| 20 | 猪繁殖与呼吸综合征病毒通过阻碍STAT1/STAT2核转位抑制Ⅰ型干扰素的信号转导显示文摘Ⅰ型干扰素(IFNs),IFN-α/β诱导抗病毒应答,在天然免疫抗病毒感染中起着重要作用。猪繁殖与呼吸综合征病毒(PRRSV)抑制Ⅰ型干扰素的合成,但其是否抑制IFN的信号转导尚未明确。本研究结果发现PRRSV干扰IFN的信号转导通路。用PRRSV VR2385株感染MARC-145细胞,经IFN-α处理的感染细胞中,干扰素刺激基因ISG15和ISG56的转录、信号转导蛋白及转录激活因子(STAT)2水平均显著低于未经过IFN-α处理过的对照感染细胞。在PRRSV感染的细胞中,IFN诱导的STAT1和STAT2磷酸化及其异源二聚体的形成都未受影响,但大部分的STAT1/STAT2/IRF9的三聚体仍存在于PRRSV感染细胞的细胞质中,表明三聚体的核易位受到了阻碍。PRRSV VR2385感染的细胞中过度表达的NSP1β抑制了ISG15和ISG56的表达,并阻止STAT1的核转位,这些都表明NSP1β可能是抑制IFN信号转导的病毒蛋白。PRRSV感染的猪肺泡巨噬细胞(PAMs)也抑制INF-α刺激的ISG基因和STAT2蛋白的表达。相反,已获批上市的的低致病性疫苗株Ingelvac PRRSV MLA感染PAMs,不需要添加外源的IFN就能活化包括趋化因子和抗病毒细胞因子等IFN诱导基因的表达,且检测不出其对IFN的信号转导产生影响。这些发现都说明PRRSV通过阻碍ISGF3的核转位阻止Ⅰ型干扰素的活化和信号转导通路。 | Deendayal Patel 程晶 | 2010 | 中国畜牧兽医2010，37，10： | 0 |
