1Grazing effects on herbage nutritive values depend on precipitation and growing season in Inner Mongolian grassland显示文摘Aims Grasslands are widely used for production of livestock,which depend on the nutritive value of herbage species.However,there are still large uncertainties as to how grazing,precipitation and growing season interactively affect herbage nutritive value.Methods Here,based on a grazing experiment with seven grazing intensities in an Inner Mongolian grassland since the year 2005,we analysed nutritive value of four dominant grass species from June to September in both a relatively wetter year(2008)and a drier year(2010).Herbage with high nutritive value is characterized by high concentration of crude protein,high concentration of cellulase digestible organic matter and low concentration of neutral detergent fibre.Important Findings We found that(i)grazing significantly increased the nutritive value of Leymus chinensis,Agropyron michnoi and Cleistogenes squarrosa but had minor effects on the nutritive value of Stipa grandis.(ii)For all species,nutritive values were greater in the wetter year than in the drier year and were greatest in the early growing season(June)and lowest at the end of the growing season(September).Inter-annual and inter-seasonal variations in nutritive value were much greater for L.chinensis and A.michnoi than for C.squarrosa and S.grandis,suggesting higher water use efficiency for the latter two species.(iii)Grazing significantly decreased the drought resistance of three species,but not of S.grandis.(iv)Grazing significantly increased inter-month variation in nutritive value for L.chinensis and A.michnoi but had relatively minor or no effects on that of C.squarrosa and S.grandis.Therefore,grazing effects on species nutritive values showed strong inter-annual and seasonal patterns,and species-specific responses might be related to species traits(i.e.water use efficiency,digestibility).Our results have important implications for ecosystem management of arid and semiarid grasslands under intense grazing and global climatic change.Haiyan Ren Guodong Han Zhichun Lan Hongwei Wan Philipp Schönbach Martin Gierus Friedhelm Taube 2016Journal of Plant Ecology2016,9,6:14
2Facile synthesis of ultrathin metal-organic framework nanosheets for Lewis acid catalysis显示文摘Ultrathin metalorganic framework (MOF) nanosheets are attracting great interest in catalysis due to their unique and intriguing two-dime nsional (2D) features. Although many progresses have been achieved, it is still highly desirable to develop novel strategies for controllable synthesis of the well-defined ultrathin MOF nanosheets. Herein we report a polyvinylpyrrolidone (PVP)-assisted route to synthesize the ultrathin Ni-MOF nanosheets characteristic of 1.5 nm in thickness, in which PVP is reacted with 2-aminoterephthalic acid (H2BDC-NH2) via formation of C=N bon d, followed by coord inatio n with Ni2+ io ns to form the ultrathi n MOF n anosheets. 4Zinc finger transcription factor Sp7/Osterix acts on bone formation and regulates col10a1a expression in zebrafish显示文摘Sp7/Osterix as a zinc finger transcription factor is expressed specifically in osteoblasts.Embryonic lethality of Sp7 knockout mice,however,has prevented from examining the functions of Sp7 in osteoblast and bone formation in live animals.Here we used TALEN,a versatile genome-editing tool,to generate one zebrafish sp7 mutant line.Homozygous sp7-/- mutant zebrafish are able to survive to adulthood.Alizarin Red staining and Micro-CT analysis showed that sp7-/- larvae and adult fish fail to develop normal opercula,and display curved tail fins and severe craniofacial malformation,while Alcian Blue staining showed no obvious cartilage defects in sp7-/- fish.Quantitative RT-PCR showed that a number of osteoblast markers including spp1,phex,col1 ala,and col1a1 b are significantly down-regulated in sp7-/- fish.Furthermore,col10a1 a,whose ortholog is the cartilage marker in mice,was shown to be a novel downstream gene of Sp7 as an osteoblast marker in zebrafish.Together,these results suggest that Sp7 is required for zebrafish bone development and zebrafish sp7 mutants provide animal models for investigating novel aspects of bone development.Pengfei Niu Zhaomin Zhong Mingyong Wang Guodong Huang Shuhao Xu Yi HOU Yilin Yan Han Wang 2017Science Bulletin2017,62,3:4

