| 1 | QTL analysis across multiple environments reveals promising chromosome regions associated with yield-related traits in maize under drought conditions显示文摘Drought is one of the most critical abiotic stresses influencing maize yield. Improving maize cultivars with drought tolerance using marker-assisted selection requires a better understanding of its genetic basis. In this study, a doubled haploid(DH) population consisting of 217 lines was created by crossing the inbred lines Han 21(drought-tolerant) and Ye 478(drought-sensitive). The population was genotyped with a 6 K SNP assay and 756 SNP(single nucleotide polymorphism) markers were used to construct a linkage map with a length of 1344 c M. Grain yield(GY), ear setting percentage(ESP), and anthesis–silking interval(ASI) were recorded in seven environments under well-watered(WW) and water-stressed(WS) regimes. High phenotypic variation was observed for all traits under both water regimes. Using the LSMEAN(least-squares mean) values from all environments for each trait, 18 QTL were detected, with 9 associated with the WW and 9 with the WS regime. Four chromosome regions,Chr. 3: 219.8–223.7 Mb, Chr. 5: 191.5–194.7 Mb, Chr. 7: 132.2–135.6 Mb, and Chr. 10: 88.2–89.4 Mb, harbored at least 2 QTL in each region, and QTL co-located in a region inherited favorable alleles from the same parent. A set of 64 drought-tolerant BC_3F_6 lines showed preferential accumulation of the favorable alleles in these four regions, supporting an association between the four regions and maize drought tolerance. QTL-by-environment interaction analysis revealed 28 ed QTL(environment-dependent QTL) associated with the WS regime and 22 associated with the WW regime for GY, ESP, and ASI. All WS QTL and55.6% of WW QTL were located in the ed QTL regions. | 2 | The effects of two free-floating plants (Eichhornia crassipes and Pistia stratiotes) on the burrow morphology and water quality characteristics of pond loach (Misgurnus anguillicaudatus) habitat显示文摘Loach exhibit conspicuous drilling behaviors in the mud of shallow waters,yet their burrow morphology and the factors affecting this behavior have received little attention.We characterized the burrow morphology and water quality of the pond loach Misgurnus anguillicaudatus in three scenarios:in tanks without plants,tanks with the free-floating plant water hyacinth Eichhornia crassipes,and tanks with water lettuce Pistia stratiotes.Water hyacinth effectively removed water TN,COD,NO3-N and NH4-N,and water lettuce removed water TP and NH4-N.Water hyacinth and water lettuce markedly reduced water turbidity and DO,increased TOC and EC.Water hyacinth purified water more effectively than water lettuce,providing a suitable habitat for loach feeding,living and burrowing.The burrow structures were V-shaped,Y-shaped,inverted L-shaped,or complicated dendritic networks composed of multiple V shapes.The hyacinth treatment was characterized by the greatest burrow volume,length,depth,and structural complexity,but the opening size was reduced by dense root mat coverage.Burrows in the water lettuce treatment were characterized by intermediate volume,length,branches and sinuosity,but they had the largest opening and pit size.The control treatment had a flat bottom with the smallest,shortest burrows.This study indicates that free-floating plants improve habitat suitability and change burrow morphology and may be used to improve loach breeding methods。 | Jinqing Wang Guihua Fu Weiyue Li Ying Shi Jicai Pang Qiang Wang Weiguang Lü Change Liu Jiansheng Liu | 2018 | Aquaculture and Fisheries2018,3,1: | 2 |

| 4 | Crystal structure of histidine-containing phosphocarrier protein from Thermoanaerobacter tengcongensis MB4 and the implications for thermostability显示文摘Protein thermostability is an inherent characteristic of proteins from thermophilic microorganisms,and therefore enables these organisms to survive at extreme temperatures.Although it is well-known that thermostable proteins are critical for the growth of thermophilic organisms,the structural basis of protein thermostability is not yet fully understood.The histidine-containing phosphocarrier (HPr) protein,a phosphate shuttle protein in the phosphoenolpyruvate-dependent sugar transport system (PTS) of bacterial species,is an ideal model for investigating protein thermostability with respect to its small size and deficiency in disulphide bonds or cofactors.In this study,the HPr protein from Thermoanaerobacter tengcongensis (TtHPr) is cloned and purified.Crystal structure with good quality has been determined at 2.3 resolution,which provides a firm foundation for exploring the thermostable mechanism.However,it shows that the crystal structure is conserved and no clue can be obtained from this single structure.Furthermore,detailed comparison of sequence and structure with the homologs from mesoor thermophilic bacteria shows no obvious rule for thermostability,but the extra salt-bridge existing only in thermophilic bacteria might be a better explanation for thermostability of HPr.Thus,mutations are performed to interrupt the salt-bridge in HPrs in thermophilic bacteria.Using site-directed mutations and the circular dichroism method,thermostability is evaluated,and the mutational variations are shown to have a faster denaturing rate than for wild-type viruses,indicating that mutations cause instability in the HPrs.Understanding the higher-temperature resistance of thermophilic and hyperthermophilic proteins is essential to studies on protein folding and stability,and is critical in engineering efficient enzymes that can work at a high temperature. | FENG ChunYan GAO Feng LIU YiWei WANG GuiHua PENG Hao MA YanHe YAN JingHua GAO George Fu | 2011 | Science China(Life Sciences)2011,54,6: | 2 |
