1Effects of Saccharomycesboulardiion fecal short-chain fatty acids and microflora in patients on long-term total enteral nutrition显示文摘AIM: To assess the effects of Sb on fecal flora and shortchain fatty acids (SCFA) in patients on long-term TEN.METHODS: Ten patients (3 females, 7 males, 59±5.5 years),on TEN for a median of 13 mo (1-125), and 15 healthy volunteers (4 females, 11 males, 32±2.0 years) received Sb (0.5 g bid PO) for 6 d. Two stool samples were taken before, on the last 2 d and 9-10 d after treatment, for SCFA measurement and for culture and bacterial identification.Values (mean±SE) were compared using sign tests and ANOVA.RESULTS: Fecal butyrate levels were lower in patients(10.1±2.9 mmol/kg) than in controls (19.2±3.9, P= 0.02).Treatment with Sb increased total fecal SCFA levels in patients (150.2±27.2 vs 107.5±18.2 mmol/kg, P = 0.02)but not in controls (129.0±28.6 vs 113.0±15.2 mmol/kg,NS). At the end of treatment with Sb, patients had higher fecal butyrate (16.0±4.4 vs 10.1 [2.9] mmol/kg, P = 0.004).Total SCFAs remained high 9 d after treatment was discontinued. Before the treatment, the anaerobe to aerobe ratio was lower in patients compared to controls (2.4±2.3 vs69.8±1.8, P = 0.003). 2miR-182参与调节胶质母细胞瘤的凋亡、生长和分化(英文)显示文摘Glioblastoma multiforme(GBM)is a lethal,therapy-resistant brain cancer consisting of numerous tumor cell subpopulations,including stem-like glioma-initiating cells(GICs),which contribute to tumor recurrence following initial response to therapy.Here,we identified miR-182 as a regulator of apoptosis,growth,and differentiation programs whose expression level is correlated with GBM patient survival.Repression of Bcl2-like12(Bcl2L12),c-Met,and hypoxia-inducible factor 2α(HIF2A)is of central importance to miR-182 anti-tumor activity,as it results in enhanced therapy susceptibility,decreased GIC sphere size,expansion,and stemness in vitro.To evaluate the tumor-suppressive function of miR-182 in vivo,we synthesized miR-182-based spherical nucleic acids(182-SNAs);i.e.,gold nanoparticles covalently functionalized with mature miR-182 duplexes.Intravenously administered 182-SNAs penetrated the bloodbrain/blood-tumor barriers(BBB/BTB)in orthotopic GBM xenografts and selectively disseminated throughout extravascular glioma parenchyma,causing reduced tumor burden and increased animal survival.Our results indicate that harnessing the anti-tumor activities of miR-182 via safe and robust delivery of 182-SNAs represents a novel strategy for therapeutic intervention in GBM.Kouri FM Hurley LA Daniel WL Day ES Hua Y Hao L Peng CY Merkel TJ Queisser MA Ritner C Zhang H James CD Sznajder JI Chin L Giljohann DA Kessler JA Peter ME Mirkin CA Stegh AH 2015中华神经外科疾病研究杂志2015,14,2:14
