1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 4miR-374b-5p suppresses RECK expression and promotes gastric cancer cell invasion and metastasis显示文摘AIM:To profile expression of micro RNAs(mi RNAs)in gastric cancer cells and investigate the effect of mi R-374b-5p on gastric cancer cell invasion and metastasis.METHODS:An mi RNA microarray assay was performed to identify mi RNAs differentially expressed in gastric cancer cell lines(MGC-803 and SGC-7901)compared with a normal gastric epithelial cell line.Upregulation of mi R-374b-5p was newly identified and confirmed via quantitative real-time reverse transcriptionPCR(q RT-PCR).MGC-803 cells were transfected with a synthesized anti-mi R-374b-5p sequence or a control vector using Lipofectamine reagent,or treated with transfection reagent alone or phosphate-buffered saline as controls.Rate of transfection was verified after 48 h by q RT-PCR.Cells were then subjected to transwell migration,wound scratch and cell counting kit-8 assays.A bioinformatic analysis to identify mi R-374b-5p target genes was performed using mi Randa,Pic Tar and Target Scan software.A dual luciferase reporter assay was performed to evaluate the influence of mi R-374b-5p on target gene activation,and q RT-PCR and Western blot were used to evaluate the levels of target m RNA and protein following transfection with mi R-374b-5p antisense oligonucleotides.RESULTS:The microarray profiling revealed downregulation of 14(fold change<0.667;P<0.05)and upregulation of 12(fold change>1.50;P<0.05)mi RNAs in MGC-803 and SGC-7901 cells compared with GES-1 controls.The upregulation of mi R-374b-5p(fold change=1.75 and 1.64 in MGC-803 and SGC-7901,respectively;P<0.05)was confirmed by q RT-PCR.Compared with the control groups,the restoration of mi R-374b-5p expression with anti-mi R-374b-5p significantly suppressed the metastasis,invasion and proliferation of MGC-803 cells.The bioinformatic analysis predicted that the 3’untranslated region(UTR)of reversion-inducing cysteine-rich protein with Kazal motif(RECK)contains three mi R-374b-5p target sequences.RECK was verified as a target gene in a dual luciferase reporter assay showing that activation of RECK 3’UTR-pmir GLO was increased by co-transfection with mi R-374b-5p.Finally,transfection of mi R-374b-5p antisense oligonucleotides increased m RNA and protein levels of RECK in MGC-803cells(P<0.05).CONCLUSION:These findings indicate that upregulation of mi R-374b-5p contributes to gastric cancer cell metastasis and invasion through inhibition of RECK expression.Juan Xie Zhi-Hui Tan Xia Tang Ming-Shu Mo Yan-Ping Liu Run-Liang Gan Yi Li Li Zhang Guo-Qing Li 2014World Journal of Gastroenterology2014,20,46:15

7Commissioning progress of the FAST显示文摘The Five-hundred-meter Aperture Spherical radio Telescope(FAST) was completed with its main structure installed on September 25, 2016, after which it entered the commissioning phase. This paper aims to introduce the commissioning progress of the FAST over the past two years. To improve its operational reliability and ensure effective observation time, FAST has been equipped with a real-time information system for the active reflector system and hierarchical commissioning scheme for the feed support system, which ultimately achieves safe operation of the two systems. For meeting the high-performance indices, a highprecision measurement system was set up based on the effective control methods that were implemented for the active reflector system and feed support system. Since the commissioning of the FAST, a low-frequency ultra-wideband receiver and 19-beam1.05-1.45 GHz receiver have been mainly used. Telescope efficiency, pointing accuracy, and system noise temperature were completely tested and ultimately achieved the acceptance indices of the telescope. The FAST has been in the process of national acceptance preparations and has begun to search for pulsars. 8Global absorption center map of the mafic minerals on the Moon as viewed by CE-1 IIM data显示文摘Determining the global distribution of minerals on the Moon has been an important goal of lunar science. Hyperspectral remote sensing is an important approach to acquiring minerals on the Moon on the global scale. The wavelength of the absorption band center is the key parameter for identifying minerals with reflectance spectra as well as remote sensing data. The global absorption center map of the mafic minerals of the Moon was produced for the first time with the Chang’E-1 IIM data. This map shows the global distribution of mafic minerals such as orthopyroxenes, clinopyroxenes, and olivine and even plagioclase feldspar of the Moon. The validation for some representative areas indicates that the global map is reliable and even more detailed than the results derived from Clementine-data. Moreover, our method is insensitive to the topography and viewing and illumination geometries. The global absorption band center map not only contributes to the lunar science research, but also has other implications to be further studied. 