1UHPLC-MS/MS analysis of cAMP and cGMP in rat plasma as potential biomarkers of Yin-Yang disharmony in traditional Chinese medicine显示文摘Cyclic 3’,5’-adenosine monophosphate(cAMP)and cyclic 3’,5’-guanosine monophosphate(cGMP)are considered as potential biomarkers for Yin-Yang disharmony in traditional Chinese medicine.However,phosphodiesterase-mediated ex vivo degradation of these molecules in biological samples may result in their underestimation.In the present study,a ultra-high performance liquid chromatography-tandem mass spectrometry(UHPLC-MS/MS)method was developed for determination of cAMP and cGMP in rat plasma,with special consideration of their stability ex vivo.Following precipitation of proteins from plasma samples with 0.4 M perchloric acid,the analytes were chromatographed on a Shimadzu Shimpack-XR-ODS II column with 2.5 mM ammonium acetate and methanol in gradient mode.The MS/MS detection was performed using multiple reaction monitoring in the positive electrospray ionization mode.The lower limit of quantification was 0.27 ng/mL for cAMP and 0.37 ng/mL for cGMP.The method was used to determine the plasma cAMP and cGMP levels in normal and Yin deficiency diabetic rats treated with or without Rehmannia glutinosa.The developed method may be useful for evaluating the regulatory effects of Chinese herbal medicine on the levels of cAMP and cGMP in the body.Xin Wang Yue Du Cuiting Wu Ming Xu Youping Liu Xin Di 2021Journal of Pharmaceutical Analysis2021,11,4:3
2Expanding the Coverage of Metabolic Landscape in Cultivated Rice with Integrated Computational Approaches显示文摘Genome-scale metabolomics analysis is increasingly used for pathway and function discovery in the post-genomics era.The great potential offered by developed mass spectrometry(MS)-based technologies has been hindered,since only a small portion of detected metabolites were identifiable so far.To address the critical issue of low identification coverage in metabolomics,we adopted a deep metabolomics analysis strategy by integrating advanced algorithms and expanded reference databases.The experimental reference spectra and in silico reference spectra were adopted to facilitate the structural annotation.To further characterize the structure of metabolites,two approaches were incorporated into our strategy,i.e.,structural motif search combined with neutral loss scanning and metabolite association network.Untargeted metabolomics analysis was performed on 150 rice cultivars using ultra-performance liquid chromatography coupled with quadrupoleOrbitrap MS.Consequently,a total of 1939 out of 4491 metabolite features in the MS/MS spectral tag(MS2T)library were annotated,representing an extension of annotation coverage by an order of magnitude in rice.The differential accumulation patterns of flavonoids between indica and japonica cultivars were revealed,especially O-sulfated flavonoids.A series of closely-related flavonolignans were characterized,adding further evidence for the crucial role of tricin-oligolignols in lignification.Our study provides an important protocol for exploring phytochemical diversity in other plant species.Xuetong Li Hongxia Zhou Ning Xiao Xueting Wu Yuanhong Shan Longxian Chen Cuiting Wang Zixuan Wang Jirong Huang Aihong Li Xuan Li 2022Genomics, Proteomics & Bioinformatics2022,20,4:0
3Catalytic adsorptive desulfurization of mercaptan,sulfide and disulfide using bifunctional Ti-based adsorbent for ultra-clean oil显示文摘Ultra-deep desulfurization of transformer oil is of great demand among power industry.In this work,the effective and deep removal of various types of organosulfurs,including mercaptan,sulfide and disulfide via catalytic adsorptive desulfurization(CADS)using bifunctional Ti-based adsorbent is reported.Compared to adsorptive desulfurization(ADS),dramatically improvement of the organosulfur uptakes were achieved under CADS process.The equilibrium adsorption capacity at 5 μg∙g^(-1) S reached up to 15.7,33.4,11.6 and 11.9 mg∙g^(-1) for propyl mercaptan(n-PM),dimethyl sulfide(DMS),di-t-butyl disulfide(DTBDS)and dibenzyl disulfide(DBDS),which was 262,477,97 and 128 times to that of ADS process,respectively,and was the highest among the reported desulfurization adsorbents.Moreover,it achieved superior breakthrough capacity of 2050,530 and 210 ml F·(g A)^(-1) at the breakthrough S concentration of 1 μg∙g^(-1) of the commercial transformer oil S containing 10,50 and 150 μg∙g^(-1),respectively.The effectiveness of CADS is associated to the transformation of sulfur species to higher polar sulfonic species with the assistance of mild oxidant,which can be readily captured by silanol groups on SiO2 through Hbonding interaction.The excellent recyclability of the adsorbent can be realized through solvent washing or oxidative air treatment.This work provides an effective and economic approach for the elimination of trace amount of mercaptan,sulfide and disulfide from transformer oil.Cuiting Yang Bowen Wu Zewei Liu Guang Miao Qibin Xia Zhong Li Michael J.Janik Guoqing Li Jing Xiao 2022Chinese Journal of Chemical Engineering2022,35,2:0
