1Role of toll like-receptor 2 in inflammatory activity of macrophage infected with a recombinant BCG expressing the C-terminus of merozoite surface protein-1 of Plasmodium falciparum显示文摘Objective: To investigate the role of toll-like receptor 2(TLR2) in inflammatory activity of macrophage infected with the recombinant Mycobacterium bovis bacillus Calmette-Guerin(rBCG). Methods: Mouse macrophage cell line J774 A.1 was infected with Mycobacterium bovis bacillus Calmette-Guerin(BCG) and rBCG cultures for 48 h in the presence or absence of 10 μg/mL of TLR2 inhibitor. Untreated macrophages were used as a negative control while lipopolysaccharide-stimulated macrophages were used as a positive control. The ability of the macrophage to engulf the BCG and rBCG in the absence or presence of TLR2 inhibitor was assessed using a phagocytic assay, while the production of inflammatory cytokines and nitric oxide by the infected macrophages was evaluated using ELISA and Griess reagent method, while the expression of the inducible nitric oxide synthase was determined using Western blot analysis. Results: The results showed that blocking TLR2 function reduced the phagocytic activity, nitric oxide production and proinflammatory cytokine secretion such as TNF-α, IL-1β and IL-12 p40 as well as inducible nitric oxide synthase expression in the infected macrophages. These data showed the importance of TLR2 in the activation of macrophages following BCG and r BCG infections. 2Needs and concerns of patients in isolation care units - learnings from COVID-19: A reflection显示文摘With strict measures in place to contain the spread of coronavirus disease 2019,many have been isolated as suspected or confirmed cases.Being isolated causes much inconvenience for the patients and family.Patients'and next-of-kins’needs and concerns during isolation will be shared together with suggestions for key process improvements.Our hospital’s Senior Patient Experience Managers contact all patients admitted to the isolation wards on a daily basis to provide some form of support.Common issues raised were gathered and strategies to help with their needs and concerns were discussed.Being in isolation is a challenging period for both patients and family.Nonetheless,we can implement measures to mitigate against the adverse effects of isolation.Patient education,effective and efficient means of communication,close monitoring for signs of distress and anxiety,and early intervention could help patients cope better with the whole isolation experience.Nursing management may want to consider implementing the measures shared in the article to manage patient’s stress while not compromising on staff safety.Peijin Esther Monica Fan Fazila Aloweni Shu Hui Lim Shin Yuh Ang Karen Perera Aik Huan Quek Hwee Koon Susan Quek Tracy Carol Ayre 2020World Journal of Clinical Cases2020,8,10:1

4Association of vitamin D and magnesium with insulin sensitivity and their influence on glycemic control显示文摘Insulin resistance increases the risk of developing diabetes,and the degree of resistance influences the glycemic control of patients with diabetes.Numerous researchers have focused on improving insulin sensitivity in order to prevent diabetes-related complications and other chronic diseases.Several studies have also linked vitamin D levels to insulin secretion and resistance,given that both vitamin D and its receptor complex play important roles in regulating pancreaticβ-cells.It has been suggested that vitamin D supplementation improves vitamin D levels,but further research is needed to confirm this as neither insulin function nor glycemic control improves when vitamin D levels increase.Magnesium is a cofactor for many enzymes.Although the role of magnesium in the management of diabetes has long been evaluated,it has not yet been determined whether magnesium supplements improve insulin function.However,several researchers have found that patients with good glycemic control have high magnesium levels.Magnesium is closely related to vitamin D and is necessary for the transport and activation of vitamin D in humans.Combined supplementation with vitamin D and magnesium improves glycemic control in patients with diabetes.Wan Nor Fazila Hafizan Wan Nik Hani Ajrina Zulkeflee Siti Nadirah Ab Rahim Tuan Salwani Tuan Ismail 2023World Journal of Diabetes2023,14,1:0
5Salvia ceratophylla L. from South of Jordan: new insights on chemical composition and biological activities显示文摘In Jordan,Salvia ceratophylla L.is traditionally used in the treatment of cancer,microbial infections,and urinary disorders.This study aimed:(1)to chemically characterize S.ceratophylla essential oil(EO)from South Jordan,by gas chromatography(GC)and gas chromatography-mass spectrometry(GC-MS);and(2)to evaluate in vitro the cytotoxic,anti-inflammatory,and antiprotozoal activities of the EO,it’s predominant components,and the hexane(A),ethyl acetate(B),methanol(C)and crude-methanol extracts(D).The analysis revealed that the EO has 71 compounds,with linalool(54.8%)as main constitu-ent.Only the hexane extract(A)showed some cytotoxic activity against SK-MEL,KB,BT-549,SK-OV-3,LLC-PK1 and VERO cells lines with IC50 between 60 and>100μg/mL.The EO inhibited NO production(IC5090μg/mL)and NF-κB activity(IC5038μg/mL).The extracts A,B,and D inhibited NO production and NF-κB activity with IC50 between 32 and 150μg/mL.Linalool considerably inhibited NO production(IC5018μg/mL).The extracts tested did not exhibit antileish-manial activity.Regarding antitrypanosomal activity,the EO exhibited significant results with IC502.65μg/mL.In conclu-sion,Jordan S.ceratophylla EO represents a rich source of linalool and bears a promising therapeutic potential for further antitrypanosomal drug development.Mohammad Sanad Abu-Darwish Célia Cabral Zulfigar Ali Mei Wang Shabana IKhan Melissa RJacob Surendra KJain Babu LTekwani Fazila Zulfiqar Ikhlas AKhan Hatem Taifour Lígia Salgueiro Thomas Efferth 2020Natural Products and Bioprospecting2020,10,5:0
