| 1 | Orthotopic transplantation model of human gastrointestinal cancer and detection of micrometastases显示文摘AIM To establish a relevant animal model ofhuman gastrointestinal cancer, which can beused for repetitive investigations, so as toimprove our understanding and management ofcarcinogenesis and cancer metastasis.METHODS Intact tissues of human colorectaland pancreatic cancers were transplanted innude mice. The biological characteristics of theoriginal and the corresponding transplantedtumors were investigated by HE staining, PASstaining and immunostaining. The metastases inthe livers and lungs of nude mice wereinvestigated by immunostaining withbiotinylated mab KL-1 and by RT-PCR using CK20specific primers.RESULTS There were totally 9 of 16 surgicalspecimens growing in nude mice subcutaneouslyand/.or orthotopically (4 of 6 colorectal and 5 of10 pancreatic cancer). Tumor cell content of thespecimens and freezing of tissue specimens areimportant factors influencing the growth oftransplanted tumor. In the group of fresh tumortissues with greater than 50% tumor cellcontent, the success rate of the transplantationwas 100% (3 cases of pancreatic cancer and 3cases of colorectal cancer). The orthotopicallytransplanted tumors resemble the original tumormorphologically and biologically, including TAAexpression such as CEA byimmunohistochemistry, and CEA level in theserum of mice. Ki-67 labeling index and theexpression of TAA especially K-ras, 17-lA andRA-96, are associated with the potential of tumorgrowth in nude mice. Micrometastases in thelungs and livers of tumor bearing mice can bedetected by immunostaining with biotinylatedmab KL-1 and CK20-specific RT-PCR.CONCLUSION An orthotopic transplantationmodel for human colon and pancreatic cancer innude mice has been set up. We have alsoestablished sensitive detection methods withCK-immunohistochemistry and CK20-RT-PCR tostudy xenotransplanted human cancer and itsmetastatic cancer cells in the liver and lung ofnude mice. | 2 | RILL EROSION PROCESS ON STEEP SLOPE LAND OF THE LOESS PLATEAU显示文摘Rill erosion processes and principle of preventing rill erosion of different applied measures are studied by rainfall simulation and field investigation. Research results show that rill erosion processes can be schematically divided into five stages, that is, concentrated flow forming process; little waterfalls in the concentrated flow forming Process; little waterfalls developing into rill head cut process; rill head cuts developing to form discontinuous rill process and discontinuous rills connecting to form continuous rill process. Rill head cuts backward erosion, rill wall collapsing and runoff washing rill bed are the main patterns of rill development. The relevant equation between critical runoff volume of occurring rill erosion and slope degree can be expressed as follows: Q = 76. 10 - 2. 10J. And relevant equation between critical slope length of occurrence Of rill erosion and slope degree is parabolic with a minimum-value. | 4 | SEDIMENT TRANSPORT MODEL OF A WATERSHED显示文摘The natural watershed is simply represented by a multi-order river net system, so the sediment transport process in watershed can be considered as sediment transport in the system. Based on this assumption, a new sediment graph model-Inverse Gaussian distribution instantaneous unit graph model is introduced in this paper. Then, the parameter estimation and other specific computational procedure are discussed. With the verification of 27 flood events measured in Chabagou watershed (in hilly and gullied areas of the Loess Plateau, Northern Shaanxi), the results are satisfactory and the model has a distinguishing feature in flexibility, model structure, and parameter numbers (only two). | 5 | ADVANCES IN MEASURING TECHNIQUES OF BED LOAD显示文摘Bed load is an important component of total load transportation. The accuracy of bed load measurement depends on the measuring equipment and sampling method. In this paper, the present situation of bed load measurement is introduced, the structure features and the sampling performance of bed lead samplers currently used in the world is evaluated. Results of field comparison of HS and TR2 type samplers used in USA, Don type sampler used in former USSR, and Chinese made Y78-1, Y80, AWT160, MB2 samplers have been summarized. From statistical point of view, selection of sampling verticals, and duration and number of repetitions in a vertical in the bed load measurement were studied. The error might be minimized if the sampling verticals were selected on the basis of equal partial bed load discharge.
