1Effects of warming and clipping on plant and soil properties of an alpine meadow in the Qinghai-Tibetan Plateau, China显示文摘Climate warming and livestock grazing are known to have great influences on alpine ecosystems like those of the Qinghai-Tibetan Plateau(QTP) in China. However, it is lacking of studies on the effects of warming and grazing on plant and soil properties in these alpine ecosystems. In this study, we reported the related research from manipulative experiment in 2010–2012 in the QTP. The aim of this study was to investigate the individual and combined effects of warming and clipping on plant and soil properties in the alpine meadow ecosystem. Infrared radiators were used to simulate climate warming starting in July 2010, while clipping was performed once in October 2011 to simulate the local livestock grazing. The experiment was designed as a randomized block consisting of five replications and four treatments: control(CK), warming(W), clipping(C) and warming+clipping combination(WC). The plant and soil properties were investigated in the growing season of the alpine meadow in 2012. The results showed that W and WC treatments significantly decreased relative humidity at 20-cm height above ground as well as significantly increases air temperature at the same height, surface temperature, and soil temperature at the depth of 0–30 cm. However, the C treatment did not significantly decrease soil moisture and soil temperature at the depth of 0–60 cm. Relative to CK, vegetation height and species number increased significantly in W and WC treatment, respectively, while vegetation aboveground biomass decreased significantly in C treatment in the early growing season. However, vegetation cover, species diversity, belowground biomass and soil properties at the depth of 0–30 cm did not differ significantly in W, C and WC treatments. Soil moisture increased at the depth of 40–100 cm in W and WC treatments, while belowground biomass, soil activated carbon, organic carbon and total nitrogen increased in the 30–50 cm soil layer in W, C and WC treatments. Although the initial responses of plant and soil properties to experimental warming and clipping were slow and weak, the drought induced by the downward shift of soil moisture in the upper soil layers may induce plant belowground biomass to transfer to the deeper soil layers. This movement would modify the distributions of soil activated carbon, organic carbon and total nitrogen. 3CPSF30-L-mediated recognition of mRNA m^(6)A modification controls alternative polyadenylation of nitrate signaling-related gene transcripts in Arabidopsis显示文摘N6-methyladenosine(m^(6)A),a ubiquitous internal modification of eukaryotic mRNAs,plays a vital role in almost every aspect of mRNA metabolism.However,there is little evidence documenting the role of m^(6)A in regulating alternative polyadenylation(APA)in plants.APA is controlled by a large protein-RNA complex with many components,including CLEAVAGE AND POLYADENYLATION SPECIFICITY FACTOR30(CPSF30).In Arabidopsis,CPSF30 has two isoforms and the longer isoform(CPSF30-L)contains a YT512-B Homology(YTH)domain,which is unique to plants.In this study,we showed that CPSF30-L YTH domain binds to m^(6)A in v itro.In the cpsf30-2 mutant,the transcripts of many genes including several important nitrate signaling-related genes had shifts in polyadenylation sites that were correlated with m^(6)A peaks,indicating that these gene transcripts carrying m^(6)A tend to be regulated by APA.Wild-type CPSF30-L could rescue the defects in APA and nitrate metabolism in cpsf30-2,but m^(6)A-binding-defective mutants of CPSF30-L could not.Taken together,our results demonstrated that m^(6)A modification regulates APA in Arabidops is and revealed that the m^(6)A reader CPSF30-L affects nitrate signaling by controlling APA,shedding new light on the roles of the m^(6)A modification during RNA 3-end processing in nitrate metabolism.Yiteng Hou Jing Sun Baixing Wu Yangyang Gao Hongbo Nie Zhentian Nie Shuxuan Quan Yong Wang Xiaofeng Cao Sisi Li 2021Molecular Plant2021,14,4:12
