1Effect of caffeic acid phenethyl ester on proliferation and apoptosis of colorectal cancer cells in vitro显示文摘AIM: To study the effect of caffeic acid phenethyl ester (CAPE)on proliferation, cell cycle, apoptosis and expression of β-catenin in cultured human colorectal cancer (CRC) cell line HCT116.METHODS: HCT116 cells were treated with CAPE at serial concentrations of 80, 40, 20, 10, 5, 2.5 mg/L. The proliferative status of HCT116 cells was measured by using methabenzthiazuron (MTT) assay. Cell cycle was analyzed by using flow cytometry (FCM) with propidium iodide (PI) labeling method. The rate of apoptosis was detected by using FCM with annexin V-FITC and PI double labeling method.β-catenin levels were determined by Western blotting.β-catenin localization in HCT116 was determined by indirect i mmunofluorescence.RESULTS: After HCT116 cells were exposed to CAPE (80,40, 20, 10, 5, and 2.5 mg/L) for 24, 48, 72, 96 h, CAPE displayed a strong growth inhibitory effect in a dose- and time-dependent manner against HCT116 cells. FCM analysis showed that the ratio of G0/G1 phase cells increased, S phase ratio decreased and apoptosis rate increased after HCT116 cells were exposed to CAPE (10, 5, and 2.5 mg/L)for 24 h. CAPE treatment was associated with decreased cytoplasmic β-catenin, nuclear β-catenin and a concurrent increase in β-catenin protein expression at cell-cell junctions.CONCLUSION: CAPE could inhibit HCT116 cell proliferation and induce cell cycle arrest and apoptosis. 2Doping bioactive elements into a collagen scaffold based on synchronous self-assembly/mineralization for bone tissue engineering显示文摘Pure collagen is biocompatible but lacks inherent osteoinductive,osteoimmunomodulatory and antibacterial activities.To obtain collagen with these characteristics,we developed a novel methodology of doping bioactive elements into collagen through the synchronous self-assembly/mineralization(SSM)of collagen.In the SSM model,amorphous mineral nanoparticles(AMN)(amorphous SrCO3,amorphous Ag3PO4,etc.)stabilized by the polyampholyte,carboxymethyl chitosan(CMC),and collagen molecules were the primary components under acidic conditions.As the pH gradually increased,intrafibrillar mineralization occurred via the self-adaptive interaction between the AMNs and the collagen microfibrils,which were self-assembling;the AMNs wrapped around the microfibrils became situated in the gap zones of collagen and finally transformed into crystals.Srdoped collagen scaffolds(Sr-CS)promoted in vitro cell proliferation and osteogenic differentiation of rat bone marrow mesenchymal stromal cells(rBMSCs)and synergistically improved osteogenesis of rBMSCs by altering the macrophage response.Ag-doped collagen scaffolds(Ag-CS)exhibited in vitro antibacterial effects on S.aureus,as well as cell/tissue compatibility.Moreover,Sr-CS implanted into the calvarial defect of a rat resulted in improved bone regeneration.Therefore,the SSM model is a de novo synthetic strategy for doping bioactive elements into collagen,and can be used to fabricate multifunctional collagen scaffolds to meet the clinical challenges of encouraging osteogenesis,boosting the immune response and fighting severe infection in bone defects.Huanhuan Liu Mingli Lin Xue Liu Ye Zhang Yuyu Luo Yanyun Pang Haitao Chen Dongwang Zhu Xue Zhong Shiqing Ma Yanhong Zhao Qiang Yang Xu Zhang 2020Bioactive Materials2020,5,4:6

4Experimental study of changes of TNF-α in rabbits with steroid-induced avascular necrosis of femoral head显示文摘Objective: To explore the effect of tumor necrosis factor-α(TNF-α)on the occurrence of steroid-induced avascular necrosis of femoral head(SANFH). Methods: Twenty-four rabbits were firstly divided into void group (n=12) and model group (n=12) at random. Prednisone Acetate was injected to the model group(0.32 mg·kg-1·d-1). In the 6 th and 8 th week, two rabbits of the two groups were killed respectively to observe whether the model was successful. The level of TNF-α in serum of the residual rabbits of the two groups was examined in Radioimmunoassay method. Results: The level of TNF-α in model group is significantly higher than that in void group(P<0.001) under the premise of the model of SANFH success by histological observation. 