| 1 | Effect of vernalization on tuberization and flowering in the Tibetan turnip is associated with changes in the expression of FLC homologues显示文摘The turnip(Brassica rapa var. rapa) is a biennial crop that is planted in late summer/early fall and forms fleshy tubers for food in temperate regions. The harvested tubers then overwinter and are planted again the next spring for flowering and seeds. FLOWERING LOCUS C(FLC) is a MADS-box transcription factor that acts as a major repressor of floral transition by suppressing the flowering promoters FT and SOC1. Here we show that vernalization effectively represses tuber formation and promotes flowering in Tibetan turnip. We functionally characterized four FLC homologues(BrrFLC1,FLC2, FLC3, and FLC5), and found that BrrFLC2 and BrrFLC1 play a major role in repressing flowering in turnip and in transgenic Arabidopsis. In contrast, tuber formation was correlated with BrrFLC1 expression in the hypocotyl and was repressed under cold treatment following the quantitative downregulation of BrrFLC1. Grafting experiments of non-vernalized and vernalized turnips revealed that vernalization independently suppressed tuberization in the tuber or hypocotyl of the rootstock or scion, which occurred in parallel with the reduction in BrrFLC1 activity. | 3 | Absolute calibration of HY-2A and Jason-2 altimeters for sea surface height using GPS buoy in Qinglan, China显示文摘GPS buoy methodology is one of the main calibration methodologies for altimeter sea surface height calibration. This study introduces the results of the Qinglan calibration campaign for the HY-2A and Jason-2 altimeters. It took place in two time slices;one was from August to September 2014, and the other was in July 2015. One GPS buoy and two GPS reference stations were used in this campaign. The GPS data were processed using the real-time kinematic (RTK) technique. The fi nal error budget estimate when measuring the sea surface height (SSH) with a GPS buoy was better than 3.5 cm. Using the GPS buoy, the altimeter bias estimate was about -2.3 cm for the Jason-2 Geophysical Data Record (GDR) Version ‘D' and from -53.5 cm to -75.6 cm for the HY-2A Interim Geophysical Data Record (IGDR). The bias estimates for Jason-2 GDR-D are similar to the estimates from dedicated calibration sites such as the Harvest Platform, the Crete Site and the Bass Strait site. The bias estimates for HY-2A IGDR agree well with the results from the Crete calibration site. | 4 | Effects of protein sources for milk replacers on growth performance and serum biochemical indexes of suckling calves显示文摘This study investigated the effects of protein sources for milk replacers on growth performance and serum biochemical indexes of suckling calves. Fifty Chinese Holstein bull calves with similar BW and age were randomly allocated to 5 groups(1 control and 4 treatments) of 10 calves in each group. Five types of milk replacers were designed to have the same level of energy and protein. The protein source for milk replacers of the control group was full milk protein(MP). The protein source of milk replacers of the 4 treatment groups was composed of MP and one vegetable protein(VP)(30 and 70% of total protein). The4 types of VP were soybean protein concentrate(SP), hydrolyzed wheat protein(WP), peanut protein concentrate(PP), and rice protein isolate(RP). Results of the experiment showed: 1) there was no significant difference on average daily gain(ADG) and feed:gain ratio(F:G) among the MP, SP and RP groups(P > 0.05), whereas the ADG and F:G of the WP and PP groups were significantly lower compared with the MP group(P < 0.05); 2) there was not a significant difference in withers height, body length and heart girth among treatment groups compared with the MP group(P > 0.05). Thereby the 4 VP milk replacers had no adverse effects on body size of calves; 3) all groups showed no significant difference in the serum contents of urea nitrogen, total protein, albumin, globulin, β-hydroxybutyrate, growth hormone, insulin-like growth factor-1, and the ratio of albumin to globulin(A:G)(P> 0.05). | 5 | Molecular cloning and expression analysis of turnip (Brassica rapa var. rapa) sucrose transporter gene family显示文摘In higher plants, sugars(mainly sucrose) are produced by