| 1 | Individualized prediction of perineural invasion in colorectal cancer: development and validation of a radiomics prediction model显示文摘Objective: To develop and validate a radiomics prediction model for individualized prediction of perineural invasion(PNI) in colorectal cancer(CRC).Methods: After computed tomography(CT) radiomics features extraction, a radiomics signature was constructed in derivation cohort(346 CRC patients). A prediction model was developed to integrate the radiomics signature and clinical candidate predictors [age, sex, tumor location, and carcinoembryonic antigen(CEA) level]. Apparent prediction performance was assessed. After internal validation, independent temporal validation(separate from the cohort used to build the model) was then conducted in 217 CRC patients. The final model was converted to an easy-to-use nomogram.Results: The developed radiomics nomogram that integrated the radiomics signature and CEA level showed good calibration and discrimination performance [Harrell's concordance index(c-index): 0.817; 95% confidence interval(95% CI): 0.811–0.823]. Application of the nomogram in validation cohort gave a comparable calibration and discrimination(c-index: 0.803; 95% CI: 0.794–0.812).Conclusions: Integrating the radiomics signature and CEA level into a radiomics prediction model enables easy and effective risk assessment of PNI in CRC. | 2 | Evaluating Suitability of Multiple Precipitation Products for the Lancang River Basin显示文摘Global reanalysis precipitation products could provide valuable meteorological information for flow forecasting in poorly gauged areas, helping to overcome a long-standing challenge in the field. But not all data sources are suitable for all regions or perform the same way in hydrological modeling, so it is essential to test the suitability of precipitation products before applying them. In this study, five widely used global high-resolution precipitation products—Asian Precipitation Highly Resolved Observational Data Integration Towards Evaluation of Water Resources(APHRODITE), National Centers for Environmental Prediction Climate Forecast System Reanalysis(NCEP-CFSR), Climate Hazards Group InfraRed Precipitation with Station Data(CHIRPS), China Gauge-based Daily Precipitation Analysis developed by China Meteorological Administration(CMA) and Agricultural Model Intercomparison and Improvement Project based on the NASA Modern-Era Retrospective Analysis for Research and Applications(AgMERRA)—were evaluated using statistical methods and a hydrological approach for their suitability for the Lancang River Basin. The results indicated that APHRODITE, CMA, AgMERRA and CHIRPS were more accurate precipitation indicators than NCEP-CFSR in terms of the multiyear average and seasonal spatial distribution pattern, all of the CHIRPS, Ag MERRA and APHRODITE perform better than CMA and NCEP-CFSR at the small, medium and high precipitation intensities ranges in subbasin11 and sunbabsin46. All five products performed better in subbasin46(a low-altitude region) than in subbasin11(a high-altitude region) on the daily and monthly scales. In addition to NCEP-CFSR, the other four products all presented encouraging potential for streamflow simulation at daily(Yunjinghong) and monthly(Yunjinghong, Jiuzhou and Gajiu) scale. Hydrological simulations forced with APHRODITE were the best of the five for the Yunjinghong station in capturing daily and monthly measured streamflow. | 3 | Application of backflush and micro-flow techniques to the analysis of C_5—C_(13) hydrocarbons in crude oils and its geochemical significance显示文摘An Agilent 6890N GC equipped with both FID and Agilent 5975 MSD analyzer has been employed to analyze C5—C13 hydrocarbon fractions of crude oils. A technical combination of program temperature volatilizer injection, backflush and micro-flow controller afforded a fine separation of C5—C13 compounds on a PONA column with the heavy part of crude oils being cut off before entering the analytical column. Both GC-FID chromatogram and GC-MS mass chromatograms (MID and full scan) could be obtained at the same time. The retension time differences of nC6—nC13 alkanes between GC and TIC were in the range of 0.02—0.58 minutes. Totally 286 peaks have been assigned group compositions. | 4 | The effects of biodegradation on the compositions of aromatic hydrocarbons and maturity indicators in biodegraded oils from Liaohe Basin显示文摘By the aid of GC-MS technique,a series of sequentially biodegraded oils from Liaohe Basin have been analyzed. The results show that the concentrations and relative compositions of various aromatic compounds in the biodegraded crude oils will change with increasing biodegradation degree. The concentrations of alkyl naphthalenes,alkyl phenanthrenes,alkyl dibenzothiophene are decreased,and the concentration of triaromatic steroids will increase with increasing biodegradation degree in biodegraded oils. Those phenomena indicate that various aromatic compounds are more easily biodegraded by bacteria like other kinds of hydrocarbons such as alkanes,but different series of aromatic compounds have a varied ability to resistant to biodegradation. The ratios of dibenzothiophene to phenenthrene(DBTH/P) and methyl dibenzothiophene to methyl phenanthrene(MDBTH/MP) are related to the features of depositional environment for source rocks such as redox and ancient salinity. However,in biodegraded oils,the two ratios increase quickly with the increase of the