2A self-monitoring microneedle patch for light-controlled synergistic treatment of melanoma显示文摘Melanoma is the most aggressive and malignant form of skin cancer.Current melanoma treatment methods generally suffer from frequent drug administration as well as difficulty in direct monitoring of drug release.Here,a self-monitoring microneedle(MN)-based drug delivery system,which integrates a dissolving MN patch with aggregation-induced emission(AIE)-active PATC microparticles,is designed to achieve light-controlled pulsatile chemo-photothermal synergistic therapy of melanoma.The PATC polymeric particles,termed D/I@PATC,encapsulate both of chemotherapeutic drug doxorubicin(DOX)and the photothermal agent indocyanine green(ICG).Upon light illumination,PATC gradually dissociates into smaller particles,causing the release of encapsulated DOX and subsequent fluorescence intensity change of PATC particles,thereby not only enabling direct observation of the drug release process under light stimuli,but also facilitating verification of drug release by fluorescence recovery after light trigger.Moreover,encapsulation of ICG in PATC particles displays significant improvement of its photothermal stability both in vitro and in vivo.In a tumor-bearing mouse,the application of one D/I@PATC MN patch combining with two cycles of light irradiation showed excellent controllable chemo-photothermal efficacy and exhibited~97%melanoma inhibition rate without inducing any evident systemic toxicity,suggesting a great potential for skin cancer treatment in clinics.Chenyuan Wang Yongnian Zeng Kai-Feng Chen Jiawei Lin Qianqian Yuan Xue Jiang Gaosong Wu Fubing Wang Yong-Guang Jia Wei Li 2023Bioactive Materials2023,,9:0
3Chlorine-substituted chiral 2D perovskite microwires with enhanced lattice rigidity for sensitive circularly polarized light detection显示文摘Chiral two-dimensional(2D) perovskites offer numerous attractive features for optoelectronics owing to their soft, deformable lattices and a high degree of chemical tunability. While tremendous advances have been made in perovskite-based direct circularly polarized light(CPL) detection, the low circular polarization anisotropy factor and sensitivity of those photodetectors arising from large lattice distortion still limit practical applications. Herein, chiral 2D perovskite-based single-crystalline microwire arrays with enhanced circular dichroism(CD) absorption are fabricated with the synergy of the capillary-bridgeconfined assembly method and chlorine-substituted phenethylamine(Cl-MBA). Compared with phenethylamine(MBA)-inserted perovskites, the smaller lattice distortion and increased halogen–halogen interaction within Cl–MBA-inserted perovskites strengthen lattice rigidity and weaken electron–phonon coupling to improve carrier transport and thermal stability, resulting in high-performance CPL photodetectors with an anisotropy factor of 0.25, responsivity exceeding 95.7±9.3 A W^(-1)and detectivity exceeding(3.05±0.30)×10^(13)Jones. 4Cold exposure promotes coronavirus infection by altering the gut microbiota and lipid metabolism to reduce host immunity显示文摘Objective:Cold exposure has been suggested to be advantageous for the spread and infection of the coronavirus,and the gut microbiota influences the severity of the infection by modulating host inflammatory and immune responses.However,it remains unclear whether the promotion of viral infection through cold exposure is linked to the gut microbiota.Methods:In this study,we performed an unbiased analysis of gut microbiota,serum,and lung tissue metabolome changes in cold-exposed and virus-infected mice,alongside the assessment of immune-inflammatory indicators in serum and lung tissue.Results:The results revealed that both cold exposure and viral infection significantly decreased the percentage of peripheral blood lymphocytes(CD4^(+)T cells,CD8^(+)T cells,and B cell)and increased the expression of inflammatory factors(IL-6,IL-1β,TNF-α,and IFN-γ).Meanwhile,cold exposure disrupted the homeostasis of gut microbiota,elevating the abundance of pathogenic bacteria(Staphylococcus)and diminishing the abundance of beneficial bacteria(Alistipes).Notably,in virus-infected mice exposed to a cold environment,the reduction in the abundance of beneficial bacteria Alistipes was more pronounced than in cases of single virus infection and cold exposure.Analysis of altered serum and lung tissue metabolites highlighted glycerophospholipids,fatty acids,and eicosanoids as the most affected metabolites by cold exposure.These metabolites,closely associated with virus infection,exhibited a significant correlation with immune-inflammatory indicators.Conclusion:These findings establish a mechanistic connection between cold exposure and virus infection,suggesting that cold exposure-induced dysregulation of gut microbiota and lipid metabolism diminishes host immunity,promoting virus infection.Gaosong Wu Yuhao Zhang Ningning Zheng Saisai Tian Jingyu Liao Wanqi Le Houkai Li Weidong Zhang 2023Frigid Zone Medicine2023,3,4:0