9An efficient methodology for utilization of K-feldspar and phosphogypsum with reduced energy consumption and CO2 emissions显示文摘The issues of reducing CO_2 emissions, sustainably utilizing natural mineral resources, and dealing with industrial waste offer challenges for sustainable development in energy and the environment. We propose an efficient methodology via the co-reaction of K-feldspar and phosphogypsum for the extraction of soluble potassium salts and recovery of SO_2 with reduced CO_2 emission and energy consumption. The results of characterization and reactivity evaluation indicated that the partial melting of K-feldspar and phosphogypsum in the hightemperature co-reaction significantly facilitated the reduction of phosphogypsum to SO_2 and the exchange of K^+(K-feldspar) with Ca^(2+)(CaSO_4 in phosphogypsum). The reaction parameters were systematically investigated with the highest sulfur recovery ratio of ~ 60% and K extraction ratio of ~ 87.7%. 10Research progress in ionic liquids catalyzed isobutane/butene alkylation显示文摘The complicated reaction mechanism and the character of competitive reactions lead to a stringent requirement for the catalyst of C_4 alkylation process. Due to their unique properties, ionic liquids(ILs) are thought to be new potential acid catalysts for C_4 alkylation. An analysis of the regular and modified chloroaluminate ILs, novel Br?nsted ILs and composite ILs used in isobutane/butene alkylation shows that the use of either ILs or ILs coupled with mineral acid as homogeneous catalysts can help to greatly adjust the acid strength. By modifying the structural parameters of the cations and anions of the ILs, the solubility of the reactants could also be adjusted, which in turn displays a positive effect on improving the activity of ILs. Immobilization of ILs is an effective way to modulate the surface adsorption/desorption properties and acid strength distribution of the solid acid catalysts. Such a process has a tremendous potential to reduce the deactivation of catalyst and enhance the activity of the solid acid catalyst. 12Measurement of boiling heat transfer coefficient in liquid nitrogen bath by inverse heat conduction method显示文摘Inverse heat conduction method (IHCM) is one of the most effective approaches to obtaining the boiling heat transfer coefficient from measured results. This paper focuses on its application in cryogenic boiling heat transfer. Experiments were conducted on the heat transfer of a stainless steel block in a liquid nitrogen bath, with the assumption of a 1D conduction condition to realize fast acquisition of the temperature of the test points inside the block. With the inverse-heat conduction theory and the explicit finite difference model, a solving program was developed to calculate the heat flux and the boiling heat transfer coefficient of a stainless steel block in liquid nitrogen bath based on the temperature acquisition data. Considering the oscillating data and some unsmooth transition points in the inverse-heat-conduction calculation result of the heat-transfer coefficient, a two-step data-fitting procedure was proposed to obtain the expression for the boiling heat transfer coefficients. The coefficient was then verified for accuracy by a comparison between the simulation results using this expression and the verifying experimental results of a stainless steel block. 13Tetrandrine citrate eliminates imatinib-resistant chronic myeloid leukemia cells in vitro and in vivo by inhibiting Bcr-Abl/β-catenin axis显示文摘Objective:To evaluate the effects of tetrandrine citrate, a novel tetrandrine salt with high water solubility, on the growth of imatinib (IM)-resistant chronic myeloid leukemia (CML) in vitro and in vivo, and reveal action molecular mechanisms. Methods:Cell viability in vitro was measured using methyl thiazolyl tetrazolium (MTT) assay. CML cell growth in vivo was assessed using a xenograft model in nude mice. Bcr-Abl and β-catenin protein levels were determined using Western blotting. Bcr-Abl messenger RNA (mRNA) was measured by reverse transcription polymerase chain reaction (RT-PCR). Flow cytometry (FCM) was used to determine cell cycle status. Results:Tetrandrine citrate inhibited the growth of IM-resistant K562 cells, primary leukemia cells, and primitive CD34 + leukemia cells, and their inhibition concentration that inhibited 50% of target cells (IC 50 ) ranged from 1.20 to 2.97 μg/ml. In contrast, tetrandrine citrate did not affect normal blood cells under the same conditions, and IC 50 values were about 10.12-13.11 μg/ml. Oral administration of tetrandrine citrate caused complete regression of IM-resistant K562 xeno-grafts in nude mice without overt toxicity. Western blot results revealed that treatment of IM-resistant K562 cells with tetrandrine citrate resulted in a significant decrease of both p210 Bcr-Abl and β-catenin proteins, but IM did not affect the Bcr-Abl protein levels. Proteasome inhibitor, MG132, did not prevent tetrandrine-mediated decrease of the p210 Bcr-Abl protein. RT-PCR results showed that tetrandrine treatment caused a decrease of Bcr-Abl mRNA. FCM analysis indicated that tetrandrine induced gap 1 (G 1 ) arrest in CML cells. Conclusions:Tetrandrine citrate is a novel orally active tetrandrine salt with potent anti-tumor activity against IM-resistant K562 cells and CML cells. 