| 1 | HDAC inhibitor chidamide synergizes with venetoclax to inhibit the growth of diffuse large B-cell lymphoma via down-regulation of MYC, BCL2, and TP53 expression显示文摘Diffuse large B-cell lymphoma(DLBCL) is an aggressive type of non-Hodgkin’s lymphoma. A total of 10%-15% of DLBCL cases are associated with myelocytomatosis viral oncogene homolog(MYC) and/or B-cell lymphoma-2(BCL2) translocation or amplification. BCL2 inhibitors have potent anti-tumor effects in DLBCL;however, resistance can be acquired through up-regulation of alternative anti-apoptotic proteins. The histone deacetylase(HDAC) inhibitor chidamide can induce BIM expression, leading to apoptosis of lymphoma cells with good efficacy in refractory recurrent DLBCL. In this study, the synergistic mechanism of chidamide and venetoclax in DLBCL was determined through in vitro and in vivo models. We found that combination therapy significantly reduced the protein levels of MYC, TP53, and BCL2 in activated apoptotic-related pathways in DLBCL cells by increasing BIM levels and inducing cell apoptosis. Moreover, combination therapy regulated expression of multiple transcriptomes in DLBCL cells, involving apoptosis, cell cycle, phosphorylation, and other biological processes, and significantly inhibited tumor growth in DLBCL-bearing xenograft mice. | 3 | Interfacial microstructure and mechanical properties of diffusion-bonded joints of titanium TC4 (Ti-6Al-4V) and Kovar (Fe-29Ni-17Co) alloys显示文摘Diffusion bonding is a near net shape forming process that can join dissimilar materials through atomic diffusion under a high pressure at a high temperature.Titanium alloy TC4(Ti-6 Al-4 V)and 4 J29 Kovar alloy(Fe-29 Ni-17 Co)were diffusely bonded by a vacuum hot-press sintering process in the temperature range of 700-850°C and bonding time of 120 min,under a pressure of 34.66 MPa.Interfacial microstructures and intermetallic compounds of the diffusion-bonded joints were characterized by optical microscopy,scanning electron microscopy,X-ray diffraction(XRD)and energy dispersive spectroscopy(EDS).The elemental diffusion across the interface was revealed by electron probe microanalysis.Mechanical properties of joints were investigated by micro Vickers hardness and tensile strength.Results of EDS and XRD indicated that(Fe,Co,Ni)-Ti,TiNi,Ti_2Ni,TiNi_2,Fe_2 Ti,Ti_(17) Mn_3 and Al_6 Ti_(19) were formed at the interface.When the bonding temperature was raised from 700 to 850°C,the voids of interface were reduced and intermetallic layers were widened.Maximum tensile strength of joints at 53.5 MPa was recorded by the sintering process at 850°C for 120 min.Fracture surface of the joint indicated brittle nature,and failure took place through interface of intermetallic compounds.Based on the mechanical properties and microstructure of the diffusion-bonded joints,diffusion mechanisms between Ti-6 Al-4 Vtitanium and Fe-29 Ni-17 Co Kovar alloys were analyzed in terms of elemental diffusion,nucleation and growth of grains,plastic deformation and formation of intermetallic compounds near the interface. | Ting-feng Song Xiao-song Jiang Zhen-yi Shao De-feng Mo De-gui Zhu Min-hao Zhu Christina H.Young Zhi-ping Luo | 2017 | Journal of Iron and Steel Research(International)2017,24,10: | 1 |

| 5 | Cross-talk between Myc and p53 in B-cell lymphomas显示文摘Myc and p53 proteins are closely associated with many physiological cellular functions,including immune response and lymphocyte survival,and are expressed in the lymphoid organs,which are sites for the development and activation of B-cell malignancies.Genetic alterations and other mechanisms resulting in constitutive activation,rearrangement,or mutation of MYC and TP53 contribute to the development of lymphomas,progression and therapy resistance by gene dysregulation,activation of downstream anti-apoptotic pathways,and unfavorable microenvironment interactions.The cross-talk between the Myc and p53 proteins contributes to the inferior prognosis in many types of B-cell lymphomas.In this review,we present the physiological roles of Myc and p53 proteins,and recent advances in understanding the pathological roles of Myc,p53,and their cross-talk in lymphoid neoplasms.In addition,we highlight clinical trials of novel agents that directly or indirectly inhibit Myc and/or p53 protein functions and their signaling pathways.Although,to date,these trials have failed to overcome drug resistance,the new results have highlighted the clinical efficiency of targeting diverse mechanisms of action with the goal of optimizing novel therapeutic opportunities to eradicate lymphoma cells. | Li Yu Tian-Tian Yu Ken H.Young | 2019 | Chronic Diseases and Translational Medicine2019,5,3: | 0 |
