1:2
19Transformation relationship among different magnetic minerals within loess-paleosol sediments of the Chinese Loess Plateau显示文摘The dominant magnetic minerals and carriers of magnetic signals within the Chinese Loess Plateau are magnetite, maghemite, hematite, and goethite. In this study, we investigated the provenance and evo- lution of magnetic minerals during loess pedogenesis, using X-ray diffraction (XRD) and optical and electron microscopy, including field emission scanning electron microscopy (FESEM) and high- resolution transmission electron microscopy (HRTEM). Our results reveal that single- and multiphase mineral assemblages among magnetic minerals in the loess-paleosol sequence have been formed. Partial oxidation of coarse eolian magnetite has occurred in the desert source area and the oxidation degree is enhanced after deposition of the dust upon the Chinese Loess Plateau. This mode of origin resulted in a microtexture consisting of an inner magnetite core surrounded by a hematite rim, and strongly affected the magnetic characteristics of the loess. Goethite coexists with hematite in the loess and paleosol, and nanometer-scale hematite is formed upon goethite rims via dehydration. 20Optimal Designs for Genomic Selection in Hybrid Crops显示文摘Improved capacity of genomics and biotechnology has greatly enhanced genetic studies in different areas. Genomic selection exploits the geno type-to-phe no type relationship at the whole-ge nome level and is being implemented in many crops? Here we show that design-thinking and data-mining techniques can be leveraged to optimize genomic prediction of hybrid performance? We phenotyped a set of 276 maize hybrids generated by crossing founder inbreds of nested association mapping populations for flowering time, ear height, and grain yield? With 10 296 310 SNPs available from the parental inbreds, we explored the pat? terns of genomic relati on ships and phenotypic variation to establish training samples based on clustering, graphic n etwork analysis, and genetic mating scheme ? Our analysis showed that training set desig ns outperformed random sampling and earlier methods that either minimize the mean of prediction error variance or maximize the mean of generalized coefficient of determination. Additional analyses of 2556 wheat hybrids from an early-stage hybrid breeding system and 1439 rice hybrids from an established hybrid breeding system validated the approaches.