6Sustainable management of Chinese grasslands—issues and knowledge显示文摘China has almost 400 Mhm^2 of grasslands,90% of which is considered degraded to varying degrees,on which 16 million herders depend for their livelihoods and many more indirectly, along the value-adding chain.Since 1950, average stocking rates across China have increased 4-fold. National policies have focused over recent decades on finding ways to rehabilitate the degraded grasslands, to sustain livestock production from them, and to improve the livelihoods of herder households, who are among the poorest people in China. A large collaborative program commenced in the early 2000 s to help find solutions to the sustainable management of grasslands.This paper summarizes key findings of many research projects, identifies where knowledge is weak and argues that the successful rehabilitation of grasslands will also require policies that provide incentives and support for herders as they move from a focus on survival to a focus on production of higher quality products, for which consumers are increasingly willing to pay. A key focus is to emphasize the improvement of animal production per head. When this is done, it naturally leads to lower stocking rates, which in turn provides the opportunities for grasslands to recover. Across a range of experiments, farm demonstrations and analyses using models, in Inner Mongolia and Gansu, a consistent result has been that a 50% reduction in stocking rates, improves net household income and starts the process of grassland rehabilitation.Rather than focusing on stocking rates, better management of grasslands could be achieved by maintaining the grasslands above critical values for herbage mass, values that help optimize botanical composition, reduce soil erosion, optimize animal growth rates and aid ecosystem functions. Managing to critical values for herbage mass is likely to be more effective than efforts to calculate sustainable stocking rates. An early summer rest is valuable for aiding grassland rehabilitation and summer productivity, but a total grazing ban(typically for 5 years)may not achieve its aims as evidence shows it may take 10–15 years to achieve a better grassland state. Lessdesirable plant species often increase in degraded grasslands that are rested and grazing can help manage those species. Surveys of herders indicate they have very mixed views on the benefits of total grazing bans that are unlikely to rehabilitate grasslands to an ideal botanical composition.The current objective is to work with grasslands that herders now have and optimize the existing composition.Grazing grasslands in winter results primarily in weight loss by animals and there is now evidence of how winter grazing reduces grassland growth in the next summer. It is better to keep animals in well-built sheds and feed them better, improved feeding through the cold months is required. 7The mechanosensitive lncRNA Neat1 promotes osteoblast function through paraspeckle-dependent Smurf1 mRNA retention显示文摘Mechanical stimulation plays an important role in bone remodeling. Exercise-induced mechanical loading enhances bone strength,whereas mechanical unloading leads to bone loss. Increasing evidence has demonstrated that long noncoding RNAs(lnc RNAs) play key roles in diverse biological, physiological and pathological contexts. However, the roles of lnc RNAs in mechanotransduction and their relationships with bone formation remain unknown. In this study, we screened mechanosensing lnc RNAs in osteoblasts and identified Neat1, the most clearly decreased lnc RNA under simulated microgravity. Of note, not only Neat1 expression but also the specific paraspeckle structure formed by Neat1 was sensitive to different mechanical stimulations, which were closely associated with osteoblast function. Paraspeckles exhibited small punctate aggregates under simulated microgravity and elongated prolate or larger irregular structures under mechanical loading. Neat1 knockout mice displayed disrupted bone formation, impaired bone structure and strength, and reduced bone mass. Neat1 deficiency in osteoblasts reduced the response of osteoblasts to mechanical stimulation. In vivo, Neat1 knockout in mice weakened the bone phenotypes in response to mechanical loading and hindlimb unloading stimulation. Mechanistically, paraspeckles promoted nuclear retention of E3 ubiquitin ligase Smurf1 m RNA and downregulation of their translation, thus inhibiting ubiquitination-mediated degradation of the osteoblast master transcription factor Runx2, a Smurf1 target. 8Mass loss and nutrient dynamics during litter decomposition in response to warming and nitrogen addition in a desert steppe显示文摘Plant litter decomposition has been studied extensively in the context of both warming and increased atmospheric nitrogen deposition. However, the temporal patterns of mass loss and nutrient release in response to warming and nitrogen addition remain unclear. A 2-year decomposition experiment aimed to examine the effects of warming and nitrogen addition on decomposition rate, and nitrogen and phosphorus dynamics. The effects of warming and nitrogen addition on decomposition of litter of Stipa breviflora, a dominant species in a desert steppe of northern China, were studied. Warming and nitrogen addition significantly enhanced litter mass loss by 10% and 16%, respectively, and moreover promoted nitrogen and phosphorus release from the litter in the first year of decomposition, followed by an immobilization period. The interactive effects of warming and nitrogen addition on mass loss, nitrogen and phosphorus concentrations of litter were also found during the decomposition. This study indicates that warming and nitrogen addition increased litter mass loss through altering litter quality. 