4Development of a Drilling and Coring Test-bed for Lunar Subsurface Exploration and Preliminary Experiments显示文摘Drill sampling has been widely employed as an effective way to acquire deep samples in extraterrestrial exploration. A novel sampling method, namely, flexible-tube coring, was adopted for the Chang'e mission to acquire drilling cores without damaging stratification information. Since the extraterrestrial environment is uncertain and different from the terrestrial environment, automated drill sampling missions are at risk of failure. The principles of drilling and coring for the lunar subsurface should be fully tested and verified on earth before launch. This paper proposes a test-bed for conducting the aforementioned experiments on earth. The test-bed comprises a rotary-percussive drilling mechanism, penetrating mechanism, drilling medium container, and signal acquisition and control system. For granular soil, coring experiments indicate that the sampling method has a high coring rate greater than 80%. For hard rock, drilling experiments indicate that the percussive frequency greatly affects the drilling efficiency. A multi-layered simulant composed of granular soil and hard rock is built to test the adaptability of drilling and coring. To tackle complex drilling media, an intelligent drilling strategy based on online recognition is proposed to improve the adaptability of the sampling drill. 6The role of glycogen synthase kinase 3 beta in brain injury induced by myocardial ischemia/reperfusion injury in a rat model of diabetes mellitus显示文摘Myocardial ischemia/reperfusion injury can lead to severe brain injury.Glycogen synthase kinase 3 beta is known to be involved in myocardial ischemia/reperfusion injury and diabetes mellitus.However,the precise role of glycogen synthase kinase 3 beta in myocardial ischemia/reperfusion injury-induced brain injury is unclear.In this study,we observed the effects of glycogen synthase kinase 3 beta on brain injury induced by myocardial ischemia/reperfusion injury in diabetic rats.Rat models of diabetes mellitus were generated via intraperitoneal injection of streptozotocin.Models of myocardial ischemia/reperfusion injury were generated by occluding the anterior descending branch of the left coronary artery.Post-conditioning comprised three cycles of ischemia/reperfusion.Immunohistochemical staining and western blot assays demonstrated that after 48 hours of reperfusion,the structure of the brain was seriously damaged in the experimental rats compared with normal controls.Expression of Bax,interleukin-6,interleukin-8,terminal deoxynucleotidyl transferase d UTP nick end labeling,and cleaved caspase-3 in the brain was significantly increased,while expression of Bcl-2,interleukin-10,and phospho-glycogen synthase kinase 3 beta was decreased.Diabetes mellitus can aggravate inflammatory reactions and apoptosis.Ischemic post-conditioning with glycogen synthase kinase 3 beta inhibitor lithium chloride can effectively reverse these changes.Our results showed that myocardial ischemic post-conditioning attenuated myocardial ischemia/reperfusion injury-induced brain injury by activating glycogen synthase kinase 3 beta.According to these results,glycogen synthase kinase 3 beta appears to be an important factor in brain injury induced by myocardial ischemia/reperfusion injury.Bo Zhao Wen-wei Gao Ya-jing Liu Meng Jiang Lian Liu Quan Yuan Jia-bao Hou Zhong-yuan Xia 2017Neural Regeneration Research2017,12,10:8
7Bilayer dissolving microneedle array containing 5-fluorouracil and triamcinolone with biphasic release profile for hypertrophic scar therapy显示文摘Hypertrophic scar(HS)is an undesirable skin abnormality following deep burns or operations.Although intralesional multi-injection with the suspension of triamcinolone acetonide(TA)and 5-fluorouracil(5-Fu)has exhibited great promise to HS treatment in clinical,the difference of metabolic behavior between TA and 5-Fu remarkably compromised the treatment efficacy.Besides,the traditional injection with great pain is highly dependent on the skill of the experts,which results in poor compliance.Herein,a bilayer dissolving microneedle(BMN)containing TA and 5-Fu(TA-5-Fu-BMN)with biphasic release profile was designed for HS therapy.Equipped with several micro-scale needle tips,the BMN could be self-pressed into the HS with uniform drug distribution and less pain.Both in vitro permeation and in vivo HS retention tests revealed that TA and 5-Fu could coexist in the scar tissue for a sufficient time period due to the well-designed biphasic release property.Subsequently,the rabbit ear HS model was established to assess