| 1 | New U-Pb age constraints on the upper Banxi Group and synchrony of the Sturtian glaciation in South China显示文摘The Nanhua basin in South China hosts well-preserved middle-late Neoproterozoic sedimentary and volcanic rocks that are critical for studying the basin evolution, the breakup of the supercontinent Rodinia, the nature and dynamics of the 'snowball' Earth and diversification of metazoans. Establishing a stratigraphic framework is crucial for better understanding the interactions between tectonic, paleoclimatic and biotic events recorded in the Nanhua basin, but existing stratigraphic correlations remain debated, particularly for pre-Ediacaran strata. Here we report new Laser Ablation Inductively Coupled Plasma Mass Spectrometry(LA-ICPMS) U-Pb zircon ages from the middle and topmost Wuqiangxi Formation(the upper stratigraphic unit of the Banxi Group) in Siduping, Hunan Province, South China. Two samples show similar age distribution, with two major peaks at ca. 820 Ma and 780 Ma and one minor peak at ca. 910 Ma, suggesting that the Wuqiangxi sandstone was mainly sourced from Neoproterozoic rocks. Two major age peaks correspond to two phases of magmatic events associated with the rifting of the Nanhua basin, and the minor peak at ca. 910 Ma may correspond to the Shuangxiwu volcanic arc magmatism, which represents pre-collision/amalgamation subduction on the southeastern margin of the Yangtze Block. The youngest zircon group from the topmost Wuqiangxi Formation has a weighted mean age of 714.6±5.2 Ma, which is likely close to the depositional age of the uppermost Banxi Group. This age, along with the ages reported from other sections, constrains that the Banxi Group was deposited between ca. 820 Ma and ca. 715 Ma. The age of 714.6±5.2 Ma from the top of the Wuqiangxi Formation is indistinguishable with the SIMS U-Pb age of 715.9± 2.8 Ma from the upper Gongdong Formation in the Sibao village section of northern Guangxi, South China. It is also, within uncertainties, overlapped with two TIMS U-Pb ages from pre-Sturtian strata in Oman and Canada. | 3 | Clinical,Laboratory and Imaging Features of High Altitude Pulmonary Edema in Tibetan Plateau显示文摘Objective To analyze characteristics of high altitude pulmonary edema(HAPE)in Chinese patients.Methods We performed a retrospective study of 98 patients with HAPE.We reviewed the medical records and summarized the clinical,laboratory and imaging characteristics of these cases,and compared the results on admission with those determined before discharge.Results Forty-eight(49.0%)patients developed HAPE at the altitude of 2800 m to 3000 m.Ninty-five(96.9%)patients were man.Moist rales were audible from the both lungs,and moist rales over the right lung were clearer than those over the left lung in fourteen patients.The white blood cells[(12.83±5.55)versus(8.95±3.23)×109/L,P=0.001)]as well as neutrophil counts[(11.34±3.81)versus(7.49±2.83)×109/L,P=0.001)]were higher,whereas the counts of other subsets of white blood cells were lower on admission than those after recovery(all P<0.05).Serum levels of alkaline phosphatase(115.8±37.6 versus 85.7±32.4 mmol/L,P=0.020),cholinesterase(7226.2±1631.8 versus 6285.3±1693.3 mmol/L,P=0.040),creatinine(85.2±17.1 versus 75.1±12.8 mmol/L,P=0.021),uric acid(401.9±114.2 versus 326.0±154.3 mmol/L,P=0.041),and uric glucose(7.20±1.10 versus 5.51±1.11 mmol/L,P=0.001)were higher,but carbondioxide combining power(CO2CP,26.7±4.4 versus 28.9±4.5 mmol/L,P=0.042)and serous calcium(2.32±0.13 versus 2.41±0.10 mmol/L,P=0.006)were lower on admission.Arterial blood gas results showed hypoxemia and respiratory alkalosis on admission.Conclusions In the present research,men were more susceptible to HAPE than women,and in the process of HAPE,the lesions of the right lung were more serious than those of the left lung.Some indicators of routine blood test and blood biochemistry of HAPE patients changed. | Zongbin Li Hongyan Chen Jiayue Li Gaoyuan Li Chunwei Liu Yundai Chen | 2018 | Chinese Medical Sciences Journal2018,33,3: | 9 |
| 4 | A genetic linkage map of hexaploid naked oat constructed with SSR markers显示文摘Naked oat is a unique health food crop in China. Using 202 F2 individuals derived from a hybrid between the variety 578 and the landrace Sanfensan, we constructed a genetic linkage map consisting of 22 linkage groups covering 2070.50 c M and including 208 simple sequence repeat(SSR) markers. The minimum distance between adjacent markers was0.01 c M and the average was 9.95 c M. Each linkage group contained 2–22 markers. The largest linkage group covered 174.40 c M and the shortest one covered 36.80 c M, with an average of 94.11 c M. Thirty-six markers(17.3%) showing distorted segregation were distributed across linkage groups LG5 to