1pRB expression in esophageal mucosa of individuals at high risk for squamous cell carcinoma of the esophagus显示文摘AIM: To investigate the pRb expression in a large group of patients with history of chronic exposure to the main risk factors for development of squamous cell carcinoma of the esophagus. METHODS: One hundred and seventy asymptomatic individuals at high risk for esophageal squamous cell carcinoma (consumption of more than 80 g of ethanol and 10 cigarettes/d for at least 10 years) underwent upper gastrointestinal endoscopy with biopsies of the esophageal mucosa. As a control group, specimens of esophageal mucosa obtained from 20 healthy subjects were also studied. Immunohistochemical assessment of the tissues was performed using a monoclonal antibody anti-pRB protein. RESULTS: Absence of the pRB staining, indicating loss of RB function, was observed in 33 (19.4%) of the individuals at risk for esophageal cancer, but in none of the healthy controls (P < 0.02). Loss of pRb expression increased in a stepwise fashion according to the severity of the histological findings (P < 0.005): normal mucosa (11/97 or 11.3%), chronic esophagitis (17/60 or 28.3%), low-grade dysplasia (3/10 or 30%), high-grade dysplasia 1/2 or 50%) and squamous cell carcinoma (1/1 or 100%). CONCLUSION: Our findings suggest that abnormal expression of the pRB protein may be implicated in the process of esophageal carcinogenesis. 4Orthognathic Surgery Combined with Partial Lipectomy of the Buccal Fat Pad: Case Report on Optimization of Esthetic Outcome显示文摘Orthognathic surgery involves correction of functional and often esthetic disharmonies. The purpose of this case report is topresent the case of a 22 years-old female with class III dentofacial deformity that required orthognathic surgery to treat hermalocclusion and buccal fat pad excision for optimizing soft-tissue facial contour. Treatment planning involved a counterclockwiserotation of the maxilomandibular complex with genioplasty based on digital 3D surgical planning using the Dolphin Imaging 11.7 3DPremium software. Prototyping splints allowed surgical procedures (bilateral sagital mandibular osteotomy associated to a Le fort Iosteotomy) that corrected position of the gnathic bones. In the same surgical procedure, the buccal fat pad was partially removed for asharp cheek contour giving a slimmer appearance to her face. After 5 months, the patient was reassessed and reported no complaintsregarding esthetics or function. 12Piwi like RNA-mediated gene silencing 1 gene as a possible major player in gastric cancer显示文摘AIM To establish a permanent piwi like RNA-mediated genesilencing 1(PIWIL1) gene knockout in AGP01 gastric cancer cell line using CRISPR-Cas9 system and analyze phenotypic modifications as well as gene expression alterations.METHODS CRISPR-Cas9 system used was purchased from Dharmacon GE Life Sciences(Lafayette, CO, United States) and permanent knockout was performed according to manufacturer's recommendations. Woundhealing assay was performed to investigate the effect of PIWIL1 knockout on migration capability of cells and Boyden chamber invasion assay was performed to investigate the effect on invasion capability. For the gene expression analysis, a one-color microarray-based gene expression analysis kit(Agilent Technologies, Santa Clara, CA, United States) was used according to the protocol provided by the manufacturer. RESULTS PIWIL1 gene knockout caused a significant decrease in AGP01 migration capacity as well as a significant decrease in cell invasiveness. Moreover, functional analysis based on grouping of all differentially expressed m RNAs identified a total of 35 genes(5 up-regulated and 30 down-regulated) encoding proteins involved in cellular invasion and migration. According to current literature, 9 of these 35 genes(DOCK2, ZNF503, PDE4 D, ABL1, ABL2, LPAR1, SMAD2, WASF3 and DACH1) are possibly related to the mechanisms used by PIWIL1 to promote carcinogenic effects related to migration and invasion, since their functions are consistent with the changes observed(being up-or down-regulated after knockout). 15Multi-parametric MRI of the prostate:Factors predicting extracapsular extension at the time of radical prostatectomy显示文摘Objective:Extracapsular extension(ECE)of prostate cancer is a poor prognostic factor associated with progression,recurrence after treatment,and increased prostate cancer-related mortality.Accurate staging prior to radical prostatectomy is crucial in avoidance of positive margins and when planning nerve-sparing procedures.Multi-parametric magnetic resonance imaging(mpMRI)of the prostate has shown promise in this regard,but is hampered by poor sensitivity.We sought to identify additional clinical variables associated with pathologic ECE and determine our institutional accuracy in the detection of ECE amongst patients who went on to radical prostatectomy.Methods:mpMRI studies performed between the years 2012 and 2014 were cross-referenced with radical prostatectomy specimens.Predictive properties of ECE as well as additional clinical and biochemical variables to identify pathology-proven prostate cancer ECE were analyzed.Results:The prevalence of ECE was 32.4%,and the overall accuracy of mpMRI for ECE was 84.1%.Overall mpMRI sensitivity,specificity,positive predictive value,and negative predictive value for detection of ECE were 58.3%,97.8%,93.3%,and 81.5%,respectively.Specific mpMRI characteristics predictive of pathologic ECE included primary lesion size((20.73±9.09)mm,mean±SD,p<0.001),T2 PIRADS score(p=0.009),overall primary lesion score(p<0.001),overall study suspicion score(p=0.003),and MRI evidence of seminal vesicle invasion(SVI)(p=0.001).Geoffrey S.Gaunay Vinay Patel Paras Shah Daniel Moreira Ardeshir R.Rastinehad Eran Ben-Levi Robert Villani Manish A.Vira 2017Asian Journal of Urology2017,4,1:1