10SOD1^(G93A) Induces a Unique PSAP-Dependent Mitochondrial Apoptosis Pathway via Bax-Bak Interaction显示文摘Amyotrophic lateral syndrome(ALS)is a progressive degenerative disorder characterized by motor neuron death and axon degeneration.Mitochondrial dysfunction plays a key role in the pathogenesis of ALS,the mechanism of which remains poorly understood.The B-cell lymphoma-2(Bcl-2)family of proteins that control and mediate mitochondrial function and apoptosis,including the pro-apoptotic members Bcl2-Associated X(Bax),are involved in ALS development.The death receptor 6(DR6)regulates motor neuron death in ALS,and DR6 antibodies can prevent axon degeneration and motor neuron damage by blocking DR6.Previous studies demonstrated that PSAP localized to mitochondria and was required for DR6-induced apoptosis.In this study,SOD1^(G93A) was transfected into the motor neuron cell line NSC-34 to serve as an ALS cell model in vitro.The data assessed the role of PSAP in SOD1^(G93A)induced apoptosis and demonstrated that the overexpression of SOD1^(G93A),but not wtSOD1,induced PARP cleavage,caspase-3 activation,cytochrome c release,and Bax translocation.PSAP,Bax,and Bak were necessary for SOD1^(G93A)induced apoptosis,as silencing PSAP inhibited SOD1^(G93A)-mediated cell death that was dependent on Bax-Bak interaction.HAN NIU XIN CHEN XUEQI FU JINGTIAN ZHANG GUODONG LI YUXIANG WANG JIAYUE SONG XUETING MA CHEN HU XUEMIN XU FUQIANG ZHANG LINLIN ZENG 2021BIOCELL2021,45,4:2
11Molecular Characterization of China Human Rabies Vaccine Strains显示文摘To understand the molecular characteristics of China human rabies vaccine strains, we report the full-length genome of the aG strain and present a comprehensive analysis of this strain and almost all available lyssavirus genomes (58 strains) from GenBank (as of Jan 6, 2011). It is generally considered that the G protein plays a predominant role in determining the pathogenicity of the virus, to this end we predicted the tertiary structure of the G protein of aG strain, CTN181 strain and wild type strain HN10 based on the crystal structure of Vesicular stomatitis virus (VSV) G. The predicted RABV G structure has a similar topology to VSV G and the ectodomain can be divided into 4 distinct domains DI-DIV. By mapping the characterized mutations to this structure between China vaccine strains and their close street strains, we speculate that the G303(P-H) mutations of CTN181 and HN10 causing DⅡ 3D change may be associated with the attenuated virulence in both strains. 13Loss-of-function of KMT5B leads to neurodevelopmental disorder and impairs neuronal development and neurogenesis显示文摘Autism spectrum disorder(ASD)is a group of neurodevelopmental disorders that cause severe social,communication,and behavioral problems.Recent studies show that the variants of a histone methyltransferase gene KMT5B cause neurodevelopmental disorders(NDDs),including ASD,and the knockout of Kmt5b in mice is embryonic lethal.However,the detailed genotype-phenotype correlations and functional effects of KMT5B in neurodevelopment are unclear.By targeted sequencing of a large Chinese ASD cohort,analyzing published genome-wide sequencing data,and mining literature,we curated 39 KMT5B variants identified from NDD individuals.A genotype-phenotype correlation analysis for 10 individuals with KMT5B pathogenic variants reveals common symptoms,including ASD,intellectual disability,languages problem,and macrocephaly.In vitro knockdown of the expression of Kmt5b in cultured mouse primary cortical neurons leads to a decrease in neuronal dendritic complexity and an increase in dendritic spine density,which can be rescued by expression of human KMT5B but not that of pathogenic de novo missense mutants.In vivo knockdown of the Kmt5b expression in the mouse embryonic cerebral cortex by in utero electroporation results in decreased proliferation and accelerated migration of neural progenitor cells.Our findings reveal essential roles of histone methyltransferase KMT5B in neuronal development,prenatal neurogenesis,and neuronal migration.Guodong Chen Lin Han Senwei Tan Xiangbin Jia Huidan Wu Yingting Quan Qiumeng Zhang Bin Yu Zhengmao Hu Kun Xia Hui Guo 2022Journal of Genetics and Genomics2022,49,9:1
