1Effect of Recql5 deficiency on the intestinal tumor susceptibility of Apc^(min) mice显示文摘AIM:To investigate whether Recql5,a DNA helicase that plays an important role in the maintenance of genome integrity,is a tumor suppressor in the gastrointestinal tract in mice.METHODS:We generated cohorts of both Recql5-proficient and Recql5-deficient Apcmin/+mice and compared the tumor susceptibility in their gastrointestinal tracts.RESULTS:Recql5 deficiency in Apcmin/+mice resulted in a significant increase in the tumor incidence in both the colon(P=0.0162)and the small intestine(P<0.01).These findings have provided the first genetic evidence for a tumor suppression role of Recql5 in the gastrointestinal tract of mice.Importantly,since mouse Recql5 and human RECQL5 are highly conserved,these findings also suggest that RECQL5 may be a tu-mor suppressor for human colon cancer.CONCLUSION:Recql5 has a tumor suppression role in the mouse gastrointestinal tract.Yiduo Hu,Xincheng Lu,Guangbin Luo,Department of Genetics,Case Comprehensive Cancer Center and University Hospitals of Cleveland,Case Western Reserve University,Cleveland,OH 44106,United States Yiduo Hu,Department of Cancer Biology,Dana-Farber Cancer Institute,Boston,MA 02115,United States 2010World Journal of Gastroenterology2010,16,12:6

5Mapping and characterization of two relevance networks from SNP and gene levels显示文摘Variations of gene expression and DNA sequence are genetically associated. The goal of this study was to build genetic networks to map from SNPs to gene expressions and to characterize the two different kinds of networks. We employed mutual information to evaluate the strength of SNP-SNP and gene-gene associations based on SNPs identity by descent (IBD) data and differences of gene expressions. We applied the approach to one dataset of Genetics of Gene Expression in Humans, and discovered that both the SNP relevance network and the gene relevance network approximated the scale-free topology. We also found that 12.09% of SNP-SNP interactions matched 24.49% of gene-gene interactions, which was consistent with that of the previous studies.