6Provenance and paleogeography of Carboniferous-Permian strata in the Bayanhot Basin:Constraints from sedimentary records and detrital zircon geochronology显示文摘The Bayanhot Basin is a superimposed basin that experienced multiple-staged tectonic movements;it is in the eastern Alxa Block,adjacent to the North China Craton(NCC)and the North Qilian Orogenic Belt(NQOB).There are well-developed Paleozoic-Cenozoic strata in this basin,and these provide a crucial window to a greater understanding of the amalgamation process and source-to-sink relationships between the Alxa Block and surrounding tectonic units.However,due to intensive post-depositional modification,and lack of subsurface data,several fundamental issues-including the distribution and evolution of the depositional systems,provenance supplies and source-to-sink relationships during the Carboniferous-Permian remain unclear and thus hinder hydrocarbon exploration and limit the geological understanding of this basin.Employing integrated outcrop surveys,new drilling data,and detrital zircon dating,this study examines the paleogeographic distribution and evolution,and provenance characteristics of the Carboniferous-Permian strata in the Bayanhot Basin.Our results show that the Bayanhot Basin experienced a long-term depositional evolution process from transgression to retrogression during the Carboniferous-late Permian.The transgression extent could reach the central basin in the early Carboniferous.The maximum regional transgression occurred in the early Permian and might connect the Qilian and North China seas with each other.Subsequently,a gradual regression followed until the end of the Permian.The northwestern NCC appeared as a paleo-uplift area and served as a sediments provenance area for the Alxa Block at that time.The NCC,Bayanwula Mountain,and NQOB jointly served as major provenances during the Carboniferous-Permian.There was no ocean separation,nor was there an orogenic belt between the Alxa Block and the NCC that provided sediments for both sides during the Carboniferous-Permian.The accretion of the Alxa and North China blocks should have been completed before the Carboniferous period.Junfeng Zhao Yijun Zhou KeWang Xuan Li Zhanrong Ma Zhengzhong Ruan DongWang Rui Xue 2021Geoscience Frontiers2021,12,3:2
7Experimental validation of a signal-based approach for structural earthquake damage detection using fractal dimension of time frequency feature显示文摘This article extends a signal-based approach formerly proposed by the authors, which utilizes the fractal dimension of time frequency feature(FDTFF) of displacements, for earthquake damage detection of moment resist frame(MRF), and validates the approach with shaking table tests. The time frequency feature(TFF) of the relative displacement at measured story is defined as the real part of the coefficients of the analytical wavelet transform. The fractal dimension(FD) is to quantify the TFF within the fundamental frequency band using box counting method. It is verified that the FDTFFs at all stories of the linear MRF are identical with the help of static condensation method and modal superposition principle, while the FDTFFs at the stories with localized nonlinearities due to damage will be different from those at the stories without nonlinearities using the reverse-path methodology. By comparing the FDTFFs of displacements at measured stories in a structure, the damage-induced nonlinearity of the structure under strong ground motion can be detected and localized. 13Effect of defects and remediation measures on the internal forces caused by a local thickness reduction in the tunnel lining显示文摘This study presents a case history of a tunnel lining with an insufficient thickness.The effect of the defects resulting from the insuf-ficient thickness of the secondary lining and their locations on the internal forces are discussed based on numerical analysis.The results show that defects increase the number of inflection points in the defect region.The increased maximum bending moment is located on the air side of the secondary lining around the defect region.The defects near the crown of the secondary lining have the most significant influence on the internal forces of the primary and secondary linings,while the defects near the sidewall of the secondary lining have a limited influence on the internal forces of the lining.A remediation measure consisting of the installation of a tertiary lining is proposed and its efficiency is analyzed.The tertiary lining significantly reduces the bending moment of the secondary lining and the axial force of the primary lining.The effect of the thickness and location of the tertiary lining on the tunnel performance is discussed.Ping Lu Dongwang Qiao Chunxiao Wu Shun Wang Xiaopei He Wenjun Zhang Haizuo Zhou 2022Underground Space2022,7,1:1

15Study on the 3-D Velocity Structure around the Focal Region of the Anqing Earthquake in January 2011 and Discussion on the Causative Fault显示文摘The Anqing MS4.8 earthquake occurred on January 19, 2011, with the epicenter lying in the foreland deformation belt along the Yangtze River of the lower Yangtze block. After the earthquake, the field work team surveyed and collected building damage data, calculating and obtaining more accurate intensity distributions. The focal mechanism of the main shock was calculated tentatively using digital seismic wave data from provincial digital seismic networks using the FOCMEC program and the first motions of P, SV and SH waves with their amplitude ratios.