LG22. | 6 | Effects of intestinal lymph on expression of neutrophil adhesion factors and lung injury after trauma-induced shock显示文摘AIM: To study how intestinal lymph after trauma-induced shock (TIS) interferes with expression of neutrophil adhesion factors (CDllb and CD18) and causes lung injury.METHODS: Thirty-two adult healthy Sprague-Dawley rats were randomly divided into four experimental groups. Groups 1 and 2 included rats with TIS caused by hitting the midupper part of both side femoral bones with a 2 500 kg rawiron, and with or without ligation of mesenteric lymph duct. Groups 3 and 4 included rats with sham-TIS and with or without ligation of mesenteric lymph duct. Expression of neutrophil CD18 and CD11b in at 1 and 3 h after a 90-min TIS/sham-TIS was evaluated. These rats were killed at 3 h after TIS/sham-TIS, and lungs were taken immediately. The main lung injury indexes (bhe MPO activity and lung injury score) were measured.RESULTS: The expressions of CD18 and CD11b at 1 and 3 h after a 90-min TIS and the main lung injury indexes were significantly increased compared with those in the sham-TIS groups (P<0.05). Moreover, at 1 and 3 h after TIS, the expressions of CD18 (32.12±1.25 and 33.46±0.98) and CD11b (29.56±1.35 and 30.56±1.85) were significantly decreased in rats with ligation of mesenteric lymph duct, compared with those (52.3±1.12 and 50.21±1.25, and 42.24±1.24 and 42.81±1.12, respectively) in those without the ligation (all P<0.05). The main lung injury indexes in rats with TIS with ligation .of mesenteric lymph duct (0.96±0.12 and 6.54±0.35) were also significantly decreased, compared with those (1.56±0.21 and 9.56±0.23) in rats with TIS without the ligation (both P<0.05). However, there was no significant difference in expressions of CD18 and CD11b and the main lung injury indexes between the two sham-TIS groups.CONCLUSION: Previous ligation of mesenteric lymph ducts prevents or alleviates the up-regulated expression of PMN CD18 and CD11b and the lung injury induced by TIS. | 7 | CRISPR/Cas9:A powerful tool for crop genome editing显示文摘The CRISPR/Cas9 technology is evolved from a type II bacterial immune system and represents a new generation of targeted genome editing technology that can be applied to nearly all organisms. Site-specific modification is achieved by a single guide RNA(usually about 20nucleotides) that is complementary to a target gene or locus and is anchored by a protospaceradjacent motif. Cas9 nuclease then cleaves the targeted DNA to generate double-strand breaks(DSBs), which are subsequently repaired by non-homologous end joining(NHEJ) or homology-directed repair(HDR) mechanisms. NHEJ may introduce indels that cause frame shift mutations and hence the disruption of gene functions. When combined with double or multiplex guide RNA design, NHEJ may also introduce targeted chromosome deletions,whereas HDR can be engineered for target gene correction, gene replacement, and gene knock-in. In this review, we briefly survey the history of the CRISPR/Cas9 system invention and its genome-editing mechanism. | 9 | Knob-associated tandem repeats on mitotic chromosomes in maize, Zea diploperennis and their hybrids显示文摘Knob-associated tandem repeats, 180-bp repeats and TR-1 elements, together with 45S rDNA were located on mitotic chromosomes of Zea diploperennis (DP),maize inbred line F102 and their hybrid. In DP, knobs on the short arm of chromosomes 1 and 4 and on the long arm ofthe chromosomes 4 and 5 are composed predominantly of the 180-bp repeats. In addition, 180-bp repeats existed together with TR-1 elements were also detected on the short arm ofchromosomes 2 and 5 and on the long arm of the chromosomes 2, 6, 7, 8 and 9. In maize inbred line F102, 180-bp repeats were present in chromosomes 7S and one homologue of chromosomes 8L. TR-1 elements appeared on satellite of chromosome 6 and no detectable hybridization site co-located with 180-bp repeats was observed in maize F102.Polymorphism of size, number, and distribution of 180-bp and TR-1 signals were revealed among different chromosomes in these two species and heteromorphism existed between some homologous chromosomes in the same species.Using these excellent landmarks, the interspecific hybrid of maize and DP were identified. | 11 | Synthetic lethal short hairpin RNA screening reveals that ring finger protein 183 confers resistance to trametinib in colorectal cancer cells显示文摘Background: The mitogen-activated extracellular signal-regulated kinase 1/2(MEK1/2) inhibitor trametinib has shown promising therapeutic effects on melanoma, but its efficacy on colorectal cancer(CRC) is limited. Synthetic lethality arises with a combination of two or more separate gene mutations that causes cell death, whereas individual