| 1 | m^6A-dependent biogenesis of circular RNAs in male germ cells显示文摘The majority of circular RNAs(circRNAs)spliced from coding genes contain open reading frames(ORFs)and thus,have protein coding potential.However,it remains unknown what regulates the biogenesis of these ORF-containing circRNAs,whether they are actually translated into proteins and what functions they play in specific physiological contexts.Here,we report that a large number of circRNAs are synthesized with increasing abundance when late pachytene spermatocytes develop into round and then elongating spermatids during murine spermatogenesis.For a subset of circRNAs,the back splicing appears to occur mostly at m^6A-enriched sites,which are usually located around the start and stop codons in linear mRNAs.Consequently,approximately a half of these male germ cell circRNAs contain large ORFs with m^6A-modified start codons in their junctions,features that have been recently shown to be associated with protein-coding potential.Hundreds of peptides encoded by the junction sequences of these circRNAs were detected using liquid chromatography coupled with mass spectrometry,suggesting that these circRNAs can indeed be translated into proteins in both developing(spermatocytes and spermatids)and mature(spermatozoa)male germ cells.The present study discovered not only a novel role of m^6A in the biogenesis of coding circRNAs,but also a potential mechanism to ensure stable and long-lasting protein production in the absence of linear mRNAs,i.e.,through production of circRNAs containing large ORFs and m^6A-modified start codons in junction sequences. | Chong Tang Yeming Xie Tian Yu Na Liu Zhuqing Wang Rebekah J.Woolsey Yunge Tang Xinzong Zhang Weibing Qin Ying Zhang Ge Song Weiwei Zheng Juan Wang Weitian Chen Xiongyi Wei Zhe Xie Rachel Klukovich Huili Zheng David R.Quilici Wei Yan | 2020 | Cell Research2020,30,3: | 12 |

| 3 | Smoking and Its Relation to Metabolic Status among Chinese Adults: Analysis of a Nationwide Survey显示文摘Objective To update the estimated prevalence rates of smoking and examine major metabolic diseases associated with smoking status in Chinese adults. Methods Using a complex, multistage, probability sampling design, we recruited a nationally representative sample of 98,658 Chinese adults aged ≥18 years in 2010. Information on current, former, never, and passive smoking status was obtained using a standard questionnaire. All estimates were weighted to represent the overall Chinese adult population. Results The estimated proportion of current smokers was 28.3% for Chinese adults aged ≥18 years. The corresponding values of former and passive smokers were 5.1% and 21.4%, respectively. Additionally, former smokers were found to have a less favorable metabolic risk profile among all categories of smoking status in both men and women. The prevalence of metabolic diseases such as diabetes and hypertension also increased with a greater number of smoking pack-years in men. | 4 | Spatial Distribution of Seismic Landslides in the Areas of 1927 Gulang M8.0 Earthquake显示文摘The 1927 Gulang M8.0 earthquake has triggered a huge number of landslides,resulting in massive loss of people’s life and property.However,integrated investigations and results regarding the landslides triggered by this earthquake are rare;such situation hinders the deep understanding of these landslides such as scale,extent,and distribution.With the support of Google Earth software,this study intends to finish the seismic landslides interpretation work in the areas of Gulang earthquake(VIII-XI degree)using the artificial visual interpretation method,and further analyze the spatial distribution and impact factors of these landslides.The results show that the earthquake has triggered at least 936 landslides in the VIII-XI degree zone,with a total landslide area of 58.6 km^2.The dense area of seismic landslides is located in the middle and southern parts of the X intensity circle.Statistical analysis shows that seismic landslides is mainly controlled by factors such as elevation,slope gradient,slope direction,strata,seismic intensity,faults and rivers.The elevation of 2000-2800 m is the high-incidence interval of the landslide.The landslide density is larger with a higher slope gradient.East and west directions are the dominant sliding directions.The areas with Cretaceous and Quaternary strata are the main areas of the Gulang seismic landslides.The X intensity zone triggered the most landslides.In addition,landslides often occur in regions near rivers and faults.This paper provides a scientific reference for exploring the development regularities of landslides triggered by the 1927 Gulang earthquake and effectively mitigating the landslide disasters of the earthquake. | XU Jixiang XU Chong HE Xiangli WEN Boyu GE Keshui BAI Yuzhu | 2020 | Earthquake Research in China2020,34,1: | 4 |

| 8 | Agricultural Development and Implication for Wetlands Sustainability: A Case from Baoqing County, Northeast China显示文摘Historical thematic maps and remote sensing data were applied to address spatiotemporal dynamics of land use/land cover(LULC)changes and its impact on wetlands sustainability based on eight LULC datasets from 1954 to 2015 in Baoqing County,Northeast China.This study demonstrated that LULC drastically changed in the past six decades due to conversion of wetlands,woodland,and grassland into cropland.The cropland was 578.8 km^2 in 1954,accounting for 5.8% of the area in Baoqing County,and it increased to 54.3% in 2015,which was nearly equivalent to 9.4 times of that in 1954.Cropland increased 4843.6 km^2 from 1954 to 2015 with average increased area of 79.4 km^2/yr.The conversion of wetlands was the main reason for cropland increase(49.7%),and woodland(18%)and grassland(16.3%)conversion were other reasons.Results also revealed that 78% of wetlands were lost during the past six decades,of which 91.2% were converted cropland.Population increasing(population across Baoqing in 2015 was 7.8 times of that in 1949),agricultural technology development was the main reason for cropland increase,institutional and economic policies also played important roles for cropland dynamics,particularly paddy field influenced by market price.Agricultural development has caused severe wetlands degradation both in area and functionality,and still being the major threads for wetlands sustainable development.Several suggestions concerning the future land use policy formulation and wetlands sustainability were proposed.They are adjusting the‘food first’agricultural policy,reinforce management for wetlands nature reserves,creating infrastructure for the rational use of surface and groundwater,harnessing the degraded cultivated land. | FANG Chong WEN Zhidan LI Lin DU Jia LIU Ge WANG Xiaodi SONG Kaishan | 2019 | Chinese Geographical Science2019,29,2: | 1 |

