| 2 | Metformin inhibits pancreatic cancer metastasis caused by SMAD4 deficiency and consequent HNF4G upregulation显示文摘Pancreatic ductal adenocarcinoma(PDAC)has poor prognosis due to limited therapeutic options.This study examines the roles of genome-wide association study identified PDAC-associated genes as therapeutic targets.We have identified HNF4G gene whose silencing most effectively repressed PDAC cell invasiveness.HNF4G overexpression is induced by the deficiency of transcriptional factor and tumor suppressor SMAD4.Increased HNF4G are correlated with SMAD4 deficiency in PDAC tumor samples and associated with metastasis and poor survival time in xenograft animal model and in patients with PDAC(log-rank P=0.036;HR=1.60,95%CI=1.03–2.47).We have found that Metformin suppresses HNF4G activity via AMPK-mediated phosphorylation-coupled ubiquitination degradation and inhibits in vitro invasion and in vivo metastasis of PDAC cells with SMAD4 deficiency.Furthermore,Metformin treatment significantly improve clinical outcomes and survival in patients with SMAD4-deficient PDAC(log-rank P=0.022;HR=0.31,95%CI=0.14–0.68)but not in patients with SMAD4-normal PDAC.Pathway analysis shows that HNF4G may act in PDAC through the cell-cell junction pathway.These results indicate that SMAD4 deficiency-induced overexpression of HNF4G plays a critical oncogenic role in PDAC progression and metastasis but may form a druggable target for Metformin treatment. | Chengcheng Wang Taiping Zhang Quan Liao Menghua Dai Junchao Guo Xinyu Yang Wen Tan Dongxin Lin Chen Wu Yupei Zhao | 2021 | Protein & Cell2021,12,2: | 5 |
| 3 | Evolution of permafrost in Northeast Chinasince the Late Pleistocene显示文摘In Northeast China, permafrost advanced and retreated several times under the influences of fluctuating paleo-climates and paleo-environments since the Late Pleistocene. During the last 60 years, many new data were obtained and studies were conducted on the evolution of permafrost in Northeast China, but so far no systematic summary and review have been made. Based on sedimentary sequences, remains of past permafrost, paleo-flora and-fauna records, and dating data, permafrost evolution since the Late Pleistocene has been analyzed and reconstructed in this paper. Paleo-temperatures reconstructed from the remains of past permafrost and those from paleo-flora and-fauna are compared, and thus the southern limit of permafrost(SLP) in each climate period is inferred by the relationship of the permafrost distribution and the mean annual air/ground temperatures(MAAT/MAGT). Thus, the evolutionary history of permafrost is here divided into five stages:(1) the Late Pleistocene(Last Glaciation, or LG)(65 to 10–8.5 ka), the Last Glaciation Maximum(LGM, 21–13 ka) in particular, the coldest period in the latest history with a cooling of about 6~10 ?C, characterized by extensive occurrences of glaciation, flourishing Mammathas-Coelodonta Faunal Complex(MCFC), widespread aeolian deposits, and significant sea level lowering, and permafrost greatly expanded southwards almost to the coastal plains(37?N–41?N);(2) the Holocene Megathermal Period(HMP, 8.5–7.0 to 4.0–3.0 ka), 3~5 ?C warmer than today, permafrost retreated to about 52°N;(3) the Late Holocene Cold Period(Neoglaciation)(4.0–3.0 to 1.0–0.5 ka), a cooling of 1~3 ?C, some earlier thawed permafrost was refrozen or attached, and the SLP invaded southwards to 46?N;(4) the Little Ice Age(LIA, 500 to 100–150 a), the latest cold period with significant permafrost expansion; and(5) climate warming since the last century, during which Northeast China has undergone extensive permafrost degradation. The frequent and substantial expansions and retreats of permafrost have greatly impacted cold-region environments in Northeast China. North of the SLP during the HMP, or in the present continuous permafrost zone, the existing permafrost was largely formed during the LG and was later overlapped by the permafrost formed in the Neoglaciation. To the south, it was formed in the Neoglaciation. However, many aspects of permafrost evolution still await further investigations, such as data integration, numerical reconstruction, and merging of Chinese permafrost history with those of bordering regions as well as collaboration with related disciplines. | 5 | Comparison of dimension reduction-based logistic regression models for case-control genome-wide association study:principal components analysis vs.partial least squares显示文摘With recent advances in biotechnology, genome-wide association study(GWAS) has been widely used to identify genetic variants that underlie human complex