mutations keep cells alive. This study aimed to identify the genes responsible for resistance to trametinib in CRC cells,using a synthetic lethal short hairpin RNA(shRNA) screening approach.Methods: We infected HT29 cells with a pooled lentiviral shRNA library and applied next-generation sequencing to identify shRNAs with reduced abundance after 8-day treatment of 20 nmol/L trametinib. HCT116 and HT29 cells were used in validation studies. Stable ring finger protein 183(RNF183)-overexpressing cell lines were generated by pcDNA4-myc/his-RNF183 transfection. Stable RNF 183-knockdown cell lines were generated by infection of lentiviruses that express RNF183 shRNA, and small interference RNA(siRNA) was used to knock down RNF183 transiently.Quantitative real-time PCR was used to determine the mRNA expression. Western blotting, immunohistochemical analysis, and enzyme-linked immunosorbent assay(ELISA) were used to evaluate the protein abundance. MTT assay,colony formation assay, and subcutaneous xenograft tumor growth model were used to evaluate cell proliferation.Results: In the primary screening, we found that the abundance of RNF183 shRNA was markedly reduced after treatment with trametinib. Trametinib induced the expression of RNF183, which conferred resistance to drug-induced cell growth repression and apoptotic and non-apoptotic cell deaths. Moreover, interleukin-8(IL-8) was a downstream gene of RNF183 and was required for the function of RNF183 in facilitating cell growth. Additionally, elevated RNF183 expression partly reduced the inhibitory effect of trametinib on IL-8 expression. | 12 | Controlling Beam Halo-Chaos by Adaptive Control Exterior Magnetic Field显示文摘In this paper, the parametric adaptive method for controlling the beam halo-chaos in the periodic focusing channels of high-current proton linacs is presented. The study of proton beam halo-chaos based on controlled beam envelope equation and the results of particles-in-cell simulations for macro-particle beam show that the proton beam chaotic envelope as well as the beam rsm radius can be controlled to the beam matched radius using this method.For the Kapchinskij-Vladimirskij (K- V) distribution of initial proton beam, all statistical physical, quantities of the beam halo-chaos are largely reduced. | 14 | Sedimentary Facies, Provenance and Geochronology of the Heshangzhen Group: Implications for the Tectonic Evolution of the Eastern Jiangnan Orogen, South China显示文摘The Neoproterozoic Jiangnan orogen plays an important role in the study of the Precambrian tectonic evolution of South China. The tectonic nature of the Neoproterozoic sedimentary basins is still controversial, due to poor understanding of the sedimentary sequences and the lack of geochronological data. Here, we present sedimentological, provenance and geochronological data from the Heshangzhen Group in the eastern Jiangnan orogen. Sedimentological analysis shows that the Luojiamen Formation was deposited in a submarine fan, and the overlying Hongchicun Formation was deposited in front of a fan delta. The youngest detrital zircons constrain the lower Luojiamen and Hongchicun formations with ages of 827.3 ± 8.4 Ma and 825 ± 12 Ma, respectively. The sandstones of the Luojiamen Formation are characterized by a large number of intermediate to felsic volcanic grains, suggesting a volcanic arc source. In contrast, quartz and sedimentary lithic grains increase in the Hongchicun Formation, showing a new input from a collisional orogenic source. Detrital zircon from six sandstone samples in the Luojiamen and Hongchicun formations yield similar age spectra of 930–820 Ma with a peak at ca. 845–860 Ma, with one main cluster at 930–820 Ma. Detrital zircons of 930–845 Ma show a positive value of εHf(t)(+2.4 to +11, mean +7.6), which is similar to the volcanic arc of the nearby Shuangxiwu Group. There are a minor group of zircons with U-Pb ages ranging from 820 Ma to 845 Ma from the middle part of the Luojiamen Formation and Hongchicun Formation, with εHf(t) values between-20 to +2.4, which are consistent with the characteristics of the Shuangqiaoshan Group. within light of the bidirectional paleocurrents in the Luojiamen Formation, it is speculated that the zircons of 820–845 Ma were recycled from the Shuangqiaoshan Group, which is derived from a continental arc to the northwest. Our data suggests that the Luojiamen Formation was formed in an inter-arc basin, while the Hongchicun Formation was formed in an accretionary wedge-top basin. When juxtaposed with the conglomeratic characteristics at the bottom of the Luojiamen Formation, it is believed that the unconformity represented by the ‘Shen Gong Movement' reflects the rapid erosion and accumulation process of island arc volcanic material.
