1Correlation between in vitro and in vivo immunomodulatory properties of lactic acid bacteria显示文摘AIM: To investigate the correlation between in vitro and in vivo immunomodulation potential of the probiotic strain and its ability to prevent experimental colitis in mice. METHODS: In vitro immunomodulation was assessed by measuring interleukin (IL)-12p70, IL-10, tumor necrosis factor alpha (TNFa) and interferon g (IFNg) release by human peripheral blood mononuclear cells (PBMCs) after 24 h stimulation with 13 live bacterial strains. A murine model of acute TNBS-colitis was next used to evaluate the prophylactic protective capacity of the same set of strains. RESULTS: A strain-specific in vivo protection was observed. The strains displaying an in vitro potential to induce higher levels of the anti-inflammatory cytokine IL-10 and lower levels of the inflammatory cytokine IL-12, offered the best protection in the in vivo colitis model. In contrast, strains leading to a low IL-10/IL-12 cytokine ratio could not significantly attenuate colitis symptoms. CONCLUSION: These results show that we could predict the in vivo protective capacity of the studied lactic acid bacteria (LAB) based on the cytokine profile we established in vitro. 2A high frequency of GBV-C/HGV coinfection in hepatitis C patients in Germany显示文摘AIM To detect infection rate of GBV-C/HGV inhepatitis C patients,to determine the methodsof higher sensitivity and the primers of higherefficiency for GBV-C/HGV RNA detection and tostudy the dominant subtype and mutation ofGBV-C/HGV.METHODS Quantitative RT-PCR for detectionpf HCV RNA concentration in serum samples,RT-nested PCR with two sets of primers fordetection of GBV-C RNA,RT-PCR ELISA with twosets of primers for detection of HGV RNA,nucleotide sequence and putative amino acidsequence analysis.RESULTS The positive rates of GBV-C RNA atthe 5’-NCR and NS3 region in 211 serums amplesfrom the patients with HCV infection were 31.8%and 22.8% respectively.The positive rates ofHGV RNA at the 5’-NCR and NS5 region in thesame samples were 47.9% and 31.8%respectively.The total positive rate of GBV-C/HGV RNA was as high as 55.5%.HCV copynumbers in the patients without GBV-C/ HGVcoinfection were statistically higher than that inthe patients with GBV-C/ HGV coinfection(P<0.01).Frequent mutation of nucleotideresidue was present in the amplificationproducts.Frameshift mutation was found in twosamples with GBV-C NS3 region nucleotidesequences.All nucleotide sequences fromamplification products showed higher homologyto HGV genome than to GBV-C genome even though part of the sequences were amplifiedwith GBV-C primers.CONCLUSION A high frequency of GBV-C/ HGV coinfection existed in the hepatitis C patients. RT-PCR ELISA was more sensitive than RT-nested PCR for detection of GBV-C/ HGV RNA. The primers derived from the 5 -NCR was more efficient than those derived from the NS3 and NS5 regions. A reverse relationship was found to exist between HCV RNA concentration and GBV-C/ HGV infection frequency. HGV was the dominant subtype of the virus in the local area. 3问号钩端螺旋体lipL21基因改建及其表达产物免疫原性变化和定位显示文摘目的:改建问号钩端螺旋体(简称钩体)lipL21基因核苷酸序列以提高表达产量并了解基因改建前后表达产物免疫原性变化,确定外膜脂蛋白LipL21在钩体表面的定位。方法:参照大肠杆菌偏爱密码子设计,合成lipL21基因核苷酸序列并构建其原核表达系统。采用SDS—PAGE和BioRad凝胶图象分析系统检测改建前后lipL21基因表达量变化。采用钩体TR/PatocI抗血清为一抗的Westernblot鉴定改建前后两种目的重组蛋白rLipL2ls的免疫反应性,显微镜凝集试验(MAT)比较改建前后rLipL21s抗血清对不同钩体血清群的交叉免疫凝集效价变化。应用免疫电镜技术对LipL21进行定位。结果:改建前后lipL21基因表达量分别为细菌总蛋白的8.5%和46.5%。两种rLipL21均能与TR/PatocI抗血清发生免疫结合反应,免疫家兔后均能产生特异性抗体。两种rLipL21兔抗血清对我国15群15型钩体参考标准株MAT效价相近,均为1:80~1:320。LipL21位于钩体外膜表面。结论:LipL21是钩体表面抗原。改建后的lipL21基因可明显提高原核表达产量,其产物可保持良好的抗原性和免疫反应性,其抗体具有广泛的交叉免疫凝集活性。罗冬娇 胡野 Dennin R H 严杰 2007浙江大学学报(医学版)2007,36,5:9