3小胶质细胞可能通过选择性吞噬丘脑底核谷氨酸能突触来代偿多巴胺能神经元丢失造成的帕金森模型鼠的功能缺失显示文摘当帕金森患者出现临床症状时,其黑质致密部的绝大部分多巴胺神经元已发生退行性改变。这提示可能存在某些代偿机制,延缓了临床症状的出现。本研究制作6-羟多巴胺诱导的偏侧帕金森大鼠模型,观察位于黑质下网状部和苍白球的激活的小胶质细胞在代偿机制中的作用。研究发现,在偏侧帕金森大鼠中,黑质下网状部聚集的激活的小胶质细胞远多于苍白球中。激活的小胶质细胞胞体增大并表达吞噬物标志物CD68和NG2蛋白多糖。激活的小胶质细胞聚集在黑质下网状部的特定部位,该部位的突触蛋白I和突触后密集蛋白95的表达降低。活化的小胶质细胞吞噬突触前、后膜上的各种蛋白,包括NMDA受体。将红色荧光标记物Di I作为顺行示踪剂注入丘脑底核,可见黑质下网状部和苍白球内的细胞吞噬了Di I。在谷氨酸刺激下,原代培养的小胶质细胞吞噬活动增强,编码吞噬相关因子的m RNA转录水平也增加。人工合成的糖皮质激素地塞米松可以减弱这种改变。给PD大鼠皮下注射地塞米松,可以抑制黑质下网状部小胶质细胞的活化,运动功能障碍进一步加重,谷氨酸能突触转录m RNA增加,GABA能突触转录m RNA水平未增加。这些发现提示,当谷氨酸能神经元活动增强时,黑质下网状部和苍白球的小胶质细胞活化,选择性吞噬丘脑底核的谷氨酸能突触。因此,在基底核区可能存在一个负反馈环路,间接地代偿了由于多巴胺能神经元丢失而导致的功能障碍。小胶质细胞在这个负反馈环路中扮演了重要的角色。Aono H Choudhury ME Higaki H Miyanishi K Kigami Y Fujita K Akiyama JI Takahashi H Yano H Kubo M Nishikawa N Nomoto M Tanaka J 唐颖馨 2017神经损伤与功能重建2017,12,5:7
42001年1月26日高纬磁层顶通量管事件的观测研究显示文摘20 0 1年 1月 2 6日 11:10~ 11:4 0UT ,ClusterⅡ卫星簇位于午后高纬磁鞘边界层和磁鞘区 ,此时行星际磁场Bz 为南向 .本文对在此期间观测到的多次磁通量管事件作了详细的研究 ,获得一系列的新发现 :(1)高纬磁鞘边界层磁通量管的出现具有准周期性 ,周期约为 78s,比目前已知的磁层顶向阳面FTE的平均周期 (8~ 11min)小得多 .(2 )这些通量管都具有强的核心磁场 ;其主轴多数在磁场最小变化方向 ,少数在中间变化方向 ,有些无法用PAA判定其方向 (需要用电流管PAA确定 ) ,这与卫星穿越通量管的相对路径有关 .(3)每个事件都存在很好的HT参考系 ,在HT参考系中这些通量管是准定常态结构 ;所有通量管都沿磁层顶表面运动 ,速度方向大体相同 ,都来自晨侧下方 .通量管的径向尺度为 1~ 2RE,与通常的FTE通量管相当 .(4 )起源于磁层的强能离子大体上沿着管轴方向由磁层向磁鞘运动 ;起源于太阳风的热等离子体沿管轴向磁层传输 .通量管为太阳风等离子体向磁层输运和磁层粒子向行星际空间逃逸提供了通道 .(5 )每个通量管事件都伴随有晨昏电场的反转 ,该电场为对流电场 .黄宗英 濮祖荫 肖池阶 宗秋刚 傅绥燕 谢伦 史全岐 曹晋滨 刘振兴 沈超 史建魁 路立 王迺权 陈涛 T Fritz K-H Glassmeier P Daly H Rème 2004地球物理学报2004,47,2:7

7Interferon-β induced in female genital epithelium by HIV-1 glycoprotein 120 via Toll-like-receptor 2 pathway acts to protect the mucosal barrier显示文摘More than 40%of HIV infections occur via female reproductive tract(FRT)through heterosexual transmission.Epithelial cells that line the female genital mucosa are the first line of defense against HIV-1 and other sexually transmitted pathogens.These sentient cells recognize and respond to external stimuli by induction of a range of carefully balanced innate immune responses.Previously,we have shown that in response to HIV-1 gp120,the genital epithelial cells(GECs)from upper reproductive tract induce an inflammatory response that may facilitate HIV-1 translocation and infection.In this study,we report that the endometrial and endocervical GECs simultaneously induce biologically active interferon-β(IFNβ)antiviral responses following exposure to HIV-1 that act to protect the epithelial tight junction barrier.The innate antiviral response was directly induced by HIV-1 envelope glycoprotein gp120 and addition of gp120 neutralizing antibody inhibited IFNβproduction.Interferon-βwas induced by gp120 in upper GECs through Toll-like receptor 2 signaling and required presence of heparan sulfate on epithelial cell surface.The induction of IFNβwas dependent upon activation of transcription factor IRF3(interferon regulatory factor 3).The IFNβwas biologically active,had a protective effect on epithelial tight junction barrier and was able to inhibit HIV-1 infection in TZM-bl indicator cells and HIV-1 replication in T cells.This is the first report that recognition of HIV-1 by upper GECs leads to induction of innate antiviral pathways.This could explain the overall low infectivity of HIV-1 in the FRT and could be exploited for HIV-1 prophylaxis.Aisha Nazli Sara Dizzell Muhammad Atif Zahoor Victor H Ferreira Jessica Kafka Matthew William Woods Michel Ouellet Ali A Ashkar Michel J Tremblay Dawn ME Bowdish Charu Kaushic 2019Cellular & Molecular Immunology2019,16,2:2
