1Epigenetic regulation in B-cell maturation and its dysregulation in autoimmunity显示文摘B cells have a critical role in the initiation and acceleration of autoimmune diseases, especially those mediated byautoantibodies. In the peripheral lymphoid system, mature B cells are activated by self or/and foreign antigens andsignals from helper T cells for differentiating into either memory B cells or antibody-producing plasma cells.Accumulating evidence has shown that epigenetic regulations modulate somatic hypermutation and class switchDNA recombination during B-cell activation and differentiation. Any abnormalities in these complex regulatoryprocesses may contribute to aberrant antibody production, resulting in autoimmune pathogenesis such as systemiclupus erythematosus. Newly generated knowledge from advanced modern technologies such as next-generationsequencing, single-cell sequencing and DNA methylation sequencing has enabled us to better understand B-cellbiology and its role in autoimmune development. Thus this review aims to summarize current research progress inepigenetic modifications contributing to B-cell activation and differentiation, especially under autoimmuneconditions such as lupus, rheumatoid arthritis and type 1 diabetes.Haijing Wu Yaxiong Deng Yu Feng Di Long Kongyang Ma Xiaohui Wang Ming Zhao Liwei Lu Qianjin Lu 2018Cellular & Molecular Immunology2018,15,7:10
2Epigenetic N6-methyladenosine modification of RNA and DNA regulates cancer显示文摘The biological roles of N6 methylation of nucleic acids have been extensively studied.Adenine methylation of RNA is the most prevalent RNA modification and has widespread effects on RNA splicing, translation, localization, and stability.Aberrant dynamic regulation of RNA N6-methyladenosine(m6 A)has been reported in numerous human diseases, including several cancers. In recent years, eukaryotic DNA N6-methyladenosine(6 mA)has also been reported and implicated in cancer progression and tumorigenesis. In this review,we summarize the contributions of N6-methyladenosine modification to cancer biology and pathogenesis in the context of both RNA and DNA.We also highlight the clinical relevance of targeting these modifications as a therapeutic strategy for cancer.Zhixian Liang Reilly LKidwell Haijing Deng Qi Xie 2020Cancer Biology & Medicine2020,17,1:6
3Three new lignan glucosides from the roots of Scutellaria baicalensis显示文摘Three new lignan glucosides, baicalensinosides A–C(1–3), were isolated from the roots of Scutellaria baicalensis. The structural elucidation was achieved by in-depth spectroscopic examinations and qualitative chemical test. Structurally, these compounds belong to the 3,4-dibenzyltetrahydrofurantype lignan glycoside with a mono-hydroxyl substitution at the 70-position of benzylidene group on the numbering system of lignans being one of their shared critical features. The anti-osteoporotic activity of the isolated compounds was assessed in an in vitro osteoprotegerin(OPG) transcriptional activity assay using dual luciferase reporter detection. At 10 μmol/L, compounds 1–3 increased the relative activating ratio of OPG transcription to 1.83, 0.84 and 0.98 times that of the control group, respectively.Hailin Long Haijing Zhang Anjun Deng Lin Ma Lianqiu Wu Zhihong Li Zhihui Zhang Wenjie Wang Jiandong Jiang Hailin Qin 2016Acta Pharmaceutica Sinica B2016,6,3:2