4Mutations in precore and core promoter region of HBV in patients with hepatic failure显示文摘Objective: To clarify the association of hepatitis Bvirus mutants in precore and core promoter regions inpatients with hepatic failure and HBeAg state.Methods: Precore and core promoter regions of 25HBV isolates from the patients with hepatic failurewere analyzed by polymerase chain reaction (PCR)direct sequencing approach.Results: Precore G-to-A1896 mutants were identified in16 (64%) of the 25 isolates. The 'hot spot' mutationsat A-to-T1762 and G-to-A1764 were present together in19 (76%) of the 25 isolates, while C-to-T1653 and T-to-C1753 existed in a mutually exclusive manner andmore frequently in hepatic failure with liver cirrhosisgroup than in hepatic failure with chronic hepatitisgroup (100% vs 50%). Both A1896 and T1762-A1764could be found frequently in HBeAg-positive subjects(77.8% and 88.9%), whereas T1653/C1753 was moreprevalent in anti-HBe-positive subjects than inHBeAg-positive subjects (93.8% vs 33.3%).Conclusions: The whole frequency of mutations in pre-core and core promoter gene will become more fre-quent as HBV infection is to be persistent. Mutationto T1653/C1753 may be useful as a marker for hepaticfailure. It requires further study whether the mixedinfection of mutants and wilds will develop and affectthe condition of HBeAg in serum along the progres-sion of liver disease.Reinhard H. 5RT-PCR检测肝炎和肝癌组织中的HCV复制中间体显示文摘应用逆转录聚合酶链反应(RT—PCR)检测11例血清抗HCV阳性的药瘾患者尸检肝组织和10例原发性肝细胞癌(PHC)患者手术切除肝癌组织中HCV基因(正链HCV RNA)和HCV复制中间体(负链HCV RNA)。结果8/11例肝组织和7/10例肝癌组织中正链HCV RNA阳性,其中各有6例负链HCV RNA亦阳性。HCV各基因区的检出率差别较大,5′NC区检出率最高、C区次之、NS区检出率较低。在同例肝(癌)组织中,若负链HCV RNA阳性,则其信号强度与正链基因无明显差别;但未发现肝癌组织中癌细胞分化程度与HCV的检出率有关。上述结果提示HCV感染与肝癌发生有密切关系,HCV的持续复制可能在癌肿形成中起作用。陈智 Dennin H 何南祥 刘克洲 沃健尔 陈明华 1995中华微生物学和免疫学杂志1995,15,2:3

15Dilemma of concepts and strategies for the prevention of spread of HIV in relation to human behavior,law and human rights显示文摘The new prevalence data regarding the estimated global number of human immunodeficiency virus positive (HIV+) cases,i.e.,including people who are either aware or unaware of their HIV infection in 2010,lead many to wonder why the increase in incidence has reached today’s unprecedented level and escalated within such a short time.This,in spite of prevention campaigns in countries affected by HIV/acquired immune deficiency syndrome (AIDS) with their urgent messages aimed at preventing HIV transmission by promoting changes in individual’s behavior.This article analyzes the background of the prevention strategies,in particular their political,social and legal concepts in terms of human rights,and reveals traits of human behavior not considered thus far.A radical reappraisal is necessary,at social and legislative levels,as well as options additional to current concepts.When ethical issues come up,they become blamed for outmoded moralistic positions.However,ignoring the reality has led to dire consequences from prioritizing individual human rights over society’s collective need to prevent the spread of HIV.Reinhard H.