diseases and traits. In case-control GWAS, typical statistical strategy is traditional logistical regression(LR) based on single-locus analysis. However, such a single-locus analysis leads to the well-known multiplicity problem, with a risk of inflating type I error and reducing power. Dimension reduction-based techniques, such as principal component-based logistic regression(PC-LR), partial least squares-based logistic regression(PLS-LR), have recently gained much attention in the analysis of high dimensional genomic data. However, the perfor?mance of these methods is still not clear, especially in GWAS. We conducted simulations and real data application to compare the type I error and power of PC-LR, PLS-LR and LR applicable to GWAS within a defined single nucleotide polymorphism(SNP) set region. We found that PC-LR and PLS can reasonably control type I error under null hypothesis.On contrast, LR, which is corrected by Bonferroni method, was more conserved in all simulation settings. In particular, we found that PC-LR and PLS-LR had comparable power and they both outperformed LR, especially when the causal SNP was in high linkage disequilibrium with genotyped ones and with a small effective size in simulation. | 10 | Effect and safety of Kanglaite injection combined with chemotherapy in the treatment of advanced colorectal cancer:a systematic review and meta-analysis显示文摘Objective:To evaluate the clinical efficacy and safety of Kanglaite injection combined with chemotherapy and chemotherapy alone in the treatment of advanced colorectal cancer.Methods:PubMed,Cochrane library,CNKI,VIP,Wanfang database were systematically searched by inputting keywords to explore the efficacy of Kanglaite injection combined with chemotherapy in the treatment of advanced colorectal cancer.A random-effects model was selected to evaluate the treatment outcomes.The screening of literature,the extraction of data and the assessment of methodology were independently undertaken by two reviewers.Meta analysis was performed using Revman5.3 software and stata15.0 software.Results:A total of 12 RCTs were included,with 792 cases.Compared with chemotherapy alone,Metaanalysis suggested that Kanglaite injection combined with chemotherapy can improve clinical efficiency(RR=1.45,95%CI:1.21-1.74,P<0.0001)and improve patients'quality of life(RR=1.55,95%CI:1.32-1.82,P<0.00001),improve the patient's immune function(CD3+cells:SWD=1.42,95%CI:1.11-1.73;CD4+/CD8+cell ratio:SWD=0.95,95%CI:0.66-1.25;NK cell activity:SWD=3.24,95%CI:2.81-3.66,P<0.00001);Leukopenia rate(RR=0.63,95%CI:0.54-0.74,P<0.0001),incidence of gastrointestinal adverse reactions(RR=0.56,95%CI:0.48-0.66,P<0.00001),incidence of peripheral neurotoxicity(RR=0.79,95%CI:0.65-0.96,P=0.02)were lower than the chemotherapy alone group,the difference was statistically significant.In improving the hand-foot syndrome and oral mucositis after chemotherapy,there was some difference between the Kanglaite injection combined with chemotherapy group and the chemotherapy alone group,but the difference was not significant.Puber bias detection and sensitivity analysis were performed with clinically effective relative risk(RR)as indicators.The results suggested that the publication bias was not obvious.The results of this study are stable.Conclusion:Compared with chemotherapy alone group in the treatment of advanced colorectal cancer,Kanglaite injection combined with chemotherapy can improve the effective rate,quality of life,immune function of patients and reduce the incidence of leukopenia after chemotherapy,gastrointestinal adverse reactions and peripheral neurotoxicity. | Jinlin Wu Zhu Yang Fengxi Long Bing Yang Xun Liu Xianman Wei Dongxin Tang | 2019 | Asian Toxicology Tesearch2019,1,1: | 1 |

| 17 | Genetic polymorphisms in genes regulating cell death and prognosis of patients with rectal cancer receiving postoperative chemoradiotherapy显示文摘Objective:The identification of biomarkers for predicting chemoradiotherapy efficacy is essential to optimize personalized treatment.This study determined the effects of genetic variations in genes involved in apoptosis,pyroptosis,and ferroptosis on the prognosis of patients with locally advanced rectal cancer receiving postoperative chemoradiotherapy(CRT).Methods:The Sequenom MassARRAY was used to detect 217 genetic variations in 40 genes from 300 patients with rectal cancer who received postoperative CRT.The associations between genetic variations and overall survival(OS)were evaluated using hazard ratios(HRs)and 95%confidence intervals(CIs)computed using a Cox proportional regression model.Functional