16Role of multi-parametric MRI of the prostate for screening and staging:Experience with over 1500 cases显示文摘Objective:Contemporary prostate cancer(PCa)screening modalities such as prostate specific antigen(PSA)and digital rectal examination(DRE)are limited in their ability to predict the detection of clinically significant disease.Multi-parametric magnetic resonance imaging(mpMRI)of the prostate has been explored as a staging modality for PCa.Less is known regarding its utility as a primary screening modality.We examined our experience with mpMRI as both a screening and staging instrument.Methods:mpMRI studies performed between 2012 and 2014 in patients without PCa were cross-referenced with transrectal ultrasonography(TRUS)biopsy findings.Statistical analyses were performed to determine association of mpMRI findings with overall cancer diagnoses and clinically significant(Gleason score≥7)disease.Subgroup analyses were then performed on patients with a history of prior negative biopsy and those without a history of TRUS biopsy.mpMRI studies were also cross-referenced with RP specimens.Statistical analyses determined predictive ability of extracapsular extension(ECE),seminal vesicle involvement(SVI),and pathologic evidence of clinically significant disease(Gleason score7).Results:Four hundred biopsy naive or prior negative biopsy patients had positive mpMRI studies.Overall sensitivity,specificity,positive and negative predictive values were 94%,37%,58%,and 87%,respectively and 95%,31%,42%,and 93%,respectively for overall cancer detection and Gleason score≥7 disease.In patients with no prior biopsy history,mpMRI sensitivity,specificity,positive and negative predictive values were 94%,36%,65%,and 82%,for all cancers,and 95%,30%,50%,and 89%for Gleason score7 lesions,respectively.In those with prior negative biopsy sensitivity,specificity,positive and negative predictive values were 94%,37%,52%,and 90% for all cancers,and 96%,32%,36%,and 96% for Gleason score7 lesions,respectively.Seventy-four patients underwent radical prostatectomy(RP)after mpMRI.Lesion size on mpMRI correlated with the presence of Gleason score7 cancers(p Z 0.005).mpMRI sensitivity,specificity,positive and negative predictive values were 84%,39%,81%,and 44% respectively,for Gleason7 cancer.For ECE and SVI,sensitivity and specificity were 58% and 98% and 44% and 97%,respectively.Conclusion:mpMRI is an accurate predictor of TRUS biopsy and RP outcomes.mpMRI has significant potential to change PCa management,particularly in the screening population,in whom a significant proportion may avoid TRUS biopsy.Further studies are necessary to determine how mpMRI should be incorporated into the current PCa screening and staging paradigms.Geoffrey Gaunay Vinay Patel Paras Shah Daniel Moreira Simon J.Hall Manish A.Vira Michael Schwartz Jessica Kreshover Eran Ben-Levi Robert Villani Ardeshir Rastinehad Lee Richstone 2017Asian Journal of Urology2017,4,1:1

18Technical Evaluation of Hybrid Clones of Corymbia spp. to Produce Market Pulp显示文摘This study presents hybrid clones of Corymbia spp.developed by Aperam BioEnergia as potential substitutes for Eucalyptus wood in the pulp industry.The biomass of Eucalyptus spp.was compared with that of Corymbia spp.by performing modified kraft pulping and basic density analyses.Comparisons were made by analyzing their respective mean annual increments of cellulose(MAI_(cel))and specific wood consumption(SWC),estimated using a kappa number of(19±1).The results showed that one of the hybrid clones(Corymbia citriodora×Corymbia torelliana,ID 4)had higher basic wood density,higher screened yield,better MAI_(cel#k19),and lower SWC than the other samples,reaching values of 0.608 t/m^(3),54.1%,24.6 ADt/ha/year(i.e.,air dry ton/hectare/year),and 2.74 m^(3)/ADt,respectively.Consequently,clone ID 4 had the highest MAI_(cel)/SWC ratio score(8.98).Given its high forest productivity and low industrial cost compared with other samples,as well as its advantages over Eucalyptus spp.wood,we concluded that clone ID 4 has great potential as an alternative biomass for pulp production.Marcelo Moreira da Costa Ricardo de Carvalho Bittencourt Thales Augusto Pinto Coelho Nogueira Larissa Soares Silva Weslley Henrique Martins da Silva Sebastião Renato Valverde Gleison Augusto Dos Santos Daniel Alexander Fernandes Coelho Claudilene Aparecida Alves Pena 2022Paper And Biomaterials2022,7,3:0
19A novel supplement with yeast b-glucan, prebiotic, minerals and Silybum marianum synergistically modulates metabolic and inflammatory pathways and improves steatosis in obese mice显示文摘Objective: To evaluate the synergic effects of a novel oral supplement formulation, containing prebiotics,yeast b-glucans, minerals and silymarin(Silybum marianum), on lipid and glycidic metabolism, inflammatory and mitochondrial proteins of the liver, in control and high-fat diet-induced obese mice.Methods: After an acclimation period, 32 male C57 BL/6 mice were divided into the following groups:nonfat diet(NFD) vehicle, NFD supplemented, high-fat diet(HFD) vehicle and HFD supplemented. The vehicle and experimental formulation were administered orally by gavage once a day during the last four weeks of the diet(28 consecutive days). We then evaluated energy homeostasis, inflammation, and mitochondrial protein expression in these groups of mice.Results: After four weeks of supplementation, study groups experienced reduced glycemia, dyslipidemia,fat, and hepatic fibrosis levels. Additionally, proliferator-activated receptor-α, AMP-activated protein kinase-1α, peroxisome proliferator-activated receptor γ co-activator-1α, and mitochondrial transcription factor A expression levels were augmented;however, levels of inhibitor of nuclear factor-κB kinase subunit a and p65 nuclear factor-κB expression, and oxidative markers were reduced.
