1Arsenic trioxide encapsulated liposomes prepared via copper acetate gradient loading method and its antitumor efficiency显示文摘In this study, arsenic trioxide(ATO) was encapsulated in liposomes via copper acetate(Cu(OAc)2) gradients and high entrapment efficiency of over 80% was obtained. The average particle size and the zeta-potential of the liposomes were detected to be 115.1 ± 29.1 nm and-21.97 ± 0.6 m V, respectively. The TEM images showed rod-like precipitates in the inner aqueous phase, which was supposed be due to the formation of insoluble ATO–Cu complex.The in vitro drug release of ATO–Cu liposomes exhibited a sustained release over 72 h, and the release rates decreased with the increase of the p H of release media. Pharmacokinetic and tissue distribution studies of ATO liposomes showed significantly reduced plasma clearance rate, increased AUC0–12h and T1/2, and improved tumor distribution of As compared to iv administration of ATO solution. The anti-tumor effect of ATO loaded liposomes to S180 tumor-bearing mice was significantly improved with a tumor inhibition rate of 61.2%,meanwhile the toxicity of encapsulated ATO was greatly decreased. 2Phyto-phospholipid complexes(phytosomes): A novel strategy to improve the bioavailability of active constituents显示文摘Although active constituents extracted from plants show robust in vitro pharmacological effects, low in vivo absorption greatly limits the widespread application of these compounds. A strategy of using phyto-phospholipid complexes represents a promising approach to increase the oral bioavailability of active constituents, which is consist of ‘‘label-friendly'phospholipids and active constituents. Hydrogen bond interactions between active constituents and phospholipids enable phospholipid complexes as an integral part. This review provides an update on four important issues related to phyto-phospholipid complexes: active constituents, phospholipids, solvents, and stoichiometric ratios. 3The use of bioactive peptides to modify materials for bone tissue repair显示文摘It has been well recognized that the modification of biomaterials with appropriate bioactive peptides could further enhance their functions.Especially,it has been shown that peptide-modified bone repair materials could promote new bone formation more efficiently compared with conventional ones.The purpose of this article is to give a general review of recent studies on bioactive peptide-modified materials for bone tissue repair.Firstly,the main peptides for inducing bone regeneration and commonly used methods to prepare peptide-modified bone repair materials are introduced.Then,current in vitro and in vivo research progress of peptide-modified composites used as potential bone repair materials are reviewed and discussed.Generally speaking,the recent related studies have fully suggested that the modification of bone repair materials with osteogenicrelated peptides provide promising strategies for the development of bioactive materials and substrates for enhanced bone regeneration and the therapy of bone tissue diseases.Furthermore,we have proposed some research trends in the conclusion and perspectives part.Cunyang Wang Yan Liu Yubo Fan Xiaoming Li 2017Regenerative Biomaterials2017,4,3:1
4Effect of covalent-binding modes of osteogenic-related peptides with artificial carriers on their biological activities in vivo显示文摘Covalent binding between bioactive substances and materials in different ways can significantly improve the bone inductivity and biological activity of bone repair materials.However,there is a lack of systematic understanding of how these binding modes affect biological activities of the active substances.In this study,four kinds of functionalized Multi-walled carbon nanotubes(MWCNTs)were prepared,ensuring the same grafting rate of different functional groups.Subsequently,two kinds of osteogenic-related peptides,bone morphogenetic protein-2 mimicking peptides and osteogenic growth mimicking peptides,were covalently bound to functionalized MWCNTs,ensuring the same molar mass of peptides bound to different functionalized MWCNTs in this process.Then the same amount of functionalized MWC-NTs/Peptides composites were introduced into the scaffolds,and through the ectopic osteogenesis model in rats and calvarial defect model in rabbits,ectopic osteogenesis and bone repair ability of the composites were analyzed.Furthermore,the effects of different covalent binding modes on peptide-induced osteogenesis and bone repair were studied.The results showed that the negative influencing trend of different covalent binding modes of osteogenic-related peptides with artificial carriers on their biological activities was in the order as follows:amide binding(carboxyl)>silane coupling>dopamine bind-ing>amide binding(amino),whose mechanism might be mainly that the covalent binding of peptides with different functional groups resulted in different charges.We believe that the results of this study have important guiding significance for the research and development of bone repair materials covalently bound with bioactive substances.Cunyang Wang Suxiang Yu Qi Guo Ke Zhang Yuntao Di Xiaoming Li 2023Journal of Materials Science & Technology2023,,9:0
