3Sorption and Dissipation of Tetracyclines in Soils and Compost显示文摘Two agricultural soils were collected from Dahu and Pinchen counties and swine manure compost (SMC) from Ping- tung County in Taiwan,China to investigate the sorption and dissipation of three tetracyclines (TCs),i.e.,oxytetracycline (OTC),tetracycline (TC) and chlortetracycline (CTC),in compost,soils and soil/compost mixtures with different organic carbon (OC) contents.There were seven treatments in total.TCs were most strongly adsorbed to SMC in all treatments due to the high OC content.When SMC was present in the soils,the sorption of TCs was significantly enhanced,which might be attributed to the increased OC content and CEC.The adsorption of TCs showed non-linear adsorption isotherms and fitted well to the Freundlich model.After 49 d of incubation at 25 ? C in soils and soil/compost mixtures in the dark,TCs elapsed in all substrates,with the time required for 50% degradation (DT 50 ) between 20 and 41 d,and the time for 90% degradation (DT 90 ) between 68 and 137 d.Soil amended with compost enhanced the stability of TCs and reduced their mobility.The dissipation of TCs in a soil environment was slow,indicating that these compounds might be persistent in soil.LI Ling-Ling HUANG Li-Dong CHUNG Ren-Shih FOK Ka-Hang ZHANG Yong-Song 2010Pedosphere2010,20,6:14
4ACSL4 is a predictive biomarker of sorafenib sensitivity in hepatocellular carcinoma显示文摘Sorafenib is the first-line treatment of advanced hepatocellular carcinoma(HCC).However,there is a lack of validated biomarkers to predict sorafenib sensitivity.In this study we investigated the role of ACSL4,a positive-activating enzyme of ferroptosis,in sorafenib-induced cell death and HCC patient outcome.We showed that ACSL4 protein expression was negatively associated with IC50 values of sorafenib in a panel of HCC cell lines(R=−0.952,P<0.001).Knockdown of ACSL4 expression by specific siRNA/sgRNA significantly attenuated sorafenib-induced lipid peroxidation and ferroptosis in Huh7 cells,and also rescued sorafenib-induced inhibition of xenograft tumor growth in vivo.We selected 29 HCC patients with surgery as primary treatment and sorafenib as postoperative adjunct therapy from a hospital-based cohort.A high proportion(66.7%)of HCC patients who had complete or partial responses to sorafenib treatment(according to the revised RECIST guideline)had higher ACSL4 expression in the pretreated HCC tissues,compared with those who had stable or progressed tumor growth(23.5%,P=0.029).Since ACSL4 expression was independent of sorafenib treatment,it could serve as a useful predictive biomarker.Taken together,this study demonstrates that ACSL4 is essential for sorafenib-induced ferroptosis and useful for predicting sorafenib sensitivity in HCC.This study may have important translational impacts in precise treatment of HCC.Ji Feng Pei-zhi Lu Guang-zhi Zhu Shing Chung Hooi Yong Wu Xiao-wei Huang Hui-qi Dai Pan-hong Chen Zhong-jie Li Wen-jing Su Chuang-ye Han Xin-ping Ye Tao Peng Jing Zhou Guo-dong Lu 2021Acta Pharmacologica Sinica2021,42,1:11

8L-Carnitine protects against tacrolimus-induced renal injury by attenuating programmed cell death via PI3K/AKT/PTEN signaling显示文摘Reducing immunosuppressant-related complications using conventional drugs is an efficient therapeutic strategy.L-carnitine(LC)has been shown to protect against various types of renal injury.In this study,we investigated the renoprotective effects of LC in a rat model of chronic tacrolimus(TAC)nephropathy.SD rats were injected with TAC(1.5 mg·kg^(−1)·d^(−1),sc)for 4 weeks.Renoprotective effects of LC were assessed in terms of renal function,histopathology,oxidative stress,expression of inflammatory and fibrotic cytokines,programmed cell death(pyroptosis,apoptosis,and autophagy),mitochondrial function,and PI3K/AKT/PTEN signaling.Chronic TAC nephropathy was characterized by severe renal dysfunction and typical histological features of chronic nephropathy.At a molecular level,TAC markedly increased the expression of inflammatory and fibrotic cytokines in the kidney,induced oxidative stress,and led to mitochondrial dysfunction and programmed cell death through activation of PI3K/AKT and inhibition of PTEN.Coadministration of LC(200 mg·kg^(−1)·d^(−1),ip)caused a prominent improvement in renal function and ameliorated histological changes of kidneys in TAC-treated rats.Furthermore,LC exerted anti-inflammatory and antioxidant effects,prevented mitochondrial dysfunction,and modulated the expression of a series of apoptosis-and autophagy-controlling genes to promote cell survival.Human kidney proximal tubular epithelial cells(HK-2 cells)were treated with TAC(50μg/mL)in vitro,which induced production of intracellular reactive oxygen species and expression of an array of genes controlling programmed cell death(pyroptosis,apoptosis,and autophagy)through interfering with PI3K/AKT/PTEN signaling.The harmful responses of HK-2 cells to TAC were significantly attenuated by cotreatment with LC and the PI3K inhibitor LY294002(25μM).In conclusion,LC treatment protects against chronic TAC nephropathy through interfering the PI3K/AKT/PTEN signaling.Hai-lan Zheng Hai-yue Zhang Chun-lian Zhu Hui-ying Li Sheng Cui Jian Jin Shang-guo Piao Yu-ji Jiang Mei-ying Xuan Ji-zhe Jin Ying-shun Jin Jung-pyo Lee Byung-ha Chung Bum-soon Choi Chul-woo Yang Can Li 2021Acta Pharmacologica Sinica2021,42,1:7