photosynthetically assimilated carbon in mesophyll cells of leaves and translocated to heterotrophic organs to ensure plant growth and development. Sucrose transporters, or sucrose carriers(SUCs), play an important role in the long-distance transportation of sucrose from source organs to sink organs, thereby affecting crop yield and quality.The identification, characterization, and molecular function analysis of sucrose transporter genes have been reported for monocot and dicot plants. However, no relevant study has been reported on sucrose transporter genes in Brassica rapa var. rapa, a cruciferous root crop used mainly as vegetables and fodder.We identified and cloned 12 sucrose transporter genes from turnips, named BrrSUC1.1 to BrrSUC6.2according to the SUC gene sequences of B. rapa pekinensis. We constructed a phylogenetic tree and analyzed conserved motifs for all 12 sucrose transporter genes identified. Real-time quantitative polymerase chain reaction was conducted to understand the expression levels of SUC genes in different tissues and developmental phases of the turnip. | 6 | Near‑Instantaneously Self‑Healing Coating toward Stable and Durable Electromagnetic Interference Shielding显示文摘Durable electromagnetic interference(EMI)shielding is highly desired,as electromagnetic pollution is a great concern for electronics’stable performance and human health.Although a superhydrophobic surface can extend the service lifespan of EMI shielding materials,degradation of its protection capability and insufficient self-healing are troublesome issues due to unavoidable physical/chemical damages under long-term application conditions.Here,we report,for the first time,an instantaneously self-healing approach via microwave heating to achieve durable shielding performance.First,a hydrophobic 1H,1H,2H,2H-perfluorooctyltriethoxysilane(POTS)layer was coated on a polypyrrole(PPy)-modified fabric(PPy@POTS),enabling protection against the invasion of water,salt solution,and corrosive acidic and basic solutions.Moreover,after being damaged,the POTS layer can,for the first time,be instantaneously self-healed via microwave heating for a very short time,i.e.,4 s,benefiting from the intense thermal energy generated by PPy under electromagnetic wave radiation.This self-healing ability is also repeatable even after intentionally severe plasma etching,which highlights the great potential to achieve robust and durable EMI shielding applications.Significantly,this approach can be extended to other EMI shielding materials where heat is a triggering stimulus for healing thin protection layers.We envision that this work could provide insights into fabricating EMI shielding materials with durable performance for portable and wearable devices,as well as for human health care. | Lihua Zou Chuntao Lan Songlin Zhang Xianhong Zheng Zhenzhen Xu Changlong Li Li Yang Fangtao Ruan Swee Ching Tan | 2021 | Nano-Micro Letters2021,13,11: | 3 |
| 7 | Comparative Cyto-molecular Analysis of Repetitive DNA Provides Insights into the Differential Genome Structure and Evolution of Five Cucumis Species显示文摘The genus Cucumis, includes the cucumber(2n = 14), melon(2n = 24), and other wild species, which is a good model for studying genome organization and evolution due to their variation in genome size and basic chromosome number. In this study, five Cucumis species with different geographical origins and basic chromosome numbers(i.e., C. sativus, C. hystrix, C. melo, C. anguria, and C. metuliferus) were used to identify and characterize the repetitive DNA in detail using a phylogenetic method. Comparative cyto-molecular genetic analysis of repetitive DNA was carried out using a graph-based clustering method, construction of Neighbor-Joining tree and fluorescence in situ hybridization(FISH).The results revealed that the five Cucumis species had differences in the repeat content of their genome, as well as in the composition of repetitive DNA and their genomic proportions. Three species from subgenus Melo showed a decreased tendency in both repeat types and genomic proportions, while two species from subgenus Cucumis also showed a decreased tendency in repeat types, but an increased tendency in genomic proportions. Phylogenic analysis of Cucumis