| 1 | Mn-containing bioceramics inhibit osteoclastogenesis and promote osteoporotic bone regeneration via scavenging ROS显示文摘Osteoporosis is caused by an osteoclast activation mechanism.People suffering from osteoporosis are prone to bone defects.Increasing evidence indicates that scavenging reactive oxygen species(ROS)can inhibit receptor activator of nuclear factorκB ligand(RANKL)-induced osteoclastogenesis and suppress ovariectomy-induced osteoporosis.It is critical to develop biomaterials with antioxidant properties to modulate osteoclast activity for treating osteoporotic bone defects.Previous studies have shown that manganese(Mn)can improve bone regeneration,and Mn supplementation may treat osteoporosis.However,the effect of Mn on osteoclasts and the role of Mn in osteoporotic bone defects remain unclear.In present research,a model bioceramic,Mn-containedβ-tricalcium phosphate(Mn-TCP)was prepared by introducing Mn intoβ-TCP.The introduction of Mn intoβ-TCP significantly improved the scavenging of oxygen radicals and nitrogen radicals,demonstrating that Mn-TCP bioceramics might have antioxidant properties.The in vitro and in vivo findings revealed that Mn^(2+)ions released from Mn-TCP bioceramics could distinctly inhibit the formation and function of osteoclasts,promote the differentiation of osteoblasts,and accelerate bone regeneration under osteoporotic conditions in vivo.Mechanistically,Mn-TCP bioceramics inhibited osteoclastogenesis and promoted the regeneration of osteoporotic bone defects by scavenging ROS via Nrf2 activation.These results suggest that Mn-containing bioceramics with osteoconductivity,ROS scavenging and bone resorption inhibition abilities may be an ideal biomaterial for the treatment of osteoporotic bone defect. | Jianmei Li Cuijun Deng Wanyuan Liang Fei Kang Yun Bai Bing Ma Chengtie Wu Shiwu Dong | 2021 | Bioactive Materials2021,6,11: | 7 |

| 7 | Identification of natural allelic variation in TTL1 controlling thermotolerance and grain size by a rice super pan-genome∞显示文摘Continuously increasing global temperatures present great challenges to food security.Grain size,one of the critical components determining grain yield in rice(Oryza sativa L.),is a prime target for genetic breeding.Thus,there is an immediate need for genetic improvement in rice to maintain grain yield under heat stress.However,quantitative trait loci(QTLs)endowing heat stress tolerance and grain size in rice are extremely rare.Here,we identified a novel negative regulator with pleiotropic effects,Thermo‐Tolerance and grain Length 1(TTL1),from the super pan‐genomic and transcriptomic data.Loss‐of‐function mutations in TTL1 enhanced heat tolerance,and caused an increase in grain size by coordinating cell expansion and proliferation.TTL1 was shown to function as a transcriptional regulator and localized to the nucleus and cell membrane.Furthermore,haplotype analysis showed that hapL and hapS of TTL1 were obviously correlated with variations of thermotolerance and grain size in a core collection of cultivars.Genome evolution analysis of available rice germplasms suggested that TTL1 was selected during domestication of the indica and japonica rice subspecies,but still had much breeding potential for increasing grain length and thermotolerance.These findings provide insights into TTL1 as a novel potential target for the development of high‐yield and thermotolerant rice varieties. | Yarong Lin Yiwang Zhu Yuchao Cui Hongge Qian Qiaoling Yuan Rui Chen Yan Lin Jianmin Chen Xishi Zhou Chuanlin Shi Huiying He Taijiao Hu Chenbo Gu Xiaoman Yu Xiying Zhu Yuexing Wang Qian Qian Cuijun Zhang Feng Wang Lianguang Shang | 2023 | Journal of Integrative Plant Biology2023,65,12: | 0 |

| 9 | Large-scale identification of expression quantitative trait loci in Arabidopsis reveals novel candidate regulators of immune responses and other processes显示文摘The extensive phenotypic diversity within natural populations of Arabidopsis is associated with differences in gene expression.Transcript levels can be considered as in-heritable quantitative traits,and used to map expression quantitative trait loci(eQTL)in genome-wide association studies(GWASs).In order to identify putative genetic de-terminants for variations in gene expression,we used pub-licly available genomic and transcript variation data from 665 Arabidopsis accessions and applied the single nucleotide polymorphism-set(Sequence)Kernel Association Test(SKAT)method for the identification of eQTL.Moreover,we used the penalized orthogonal-components regression(POCRE)method to increase the power of statistical tests.Then,gene annotations were used as test units to identify genes that are associated with natural variations in transcript accumulation,which correspond to candidate regulators,some of which may have a broad impact on gene ex-pression.Besides increasing the chances to identify real as-sociations,the analysis using POCRE and SKAT significantly reduced the computational cost required to analyze large datasets.As a proof of concept,we used this approach to identify eQTL that represent novel candidate regulators of immune responses.The versatility of this approach allows its application to any process that is subjected to natural var-iation among Arabidopsis accessions. | Xingang Wang Min Ren Danni Liu Dabao Zhang Cuijun Zhang Zhaobo Lang Alberto PMacho Min Zhang Jian-Kang Zhu | 2020 | Journal of Integrative Plant Biology2020,62,10: | 0 |
