1α粒子照射诱发细胞存活的旁效应及机理研究显示文摘以人-中国仓鼠卵巢杂交细胞(AL细胞)为靶,通过在照射源与受照射细胞间插入网格对细胞进行定比照射,以及用受照射细胞培养液培养未受照射细胞两种方式,检测未受照射细胞存活率变化,研究α粒了照射细胞时对未受照射细胞存活的影响。通过观察自由基清除剂二甲基亚砜(DMSO)和细胞间通讯阻断剂Lindane对α粒了照射诱发的细胞存活旁效应的抑制作用探讨其可能的机理。研究结果表明,α粒子照射细胞时的未受照射细胞,以及受α粒子照射细胞的培养液培养的未受照射细胞,其存活率均明显低于预期值或正常细胞水平;而DMSO和Lindane均能明显提高细胞存活率。这提示α粒子照射细胞存在旁效应,活性氧和细胞间通讯在α粒子诱发细胞存活旁效应中可能起着重要作用。陆颖 杨陟华 曹珍山 樊飞跃 朱茂祥 Tom K Hei 2003辐射研究与辐射工艺学报2003,21,1:6
2转化生长因子β诱导的基因抑制恶性间皮瘤细胞增殖的体外研究显示文摘目的研究转化生长因子β诱导的基因(transforming growth factor-βinduced gene,TGFBI)在体外是否能抑制恶性间皮瘤细胞NCI-H28的增殖。方法将外源性TGFBI稳定性转染到恶性间皮瘤细胞NCI-H28中,绘制生长曲线图,测定其细胞增殖、接种效率和软琼脂克隆形成,并进行细胞周期的分析。结果外源性TGFBI在恶性间皮瘤细胞NCI-H28中能够稳定地高表达;当外源性的TGFBI转入到肿瘤细胞NCI-H28后,TGFBI高表达的细胞倍增时间增加了4.38倍;与转染空载体的肿瘤细胞相比:NCI-H28的相对接种效率降低了69.68%,外源性TGFBI表达的T28细胞形成了较小的软琼脂克隆;TGFBI可以使肿瘤细胞阻滞在G1期,并延缓肿瘤细胞进入S期的时间。结论 TGFBI可以抑制恶性间皮瘤细胞的体外增殖。曹艳梅 张鹤美 高四海 贺金奖 童建 张增利 Tom K Hei 李冰燕 2013肿瘤防治研究2013,40,12:3