biodegradation degree,indicating that they have lost their geochemical significance. In this case,they could not be used to evaluate the features of depositional environment. Methyl phenanthrene index,methyl phenanthrene ratio and methyl dibenzoyhiophene ratio are useful aromatic maturity indicators for the crude oils and the source rocks without vitrinite. But for biodegraded oils,those aromatic maturity indicators will be affected by biodegradation and decrease with the increase of the biodegradation degree. | 5 | Detection and significance of higher thiadiamondoids and diamondoidthiols in oil from the Zhongshen 1C well of the Tarim Basin, NW China显示文摘Oil and gas breakthroughs have been achieved in the Zhongshen 1(ZS1) and 1 C(ZS1 C) wells in Cambrian pre-salt from the Tarim Basin in northwest China. However, Middle and Lower Cambrian reservoirs reveal substantial differences in the geochemistry and secondary alteration characteristics between the oils collected from the two wells. High concentrations of thiadiamondoids and diamondoidthiols, including thiatetramantanes, tetramantanethiols, thiapentamantanes, and pentamantanethiols, are detected in the organic sulfur compound fraction of concentrated oil collected from the ZS1 C well, which samples the Lower Cambrian Xiaoerbulake Formation. Higher diamondoids, such as tetramantanes, pentamantanes, hexamantanes, and cyclohexamantane, also occur in the saturate fractions of the concentrated ZS1 C oil. The presence of these compounds is verified by mass spectra analysis and comparison with previous studies. During thermochemical sulfate reduction(TSR), the cage of higher diamondoids is interpreted to open because of sulfur radicals forming open-cage higher diamondoid-like thiols, followed by cyclization that leads to the formation of high thiadiamondoids. Using D_(16)-adamantane as an internal standard, the concentrations of lower diamondoids and thiadiamondoids of non-concentrated Cambrian oil from well ZS1 C are 83874 and8578 μg/g, respectively, which are far higher than Cambrian oil from well ZS1 and most Ordovician oils in the Tarim Basin. | 6 | The effects of biodegradation on biomarker maturity indicators in sequentially biodegraded oils from Liaohe Basin,China显示文摘By aid of gas chromatogram/mass spectrometry(GC-MS) ,the distributions and the compositions of biomarkers in a set of sequentially biodegraded oils from Liaohe Basin,China,have been quantitatively analyzed,and it has been found that during the biodegradation process of crude oils,the molecular maturity parameters such as Ts/Tm,homohopane C31 22S/(22S+22R) and sterane C29 20S/(20S+20R) ratios will be affected to different extent. The results show that except homohopane C31 22S/(22S+22R) ratio,Ts/Tm ratio will decrease with increasing biodegradation,but for C29 20S/(20S+20R) ratio,it will almost remain constant in slightly and moderately biodegraded oils,and then will increase quickly in severely biodegraded oils. The main reason is that there are some differences in the ability of resistant biodegradation for different isomer of biomarkers with different stereo configuration,resulting in the fact that destroying rate by bacteria for those biomarkers with weak ability will be higher than those with strong ability in resistant biodegradation. For example,18α(H) -22,29,30-trisnorhopanes(Ts) will be destroyed more quickly than 17α(H) -22,29,30-trisnorshopanres(Tm) ,and 20R isomer is more quickly than 20S isomer for C29 sterane,resulting in the relative ratios changed with increasing biodegradation. | 15 | Anti-inflammatory and anabolic biphasic scaffold facilitates osteochondral tissue regeneration in osteoarthritic joints显示文摘Osteochondral defects (OCD) are common but difficult to heal due to the low intrinsic repair capacity of cartilage and its complex hierarchical structure. In osteoarthritis (OA), OCD become more challenging to repair as both cartilage and subchondral bone regeneration are further impaired due to the arthritic environment. Numerous biomaterials have been developed and tested in osteochondral defects while ignoring the inflammatory environment. To target this challenging underlying pathophysiology, we designed and fabricated a biphasic porous and degradable scaffold incorporating anti-inflammatory and anabolic molecules by low-temperature rapid prototyping technology, and its effects on promoting osteochondral regeneration were evaluated using our well-established OA-OCD rabbit model. The biphasic porous scaffolds consisted of poly lactic-co-glycolic acid (PLGA) with kartogenin (KGN) for cartilage repair and PLGA and β-calcium phosphate (PLGA/β-TCP) with cinnamaldehyde (CIN) for subchondral bone repair. KGN is a molecule for promoting chondrogenesis and CIN is a phytomolecule for enhancing osteogenesis and alleviating inflammation. The biphasic scaffolds PLGA/KGN-PLGA/β-TCP/CIN (PK/PTC) with bio-mimic structure provided stable mechanical properties and exhibited excellent biocompatibility to support cell adhesion, proliferation, migration, and distribution. Furthermore, KGN and CIN within biphasic scaffolds could be released in a controlled and sustained mode, and the biphasic scaffold degraded slowly in vitro . Evaluating the repair of 16-weeks post-implantation into critically sized OA-OCD rabbit models revealed that the biphasic scaffold could promote subchondral bone and cartilage regeneration, as well as reverse subchondral osteosclerosis caused by inflammation in vivo .
