| 1 | Characterization and Mapping of a Salt-Sensitive Mutant in Rice (Oryza sativa L.)显示文摘A salt-sensitive mutant designated rice salt sensitive 2 (rss2) was isolated from the M2 generation of the rice cultivar Nipponbare (Oryza sativa L. ssp. japonica) mutagenized with ethyl methanesulfonate (EMS). This mutant exhibited a greater decrease in salt tolerance with a significant increase in Na+ content in its shoots. Genetic analysis indicated that the increase in Na+ in rss2 was controlled by a single recessive gene. Further genome-wide analysis of the linkage map constructed from the F2 population of rss2/Zhaiyeqing 8 (ZYQ8) showed that two quantitative trait loci (QTLs) on chromosomes 1 and 6 were responsible for the Na+ concentration in shoots, which explained 14.5% and 53.3%, respectively, of the phenotypic variance. The locus on chromosome 1, but not that on chromosome 6, was also detected in the F2 population of Nipponbare/ZYQ8, suggesting that the QTL on chromosome 6 was responsible for the salt sensitivity in rss2. By analyzing the recombination events in 220 mutant individuals of an enlarged mapping population of rss2/ZYQ8, the rss2 locus was precisely mapped to an interval of 605.3 kb between insertion/deletion (InDel) markers IM21962 and IM22567. | 3 | microRNA-18a Promotes Cell Migration and Invasion Through Inhibiting Dicer l Expression in Hepatocellular Carcinoma In Vitro显示文摘Objective To investigate the effects of microRNA-18a(miR-18a) on migration and invasion of hepatocellular carcinoma(HCC) cells,and its possible mechanism associated with Dicer l. Methods HepG 2 and HepG 2.2.15 cells were transfected with miR-18 a inhibitor using Lipofectamine. Cell invasion was evaluated by transwell invasion assay,and cell migration was detected by transwell migration and wound-healing assays. Moreover,luciferase reporter assay was used to identify whether Dicer expression was regulated by miR-18 a. Real-time RT-PCR and western blot were performed to analyze Dicer 1 expression. In addition,a functional restoration assay was performed to investigate whether miR-18 a promotes HCC cell migration and invasion by directly targeting Dicer 1. Results miR-18 a inhibitor can suppress the migration and invasion of HCC cells. Furthermore,suppression of Dicer l expression by small interfering RNA essentially abolished the inhibition of cell migration and invasion induced by miR-18 a inhibitor,restorating these activities to levels similar to the parental HCC cells. Interestingly,suppression of miR-18 a in HCC cells resulted in enhanced expression of Dicer l. In addition,the results of a luciferase assay demonstrated targeted regulation of Dicer l by miR-18 a. | 16 | Ultrasonic Characteristics and Severity Assessment of Lung Ultrasound in COVID-19 Pneumonia:A Retrospective,Observational Study显示文摘The clinical application of lung ultrasound(LS)in the assessment of coronavirus disease 2019(COVID-19)pneumonia severity remains limited,Herein,we investigated the role of LUS imaging in CoVID-19pneumonia patients and the relationship between LUS findings and disease severity.This was a retro-spective,observational study at Tongji Hospital,on 48 recruited patients with COVID-19 pneu-monia,including 32 non-critically ill patients and 16critically ill patients.LUS was performed and the respiratory rate oxygenation(ROX)index,disease severity,and confusion,blood urea nitrogen,respira-tory rate,blood pressure,and age(CURB-65)score were recorded on days 0-7,8-14,and 15-21 after symptomonset.Lung images were divided into 12 regions,and the luS score(0-36 points)was calcu-lated.hestcomputed tomography(CT)scores(0-20 points)were also recorded on days O-7.Coelations between the LUS score,ROX index,and CURB-65 scores were examined.LUS detected COVID-19 pneumonia in 38patients.LUS signs included B lines(34/38,89.5%),consolidations(6/38,15.8%),and pleural effusions(2/38,5.3%).Most cases showed more than one lesion(32/38,84.2%)and involved both lungs(28/38,73.7%).Compared with non-critically ill patients,the LUS scores of critically ill patients were higher(12(10-18)vs 2(0-5),p<0.001).The LUS score showed significant negative cor-relations with the ROX indexon days O-7(r=-0.85,p<0.001),days 8-14(r=-0.71,p<0.001),and days 15-21(r=-0.76,p<0.001)after symptom onset.However,the LUS score was positively correlated with the CT score(r=0.82,p<0.001).The number of patients with LUS-detected lesions decreased from 27 cases(81.8%)to 20 cases(46.5%),and the lus scores significantly decreased from 4(2-10)to 0(0-5),(p<0.001) from days O-7 to 17-21.We conclude that LUS can detect lunglesions in COVID-19 pneumo-nia patients in a portable,real-time,and safe manner.Thus,LUS is helpful in assessing COVID-19 pneu-monia severity in critically ill patients. | Fengxue Zhu Xiujuan Zhao Tianbing Wang Zhenzhou Wang Fuzheng Guo Haiyan Xue Panpan Chang Hansheng Liang Wentao Ni Yaxin Wang Lei Chen Baoguo Jiang | 2021 | Engineering2021，7，3： | 1 |
