| 1 | 共聚物在超声介导基因转染中的新功能显示文摘目的探讨超声和共聚物在促进基因转染中是否有协同作用.方法应用3种水溶性不同的共聚物F127,L61和P85.超声参数设定为1 MHz,1~3 W/cm2,脉冲作功周期为20%.选用C2C12,3T3-MDEI,和CHO 3种细胞系为研究对象.质粒DNA为可产生绿色荧光蛋白的GFP.应用荧光显微镜和流式细胞仪(FACS)评价转染率,台盼蓝染色评价细胞的成活率.每一实验结果至少重复3次以上.结果与以前的研究结果相同,超声可介导基因转染,但随着超声能量的增加,细胞的破坏也增加;单独应用3种共聚物未观察到基因转染.但当与超声同时应用,在较低浓度时共聚物可使超声介导的基因转染率提高2~4倍.结论在较低浓度时,共聚物可显著提高超声介导的基因转染作用. | 陈云超 张青萍 LIANG Hai-Dong David O.