| 11 | Deep brain stimulation of the subthalamic nucleus for essential tremor显示文摘Essential tremor (ET),characterized by a postural and/or kinetic tremor and primarily manifested in the upper extremities,head,and other parts of the body,is one of the most common neurological disorders.1 The traditional target for the neurosurgical treatment of ET,the ventral intermedius nucleus (Vim) of the thalamus,has confirmed that chronic deep brain stimulation (DBS) is an effective,standard,and primary procedure for ET.However,the loss of tremor suppression due to tolerance of chronic Vim stimulation,accompanied by other adverse effects,such as paresthesias,dysarthria,dysequilibrium,hyperhidrosis,and localized pain,has necessitated changes in the stimulation target in some patients.Therefore,an alternative target for ET stimulation is required.Although targeting the subthalamic nucleus (STN) also has complications,such as dyskinesia,dysarthria,paresthesias,diplopia,mood changes,depression,and restless leg syndrome,in view of the evidence that STN could be the target for patients with tremors associated with Parkinson's disease (PD),STN was elected in two ET patients in our study with positive effects.Our research is reported to have explored the possibilities of STN-DBS in the treatment of ET. | MENG Fan-gang KAO C. | 13 | Generation of recombinant rabies virus ERA strain applied to virus tracking in cell infection显示文摘The mechanism of rabies virus (RABV) infection still needs to be further characterized. RABV particle with self-fluorescent is a powerful viral model to visualize the viral infection process in cells. Herein, based on a reverse genetic system of the Evelyn-Rokitnicki-Abelseth (rERA) strain, we generated a recombinant RABV rERA-N/mCherry strain that stably expresses an additional ERA nucleoprotein that fuses with the red fluorescent protein mCherry (N/mCherry). The rERA-N/mCherry strain retained growth property similar to the parent strain rERA in vitro. The N/mCherry expression showed genetic stability during passage into mouse neuroblastoma (NA) cells and did not change the virulence of the vector. The rERA-N/mCherry strain was then utilized as a visual viral model to study the RABV-cell binding and internalization. We directly observed the red self-fluorescence of rERA-N/mCherry particles binding to the cell surface, and further co-localizing with clathrin in the early stage of infection in NA cells by fluorescence microscopy. Our results showed that the rERA-N/mCherry strain uses clathrin-dependent endocytosis to enter cells, which is consistent with the well-known mechanism of RABV invasion. | 17 | Mechanoresponsive MiR-138-5p Targets MACF1 to Inhibit Bone Formation显示文摘Mechanical stimuli play an essential role in maintaining bone remodeling and skeletal integrity.Meanwhile,bone can respond to the changes of mechanical condition to adjust its mass and architecture.Clinical studies discover that bedridden patients showed osteoporotic T-scores and low bone mineral density,and long-term immobilized patients presented reduced markers of bone formation.However,as bone formation mediated by osteoblast differentiation is a complex process,the underlying molecular mechanism of mechanical stimuli regulating bone formation is still unclear.Recent evidences show that microRNAs(miRNAs)are involved in mechanical stimuli regulating bone formation or osteoblast differentiation.Nevertheless,no direct evidence identifies mechanoresponsive miRNA in both human and animal bones,and clarifies its mechanoresponsive role under different mechanical conditions(e.g.mechanical unloading,reloading,loading).In the current study,we screened for differentially expressed miRNAs in bone specimens of bedridden patients with fractures,then identified that the expression of miR-138-5p,but not the other miRNAs,altered withbedridden time and was negatively correlated with the expression of the bone formation marker genes Alp(alkaline phosphatase).Moreover,miR-138-5p was up-regulated with reduced bone formation during unloading and down-regulated with increased bone formation during reloading in hind4imb unloaded mice.In addition,miR-138-5p was verified to be responsive to different mechanical unloading condition and cyclic mechanical stretch condition in primary osteogenic cells,respectively.Further in vitro data suggested that mechanoresponsive miR-138-5p directly targeted microtubule actin crosslinking factor 1(MACF1)to inhibit osteoblast differentiation.In vivo,we constructed an osteoblastic miR-138-5p transgenic mice model(TG138)with the Runx2promoter,and found that overexpression miR-138-5p supressed bone formation.Moreover,osteoblast-targeted inhibition of miR-138-5p sensitized bone anabolic response to mechanical loading in TG138 mice.Predominantly,the osteoblast-targeted inhibition of miR-138-5p could counteract bone formation reduction induced by hind limb unloading.Taken together,the mechanoresponsive miR-138-5p inhibited bone anabolic response for developing a novel bone anabolic sensitization strategy. | Zhihao Chen Zhao Fan Liang Chao Lifang Hu Chen Lei Zhang Yan Yin Chong Dijie Li Tian Ye Wuxia Qiu Kewen Zhang Chaofei Yang Xiaona Li Li Yu Weiyi Chen Zhang Ge Qian Airong | 2019 | 医用生物力学2019,34,A01: | 1 |