14Comparison of the short-term and long-term outcomes of laparoscopic hysterectomies and of abdominal hysterectomies: a case study of 4,895 patients in the Guangxi Zhuang Autonomous Region, China显示文摘Background: To evaluate the short-term and long-term outcomes after laparoscopic hysterectomy(LH) compared with abdominal hysterectomy(AH) in case of benign gynecological disease. Methods: A multi-center cohort retrospective comparative study of population among 4,895 hysterectomies(3,539 LH vs.1,356 AH) between 2007 and 2013 was involved. Operative time(OT), estimated blood loss(EBL), intra-operative and post-operative complications, passing flatus; days with indwelling catheter, questionnaires covering pelvic floor functions and sexual functions were assessed.Results: The EBL(174.1±157.4 vs. 263.1±183.2 cc, LH and AH groups, respectively), passing flatus(38.7±14.1 vs. 48.1±13.2 hours), days with indwelling catheter(1.5±0.6 vs. 2.2±0.8 days), use of analgesics(6.5% vs. 73.1%), intra-operative complication rate(2.4% vs. 4.1%), post-operative complication rate(2.3% vs. 5.7%), post-operative constipation(12.1% vs. 24.6%), mild and serious stress urinary incontinence(SUI) post-operative(P<0.001; P=0.014), and proportion of Female Sexual Functioning Index(FSFI) total score <26.55 post-operative(P<0.001) of the LH group were significantly less than those of AH group. There wereno significant differences in OT(106.5±34.5 vs. 106.2±40.3 min) between the two groups.Conclusions: LH is a safe and efficient operation for improving patients' long-term quality of life(Qo L), and LH is a cost-effectiveness procedure for treating benign gynecological disease. 16Image Signature Based Mean Square Error for Image Quality Assessment显示文摘Motivated by the importance of Human visual system(HVS) in image processing, we propose a novel Image signature based mean square error(ISMSE)metric for full reference Image quality assessment(IQA).Efficient image signature based describer is used to predict visual saliency map of the reference image. The saliency map is incorporated into luminance difference between the reference and distorted images to obtain image quality score. The effect of luminance difference on visual quality with larger saliency value which is usually corresponding to foreground objects is highlighted. 19The fundamental performance of FAST with 19-beam receiver at L band显示文摘The Five-hundred-meter Aperture Spherical radio Telescope(FAST) has passed national acceptance and finished one pilot cycle of ‘Shared-Risk’ observations. It will start formal operation soon. In this context, this paper describes testing results of key fundamental parameters for FAST, aiming to provide basic support for observation and data reduction of FAST for scientific researchers. The 19-beam receiver covering 1.05–1.45 GHz was utilized for most of these observations. The fluctuation in electronic gain of the system is better than 1% over 3.5 hours, enabling enough stability for observations. Pointing accuracy,aperture efficiency and system temperature are three key parameters for FAST. The measured standard deviation of pointing accuracy is 7.9′′, which satisfies the initial design of FAST. When zenith angle is less than 26.4°, the aperture efficiency and system temperature around 1.4 GHz are ~0.63 and less than 24 K for central beam, respectively. The sensitivity and stability of the 19-beam backend are confirmed to satisfy expectation by spectral HI observations toward NGC 672 and polarization observations toward 3 C 286. 20Numerical Simulation of Austenite Recrystallization in CSP Hot Rolled C-Mn Steel Strip显示文摘An integrated mathematical model is developed to predict the microstructure evolution of C-Mn steel during multipass hot rolling on the CSP production line,and the thermal evolution,the temperature distribution,the deformation,and the austenite recrystallization are simulated.The characteristics of austenite recrystallization of hot rolled C-Mn steel in the CSP process are also discussed.The simulation of the microstructure evolution of C-Mn steel ZJ510L during CSP multipass hot rolling indicates that dynamic recrystallization and metadynamic recrystallization may easily occur in the first few passes,where nonuniform recrystallization and inhomogeneous grain size microstructure may readily occur;during the last few passes,static recrystallization may occur dominantly,and the microstructure will become more homogeneous and partial recrystallization may occur at relatively low temperature.TANG Guang-bo LIU Zheng-dong DONG Han GAN Yong KANG Yong-lin LI Lie-jun MAO Xin-ping 2007Journal of Iron and Steel Research(International)2007,14,4:4
