| 1 | Genetically modified CD7-targeting allogeneic CAR-T cell therapy with enhanced efficacy for relapsed/refractory CD7-positive hematological malignancies:a phase I clinical study显示文摘Chimeric antigen receptor(CAR)-T cell therapy against T cell malignancies faces major challenges including fratricide between CAR-T cells and product contamination from the blasts.Allogeneic CAR-T cells,generated from healthy donor T cells,can provide ready-to-use,blast-free therapeutic products,but their application could be complicated by graft-versus-host disease(GvHD)and host rejection.Here we developed healthy donor-derived,CD7-targeting CAR-T cells(RD13-01)with genetic modifications to resist fratricide,GvHD and allogeneic rejection,as well as to potentiate antitumor function.A phase I clinical trial(NCT04538599)was conducted with twelve patients recruited(eleven with T cell leukemia/lymphoma,and one with CD7-expressing acute myeloid leukemia).All patients achieved pre-set end points and eleven proceeded to efficacy evaluation.No dose-limiting toxicity,GvHD,immune effector cell-associated neurotoxicity or severe cytokine release syndrome(grade≥3)were observed.28 days post infusion,81.8%of patients(9/11)showed objective responses and the complete response rate was 63.6%(7/11,including the patient with AML).3 of the responding patients were bridged to allogeneic hematopoietic stem cell transplantation.With a median follow-up of 10.5 months,4 patients remained in complete remission.Cytomegalovirus(CMV)and/or Epstein-Barr virus(EBV)reactivation was observed in several patients,and one died from EBV-associated diffuse large B-cell lymphoma(DLBCL).Expansion of CD7-negative normal T cells was detected post infusion.In summary,we present the first report of a Phase I clinical trial using healthy donor-derived CD7-targeting allogeneic CAR-T cells to treat CD7^(+)hematological malignancies.Our results demonstrated the encouraging safety and efficacy profiles of the RD13-01 allogeneic CAR-T cells for CD7^(+)tumors. | Yongxian Hu Yali Zhou Mingming Zhang Houli Zhao Guoqing Wei Wengang Ge Qu Cui Qitian Mu Gong Chen Lu Han Tingting Guo Jiazhen Cui Xiaoyan Jiang Xiujun Zheng Shuhui Yu Xiaolong Li Xingwang Zhang Mingxi Chen Xiuju Li Ming Gao Kang Wang Cheng Zu Hao Zhang Xiaohong He Yanbin Wang Dongrui Wang Jiangtao Ren He Huang | 2022 | Cell Research2022,32,11: | 2 |
| 2 | Crumpled,high-power,and safe wearable Lithium-Ion Battery enabled by nanostructured metallic textiles显示文摘Textile-based flexible Lithium-Ion Batteries(LIBs)show promising mechanical flexibility that is appealing for a wide variety of wearable and flexible electronic applications.The flexibility of flexible LIBs nowadays is still limited.In addition,their power performance is too low to enable high-speed charging,due to the low conductivity of the textiles.Here,we develop highly electrically conductive metallic fabrics,which are fabricated by coating nanostructured Ni or Cu(nano-reliefs)on woven cotton fabrics,as current collectors to enable crumpled,high-power,and safe wearable LIBs.The nanostructured metal coating not only effectively increases the contact area between current collectors and active materials,but also shortens the charge carrier transport paths,so that LIBs constructed on these nanostructured metallic cotton fabrics exhibit a high power density of 439 W/L and superior electrochemical stability under various mechanical deformations including folding,twisting,squeezing,and impacting.This type of nanostructured metallic textile is highly desirable for portable and wearable electronic applications. | Dongrui Wang Jian Chang Qiyao Huang Dongdong Chen Peng Li Yau-Wai Denis Yu Zijian Zheng | 2021 | Fundamental Research2021,1,4: | 1 |

| 4 | Time interval measurement with linear optical sampling at the femtosecond level显示文摘High-precision time interval measurement is a fundamental technique in many advanced applications,including time and distance metrology,particle physics,and ultra-precision machining.However,many of these applications are confined by the imprecise time interval measurement of electrical signals,restricting the performance of the ultimate system to a few picoseconds,which limits ultrahigh precision applications.Here,we demonstrate an optical means for the time interval measurement of electrical signals that can successfully achieve femtosecond(fs)level precision.The setup is established using the optical frequency comb(OFC)based linear optical sampling(LOS)technique to realize timescale-stretched measurement.We achieve a measurement precision of 82 fs for a single LOS scan measurement and 3.05 fs for the 100-times average with post-processing,which is three orders of magnitude higher than the results of older electrical methods.The high-precision time interval measurement of electrical signals can substantially improve precision measurement technologies. | DONGRUI YU ZIYANG CHEN XUAN YANG YUNLONG XU ZIYI JIN PANXUE MA YUFEI ZHANG SONG YU BIN LUO HONG GUO | 2023 | Photonics Research2023,11,12: | 0 |