experiments were performed to determine the functions of the arachidonate 5-lipoxygenase(ALOX5)gene and the ALOX5 rs702365 variant.Results:We detected 16 genetic polymorphisms in CASP3,CASP7,TRAILR2,GSDME,CASP4,HO-1,ALOX5,GPX4,and NRF2 that were significantly associated with OS in the additive model(P<0.05).There was a substantial cumulative effect of three genetic polymorphisms(CASP4 rs571407,ALOX5 rs2242332,and HO-1 rs17883419)on OS.Genetic variations in the CASP4 and ALOX5 gene haplotypes were associated with a higher OS.We demonstrated,for the first time,that rs702365[G]>[C]represses ALOX5 transcription and corollary experiments suggested that ALOX5 may promote colon cancer cell growth by mediating an inflammatory response.Conclusions:Polymorphisms in genes regulating cell death may play essential roles in the prognosis of patients with rectal cancer who are treated with postoperative CRT and may serve as potential genetic biomarkers for individualized treatment. | Hongxia Chen Luxi Yin Jie Yang Ningxin Ren Jinna Chen Qixuan Lu Ying Huang Yanru Feng Weihu Wang Shulian Wang Yueping Liu Yongwen Song Yexiong Li Jing Jin Wen Tan Dongxin Lin | 2023 | Cancer Biology & Medicine2023,20,4: | 0 |
| 18 | Astragalus injection as an adjuvant treatment for colorectal cancer:a meta-analysis显示文摘Background:The combination of Chinese patent medicine Astragalus injection and Western medicine has achieved a certain clinical effect in colorectal cancer patients.However,due to the uneven basic conditions and research indicators of these clinical trials,it is difficult to comprehensively evaluate the effect of Astragalus injection in the adjuvant treatment of colorectal cancer.This study aimed to systematically evaluate the efficacy and safety of Astragalus injection as an adjuvant treatment for colorectal cancer.Methods:The Cochrane Library,VIP database,Wanfang database,and Chinese Academic Journal Full Text database were searched for potentially eligible articles from inception to December 15,2018.Randomized controlled trials in which patients were diagnosed with colorectal cancer were included.Patients in the control group received chemotherapy alone or combined with other drugs,or chemotherapy combined with radiotherapy.Patients in the experimental group were treated with Astragalus injection combined with interventions in the control group.Results:A total of 8 articles were included.Compared with Western medicine alone,the Astragalus injection could improve the therapeutic effect(RR=1.18,95%CI(1.01,1.38),P=0.03),improved the quality of life of colorectal cancer patients(SMD=1.18,95%CI(0.86,1.50),P<0.001),inhibited leukopenia(RR=0.55,95%CI(0.42,0.71),P<0.001),reduced neurotoxicity(RR=0.43,95%CI(0.34,0.56),P<0.001),and reduced the incidence of nausea and vomiting(RR=0.67,95%CI(0.55,0.80),P<0.001).Conclusion:Astragalus injection can reduce the toxicity and improve the efficiency of the conventional Western medicine in the treatment of colorectal cancer. | Jia Wang Zhu Yang Fengxi Long Li Luo Jinlin Wu Ting Yu Dongxin Tang | 2020 | Traditional Medicine Research2020,5,1: | 0 |
| 19 | Simulation of Ultrasonic Propagation in Transformers within Thermal Fields and Intelligent Methodology for Hot-spot Temperature Recognition显示文摘Hot-spot temperature of transformer windings is a crucial indicator of internal defects.However,current methods for measuring the hot-spot temperature of transformers do not apply to those already in operation and suffer from data lag.This study introduces a novel inversion method that combines ultrasonic sensing technology,multiphysics simulation,and the K-nearest neighbors algorithm.Leveraging the penetrative ability and temperature sensitivity of ultrasonic sensing,a detailed physical field simulation model was established.This study extensively investigates the characteristics of ultrasonic wave signals inside transformers.The investigation includes different temperature fields,ranging from 40℃ to 110℃ at 10℃ intervals,and various ultrasonic wave emitter conditions.By extracting the key features of the acoustic signals,such as the peak time,propagation time,and peak amplitude,an accurate inversion of the winding hot-spot temperature is successfully achieved.The results demonstrate that this method achieves a high accuracy rate(98.57%)in inverting the internal winding hot-spot temperatures of transformers,offering an efficient and reliable new approach for measuring winding hot-spot temperatures. | Dongxin He Dechao Yang Xinhua Guo Jiefeng Liu Haoxin Guo Qingquan Li Gilbert Teyssedre | 2024 | Chinese Journal of Electrical Engineering2024,10,1: | 0 |