therapeutic efficacy.The histological analysis showed that TA-5-Fu-BMN could significantly reduce abnormal fibroblast proliferation and collagen fiber deposition.It was also found that the value of scar elevation index was ameliorated to a basal level,together with the downregulation of mRNA and protein expression of Collagen I(Col I)and transforming growth factor-β1(TGF-β1)after application of TA-5-Fu-BMN.In conclusion,the BMN with biphasic release profiles could serve as a potential strategy for HS treatment providing both convenient administrations as well as controlled drug release behavior.Beibei Yang Yating Dong Yifeng Shen Ailin Hou Guilan Quan Xin Pan Chuanbin Wu 2021Bioactive Materials2021,6,8:8
8Facile synthesis of graphene via reduction of graphene oxide by artemisinin in ethanol显示文摘The preparation of reduced graphene oxide(RGO)by chemical reduction of graphene oxide(GO)usually involves highly toxic reducing agents which are harmful to the environment and human health.In this paper,a mediated facile and relative green approach for the preparation of RGO in ethanol using artemisinin as a reducing agent is reported for the first time.The morphology and de-oxidation efficiency of the obtained RGO were characterized by transmission electron microscope(TEM),atomic force microscope(AFM),and X-ray photoelectron spectroscopy(XPS).The results showed that artemisinin can effectively reduce GO into few-layered RGO with a high carbon to oxygen ratio(11.7).The mechanism for elimination of oxygen-containing functional groups decorated on GO nanosheets by artemisinin was proposed.The important features of relatively environmentally friendly and facile operation procedures endow this approach with great promise in the mass production of RGO and various graphene-based materials,especially for biomaterials.Dandan Hou Qinfu Liu Xianshuai Wang Ying Quan Zhichuan Qiao Li Yu Shuli Ding 2018Journal of Materiomics2018,4,3:6
9New research trends on high-precision time transfer technology显示文摘High-precision time transfer plays an important role in the areas of fundamental research and applications. Accompanying w ith the remarkable improvements in the ability of generating and measuring high-accuracy time-frequency signal,seeking for new time-transfer techniques betw een distant clocks w ith much further improved accuracy attracts attentions w orld-w idely. The time-transfer technique based on optical pulses has the highest precision presently,and the further improvement in the accuracy is heavily dependent on the time-domain properties of the pulse as w ell as the sensitivity of the applied measurement on the exchanged pulse. The application of optical frequency comb in time transfer for a precision up to femtosecond level are currently the focus of much interest,and has recently achieved many breakthroughs. Further investigations show that,utilizing quantum techniques,i.e. quantum measurement technique and quantum optical pulse source,can lead to a new limit on the measured timing information. Furthermore,it can be immune from atmospheric parameters,such as pressure,temperature,humidity and so on.Such quantum improvements on time-transfer have a bright prospect in the future applications requiring extremely high-accuracy timing and ranging. 10Excess Commuting in Transitional Urban China:A Case Study of Guangzhou显示文摘During the reform era, Chinese cities witnessed dramatic institutional transformation and spatial restructuring in general and profound change of commuting patterns in particular. Using household surveys collected in Guangzhou, China, in 2001, 2005 and 2010, excess commuting measurements are estimated. Excess commuting shows an overall trend of increasing during 1990–1999, and then declining during 2000–2010. We argue that deepening marketization of the jobs and housing sectors has induced spatial separation of jobs and housing. In other words, institutional transition and urban spatial restructuring are underpinning the changes of commuting patterns in Chinese cities. Excess commuting has strong relationship with individual socio-demographic status, which is by and large due to the increasing flexibilities of jobs and housing location choices enjoyed by urban residents. 