10Bifidobacterium infantis attenuates colitis by regulating T cell subset responses显示文摘AIM:to investigate the effect of Bifidobacterium infantis(B.infantis)on the T cell subsets and in attenuating the severity of experimental colitis in mice.METHODS:Normal BALB/c mice were fed different doses of B.infantis for 3 wk,and T cell subsets and related cytokine profiles in mesenteric lymph nodes(MLNs)were detected by flow cytometry and realtime RT-PCR.Colitis was induced by administration of trinitrobenzene sulfonic acid(TNBS)in mice.Before colitis induction,mice were fed high dose B.infantisfor 3 wk.Cytokine profiles in MLNs and histological changes of colonic tissue were examined 6 d after colitis induction.RESULTS:No significant change in cytokine profiles was observed in normal mice fed low dose B.infantis.However,Th1-related cytokines(IL-2,IFN-γ,IL-12p40),Th17-related transcription factor and cytokines(RORγt,IL-21,IL-23),regulatory T cell(Treg)-related transcription factor and cytokines(Foxp3,IL-10)were increased in normal mice fed high dose B.infantis.Furthermore,flow cytometry assay showed B.infantis increased the numbers of CD4+Foxp3+Tregs and Th17cells in MLNs.Colitis was successfully induced by TNBS in mice,characterized by colonic inflammation and aberrant Th1 and Th17 responses.Feeding high dose B.infantis for 3 wk before colitis induction decreased the inflammatory cell infiltration and goblet cell depletion and restored the intestinal epithelium.In addition,B.infantis feeding reduced Th1-related cytokines(T-bet,IL-2 and IFN-γ)and Th17-related cytokines(IL-12p40,RORγt,IL-17A,IL-21 and IL-23),and increased Tregrelated molecules(Foxp3,IL-10 and TGF-β)in colitis mice.CONCLUSION:B.infantis effectively attenuates TNBSinduced colitis by decreasing Th1 and Th17 responses and increasing Foxp3+Treg response in the colonic mucosa.Li Zuo Kai-Tao Yuan Li Yu Qing-Hong Meng Peter Chee-Keung Chung Ding-Hua Yang 2014World Journal of Gastroenterology2014,20,48:5

14The strategic framework of tuberculosis control and prevention in the elderly:a scoping review towards End TB targets显示文摘With the rapid pace of population ageing,tuberculosis(TB)in the elderly increasingly becomes a public health challenge.Despite the increasing burden and high risks for TB in the elderly,targeted strategy has not been well understood and evaluated.We undertook a scoping review to identify current TB strategies,research and policy gaps in the elderly and summarized the results within a strategic framework towards End TB targets.Databases of Embase,MEDLINE,Global health and EBM reviews were searched for original studies,review articles,and policy papers published in English between January 1990 and December 2015.Articles examining TB strategy,program,guideline or intervention in the elderly from public health perspective were included.Nineteen articles met the inclusion criteria.Most of them were qualitative studies,issued in high-and middle-income countries and after 2000.To break the chain of TB transmission and reactivation in the elderly,infection control,interventions of avoiding delay in diagnosis and containment are essential for preventing transmission,especially in elderly institutions and aged immigrants;screening of latent TB infection and preventive therapy had effective impacts on reducing the risk of reactivation and should be used less reluctantly in older people;optimizing early case-finding with a high index of suspicion,systematic screening for prioritized high-risk groups,initial empirical and adequate follow-up treatment with close monitoring and evaluation,as well as enhanced programmatic management are fundamental pillars for active TB elimination.Evaluation of TB epidemiology,risk factors,impacts and cost-effectiveness of interventions,adopting accurate and rapid diagnostic tools,shorter and less toxic preventive therapy,are critical issues for developing strategy in the elderly towards End TB targets.TB control strategies in the elderly were comprehensively mapped in a causal link pathway.The framework and principals identified in this study will help to evaluate and improve current program,develop targeted strategy,as well as raise more discussions on the research priority settings and policy transitions.Given the scarceness of policy and evaluated interventions,as well as the unawareness of shifting TB epidemiology and strategy especially in developing countries,the increasing need of a ready TB program for the elderly warrants further research.Jun Li Pui-Hong Chung Cyrus L.K.Leung Nobuyuki Nishikiori Emily Y.Y.Chan Eng-Kiong Yeoh 2017Infectious Diseases of Poverty2017,6,1:4

16Monosialoganglioside 1 may alleviate neurotoxicity induced by propofol combined with remifentanil in neural stem cells显示文摘Monosialoganglioside 1(GM1) is the main ganglioside subtype and has neuroprotective properties in the central nervous system. In this study, we aimed to determine whether GM1 alleviates neurotoxicity induced by moderate and high concentrations of propofol combined with remifentanil in the immature central nervous system. Hippocampal neural stem cells were isolated from newborn Sprague-Dawley rats and treated with remifentanil(5, 10, 20 ng/m L) and propofol(1.0, 2.5, 5.0 μg/m L), and/or GM1(12.5, 25, 50 μg/m L). GM1 reversed combined propofol and remifentanil-induced decreases in the percentage of 5-bromodeoxyuridine(+) cells and also reversed the increase in apoptotic cell percentage during neural stem cell proliferation and differentiation. However, GM1 with combined propofol and remifentanil did not affect β-tubulin(+) or glial fibrillary acidic protein(+) cell percentage during neural stem cell differentiation.