Ty1/Copia, Ty3/Gypsy, and 45 S rDNA ITS regions revealed that C. sativus, C. hystrix, and C. melo were closely related species, which C. sativus and C. hystrix were closer, while C. anguria and C. metuliferus were closer to each other and further from the other three species. Differential accumulation and elimination of different repeat types divergently shaped the genomic architecture of these five Cucumis species, contributing to the genome’s evolution and diversification. | 15 | Nuclear PLD1 combined with NPM1 induces gemcitabine resistance through tumorigenic IL7R in pancreatic adenocarcinoma显示文摘Objective:Pancreatic ductal adenocarcinoma(PDAC)is a highly malignant gastrointestinal cancer with a 5-year survival rate of only 9%.Of PDAC patients,15%-20%are eligible for radical surgery.Gemcitabine is an important chemotherapeutic agent for patients with PDAC;however,the efficacy of gemcitabine is limited due to resistance.Therefore,reducing gemcitabine resistance is essential for improving survival of patients with PDAC.Identifying the key target that determines gemcitabine resistance in PDAC and reversing gemcitabine resistance using target inhibitors in combination with gemcitabine are crucial steps in the quest to improve survival prognosis in patients with PDAC.Methods:We constructed a human genome-wide CRISPRa/dCas 9 overexpression library in PDAC cell lines to screen key targets of drug resistance based on sgRNA abundance and enrichment.Then,co-IP,ChIP,ChIP-seq,transcriptome sequencing,and qPCR were used to determine the specific mechanism by which phospholipase D1(PLD1)confers resistance to gemcitabine.Results:PLD1 combines with nucleophosmin 1(NPM1)and triggers NPM1 nuclear translocation,where NPM1 acts as a transcription factor to upregulate interleukin 7 receptor(IL7R)expression.Upon interleukin 7(IL-7)binding,IL7R activates the JAK1/STAT5 signaling pathway to increase the expression of the anti-apoptotic protein,BCL-2,and induce gemcitabine resistance.The PLD1 inhibitor,Vu0155069,targets PLD1 to induce apoptosis in gemcitabine-resistant PDAC cells.Conclusions:PLD1 is an enzyme that has a critical role in PDAC-associated gemcitabine resistance through a non-enzymatic interaction with NPM1,further promoting the downstream JAK1/STAT5/Bcl-2 pathway.Inhibiting any of the participants of this pathway can increase gemcitabine sensitivity. | Danqi Fu Jingrui Yan Zhaoyu Zhang Yang Liu Xiaoqing Ma Jinsheng Ding Shengyu Yang Ran Zhao Antao Chang Chuntao Gao Jing Liu Tiansuo Zhao Xiuchao Wang Chongbiao Huang Song Gao Ying Ma Bo Tang Yukuan Feng Hongwei Wang Jihui Hao | 2023 | Cancer Biology & Medicine2023,20,8: | 0 |

| 19 | GRASSLAND AGRICULTURE IN CHINA--A REVIEW显示文摘Interactions between crops and livestock have been at the core of the evolution of many agricultural systems.In this paper,we review the development and characteristics of mixed crop-livestock systems,with a focus on grassland-based systems,as these cover large areas in China,and face several challenges.Following the transition from the original hunting and foraging systems to a sedentary lifestyle with integrated crop-livestock production systems some 8000 years ago,a range of different mixed systems have developed,depending on rainfall,solar radiation and temperature,culture and markets.We describe 5 main types of integrated systems,(1)livestock and rangeland,(2)livestock and grain production,(3)livestock and crop-grassland rotations,(4)livestock,crops and forest(silvo-pasture),and(5)livestock,crops and fish ponds.Next,two of these mixed systems are described in greater detail,i.e.,the mountain-oasis-desert system and its modifications in arid and semi-arid regions,and the integrated crop-livestock production systems on the Loess Plateau.In general,crop-livestock interactions in integrated systems have significant positive effects on crop production,livestock production,energy use efficiency and economic profitability.We conclude that improved integration of crop-livestock production systems is one of the most important ways for achieving a more sustainable development of animal agriculture in China. | Fujiang HOU Qianmin JIA Shanning LOU Chuntao YANG Jiao NING Lan LI Qingshan FAN | 2021 | Frontiers of Agricultural Science and Engineering2021,8,1: | 0 |
