2Carbon Isotope Features of the Sugetbrak Section in the Aksu-Wushi Area, Northwest China: Implications for the Precambrian/Cambrian Stratigraphic Correlations显示文摘The upper Qigeblaq Formation(Fm) dolostones and the Yurtus Fm phosphatic cherts, black shales, limestones, and dolostones are widely distributed in the Precambrian/Cambrian transitional succession of the Aksu-Wushi area. Negative δ13C excursion above the Yurtus Fm/Qigeblaq Fm boundary was determined in this study. The pronounced negative carbon isotope excursion occurs in the phosphatic chert layers at the bottom of the Cambrian Yurtus Fm, below which the first appearance of the Asteridium- Heliosphaeridium-Comasphaeridium(AHC) acritarch assemblage zone. The δ13C curve of the lower part of the Yurtus Fm in the Aksu-Wushi area was found to be correlated with the early Cambrian δ13C curves of the Zhujiaqing Fm(Daibu Member), the lower part of the Yanjiahe Fm on the Yangtze Platform in China, the lower Tal Fm in India, the Sukharikha Fm in Siberia, and the upper part of the Tsagaan Oloom Fm in Mongolia through biostigraphy. The lower part of the Yurtus Fm in the Tarim Basin is at the Nemakit-Daldynian stage, and the Precambrian/Cambrian boundary of the Aksu-Wushi area may be located in the phosphatic chert unit which just below the first appearance AHC acritarch assemblage zone. 7Targeting ATAD3A-PINK1-mitophagy axis overcomes chemoimmunotherapy resistance by redirecting PD-L1 to mitochondria显示文摘Only a small proportion of patients with triple-negative breast cancer benefit from immune checkpoint inhibitor(ICI)targeting PD-1/PD-L1 signaling in combination with chemotherapy.Here,we discovered that therapeutic response to ICI plus paclitaxel was associated with subcellular redistribution of PD-L1.In our immunotherapy cohort of ICI in combination with nab-paclitaxel,tumor samples from responders showed significant distribution of PD-L1 at mitochondria,while non-responders showed increased accumulation of PD-L1 on tumor cell membrane instead of mitochondria.Our results also revealed that the distribution pattern of PD-L1 was regulated by an ATAD3A-PINK1 axis.Mechanistically,PINK1 recruited PD-L1 to mitochondria for degradation via a mitophagy pathway.Importantly,paclitaxel increased ATAD3A expression to disrupt proteostasis of PD-L1 by restraining PINK1-dependent mitophagy.Clinically,patients with tumors exhibiting high expression of ATAD3A detected before the treatment with ICI in combination with paclitaxel had markedly shorter progression-free survival compared with those with ATAD3A-low tumors.Preclinical results further demonstrated that targeting ATAD3A reset a favorable antitumor immune microenvironment and increased the efficacy of combination therapy of ICI plus paclitaxel.In summary,our results indicate that ATAD3A serves not only as a resistant factor for the combination therapy of ICI plus paclitaxel through preventing PD-L1 mitochondrial distribution,but also as a promising target for increasing the therapeutic responses to chemoimmunotherapy.Xiao-Qing Xie Yi Yang Qiang Wang Hao-Fei Liu Xuan-Yu Fang Cheng-Long Li Yi-Zhou Jiang Shuai Wang Hong-Yu Zhao Jing-Ya Miao Shuai-Shuai Ding Xin-Dong Liu Xiao-Hong Yao Wen-Tao Yang Jun Jiang Zhi-Ming Shao Guoxiang Jin Xiu-Wu Bian 2023Cell Research2023,33,3:1
8Diffusion Characteristics of Swells in the North Indian Ocean显示文摘Research on the diffusion characteristics of swells contributes positively to wave energy forecasting, swell monitoring, and early warning. In this work, the South Indian Ocean westerly index(SIWI) and Indian Ocean swell diffusion effect index(IOSDEI) are defined on the basis of the 45-year(September 1957–August 2002) ERA-40 wave reanalysis data from the European Centre for Medium-Range Weather Forecasts(ECMWF) to analyze the impact of the South Indian Ocean westerlies on the propagation of swell acreage. The following results were obtained: 1) The South Indian Ocean swell mainly propagates from southwest to northeast. The swell also spreads to the Arabian Sea upon reaching low-latitude waters. The 2.0-meter contour of the swell can reach northward to Sri Lankan waters. 2) The size of the IOSDEI is determined by the SIWI strength. The IOSDEI requires approximately 2–3.5 days to fully respond to the SIWI. The correlations between SIWI and IOSDEI show obvious seasonal differences, with the highest correlations found in December–January–February(DJF) and the lowest correlations observed in June–July–August(JJA). 3) The SIWI and IOSDEI have a common period of approximately 1 week in JJA and DJF. 11Expression of human erythropoietin directed by mWAP promoter in m鈓mary gland of transgenic mice显示文摘The present work has generated transgenic mice with a hybrid gene construct consisting of genomic sequences encoding human erythropoietin (hEPO) and governed by regulatory sequences of mouse whey acidic protein (mWAP). The construct proved effective by transient expression in lactating animal. After introducing hybrid gene construct into single-cell embryo via pronuclear microinjection, surviving embryo are reimplanted into pseudopregnant foster mother mouse. 58 mice of 86 generation zero mice obtained were identified to be positive by PCR-Southern blot and genomic DNA Southern blot methods. 12Pancreatic cancer–Palliative therapy: surgery (bypass/resection)显示文摘Objective: We compare the outcome of palliative pancreaticoduodenectomy and palliative surgical bypass in patients with advanced pancreatic carcinoma in our hospital. Recent published related articles are also reviewed. Methods: A respective analysis was performed comparing the perioperative parameters and outcome of 20 patients who underwent pancreaticoduodenectomy with a gross suspected cancer residue and 30 patients who underwent a surgical bypass, all of the patients were diagnosed as in advanced stages intra-operatively. Results: The two groups were comparable with patient characteristics, including age, gender, initial symptoms and concomitant major organ diseases. Tumors are similar in size and intra-operatively diagnosed as in advanced stages in both groups. All of the patients in the resection group were microscopically proved having cancer residue. One postoperative mortality occurred in the resection group (5%), zero in the bypass group (P > 0.05). Overall complications were significantly higher in the resection group (30% vs. 0, P < 0.01), including 2 patients developed Acute Respiratory Distress Syndrome (ARDS), zero in the bypass group (P < 0.01); hemorrhage and transfusions in the resection group were much more than that in the bypass group (P < 0.05). Hospital stay after resection was significantly longer than bypass (20 vs. 12 days, P < 0.01). Hospital fee after resection was 4 times more than after bypass (median 61.500 vs. 15. 300 yuan, P < 0.01). Survival was significantly longer after resection (median 12.2 vs. 7.1 months, P < 0.01). Conclusion: Our results show that palliative resection in advanced pancreatic carcinoma lengthens the survival time of the patients, but this is paid for significantly higher complications than bypass.Xiaoping Chen Guoxiang Yao 2007The Chinese-German Journal of Clinical Oncology2007,6,2:0