11Expression profile of microRNAs in gastrointestinal stromal tumors revealed by high throughput quantitative RT-PCR microarray显示文摘AIM: To investigate the microRNA(miRNA) expression profile in gastrointestinal stromal tumor(GIST) tissues that could serve as a novel diagnostic biomarker for GIST detection.METHODS: We performed a quantitative real-time quantitative reverse transcriptase polymerase chain reaction assay to analyze the expression of 1888 miRNAs in a sample set that included 54 GIST tissue samples.RESULTS: We found that dysregulation of several miRNAs may be related to the malignant potential of GISTs. Six of these miRNAs, hsa-let-7c, miR-218,miR-488#, miR-4683, miR-34c-5p and miR-4773, were selected as the final list of biomarkers to separate the malignant GISTs(M group) from the benign GISTs(B group). In addition, MiR-29b-2#, hsa-let-7c, miR-891 b, miR-218, miR-204, miR-204-3p, miR-628-5p,miR-744, miR-29c#, miR-625 and miR-196 a were used to distinguish between the borderline(BO group) and M groups. 12The Chinese patent medicine, Jin-tang-ning, ameliorates hyperglycemia through improving β cell function in pre-diabetic KKAy mice显示文摘Jin-tang-ning(JTN), a Chinese patent medicine, mainly comprised of Bombyx mori L., has been proved to show α-glucosidase inhibitory efficacy and clinically effective for the treatment of type 2 diabetes(T2 DM). Recently, we have reported that JTN could ameliorate postprandial hyperglycemia and improved β cell function in monosodium glutamate(MSG)-induced obese mice,suggesting that JTN might play a potential role in preventing the conversion of impaired glucose tolerance(IGT) to T2 DM. In this study, we evaluated the effect of JTN on the progression of T2 DM in the pre-diabetic KKAy mice. During the 10 weeks of treatment,blood biochemical analysis and oral glucose tolerance tests were performed to evaluate glucose and lipid profiles. The β cell function was quantified using hyperglycemic clamp at the end of the study. JTN-treated groups exhibited slowly raised fasting and postprandial blood glucose levels, and also ameliorated lipid profile. JTN improved glucose intolerance after 8 weeks of treatment. Meanwhile, JTN restored glucose-stimulated first-phase of insulin secretion and induced higher maximum insulin levels in the hyperglycemic clamp.Thus, to investigate the underlying mechanisms of JTN in protecting β cell function, the morphologic changes of the pancreatic islets were observed by optical microscope and immunofluorescence of hormones(insulin and glucagon). Pancreatic protein expression levels of key factors involving in insulin secretion-related pathway and ER stress were also detected by Western blot. Pre-diabetic KKAy mice exhibited a compensatory augment in β cell mass and abnormal α cell distribution. Long-term treatment of JTN recovered islet morphology accompanied by reducing α cell area in KKAy mice. JTN upregulated expression levels of glucokinase(GCK), pyruvate carboxylase(PCB) and pancreas duodenum homeobox-1(PDX-1), while down-regulating C/EBP homologous protein(Chop)expression in pancreas of the hyperglycemic clamp, which indicated the improvement of mitochondrial metabolism and relief of endoplasmic reticulum(ER) stress of β cells after JTN treatment. 15Effects of rice or wheat residue retention on the quality of milled japonica rice in a rice–wheat rotation system in China显示文摘In rice–wheat rotation systems, crop straw is usually retained in the field at land preparation in every, or every other, season. We conducted a 3-year-6-season experiment in the middle–lower Yangtze River Valley to compare the grain qualities of rice under straw retained after single or double seasons per year. Four treatments were designed as: both wheat and rice straw retained(WR), only rice straw retained(R), only wheat straw retained(W), and no straw retained(CK). The varieties were Yangmai 16 wheat and Wuyunjing 23 japonica rice. The results showed contrasting effects of W and R on rice quality. Amylopectin content, peak viscosity, cool viscosity, and breakdown viscosity of rice grain were significantly increased in W compared to the CK, whereas gelatinization temperature,setback viscosity, and protein content significantly decreased. In addition, the effect of WR on rice grain quality was similar to that of W, although soil fertility was enhanced in WR due to straw being retained in two cycles. The differences in protein and starch contents among the treatments might result from soil nitrogen supply.
