5Up-regulation of PIK3CA promotes metastasis in gastric carcinoma显示文摘AIM:To explore expressions of PIK3CA in the progression of gastric cancer from primary to metastasis and its effects on activation of phosphatidylinositol 3-kinase(PI3K)/Akt pathway.METHODS:mRNA and protein levels of PIK3CA were assessed,respectively,by real-time quantitative polymerase chain reaction and immunohistochemistry in specimens of normal gastric mucosa,primary foci and lymph node and distant metastasis of gastric cancer.Akt and phosphorylated Akt protein were also examined by Western blotting in these tissues,in order to analyze the effect of PIK3CA expression level changes on the activation of PI3K/Akt signaling pathway.RESULTS:PIK3CA mRNA in lymph node metastasis were approximately 5 and 2 folds higher,respectively,than that in the corresponding normal gastric mucosa and primary gastric cancer tissues(P<0.05),while no statistical significance was found compared with distant metastasis.Immunohistochemically,PIK3CA protein expression was discovered in 7(35%)specimens of 20 primary foci vs 10(67%)of 15 of lymph node metastasis or 11(61%)of 18 of distant metastasis(35%vs 67%,P=0.015;35%vs 61%,P=0.044).With the increased level of PIK3CA expression,the total Akt protein expression remained almost unchanged,but p-Akt protein was upregulated markedly.CONCLUSION:Increased expression of PIK3CA is expected to be a promising indicator of metastasis in gastric cancer.Up-regulation of PIK3CA may promote the metastasis of gastric cancer through aberrant activation of PI3K/Akt signaling.Liu JF Zhou XK Chen JH Yi G Chen HG Ba MC Lin SQ Qi YC. 2010World Journal of Gastroenterology2010,16,39:20
6Can acupuncture therapy reduce preoperative anxiety?A systematic review and meta-analysis显示文摘Background:Acupuncture therapy has shown promise for effectively relieving preoperative anxiety.Nevertheless,previous findings from randomized controlled trials(RCTs)are inconsistent and must be examined in detail.Objective:This study systematically evaluates the efficacy and safety of acupuncture therapy for preoperative anxiety as well as the quality of evidence supporting this application.Search strategy:The China National Knowledge Infrastructure Database,Wanfang Data Journal Database,Chinese Biomedical Literature Database,Chongqing VIP,Embase,Pub Med and Cochrane Library Databases were queried from their inception to 19,February 2020,using keywords such as“acupuncture therapy,”“preoperative”and“anxioty.”Manual searches expanded the search breadth and included conference abstracts and other reference lists.Inclusion criteria:RCTs were included in the current study if they contained a comparison between a group of anxiety patients that received acupuncture therapy and a control group that received sham acupuncture.Data extraction and analysis:Literature was reviewed,and various articles were selected using the Note Express 3.2.0 software.Two researchers independently screened and extracted data and evaluated the risk of bias in the included studies.The Rev Man 5.3 software was used for data aggregation and the Grades of Recommendation,Assessment,Development and Evaluation(GRADE)assessment was used to evaluate the quality of the study outcomes.Results:Twelve studies were included in the review,containing a total of 916 patients.Meta-analysis showed that,compared with the control group,patients who received acupuncture therapy had reduced State-Trait Anxiety Inventory Scale(STAI-S)score(mean difference[MD]=–9.07,95%confidence interval[CI][–13.19 to–4.96],P<0.0001)and Visual Analogue Scale(VAS)score(MD=–1.37,95%CI[–2.29 to–0.45],P=0.003).However,for the Hamilton Anxiety Scale(HAMA)score,there was no difference between the two groups(MD=–3.98,95%CI[–12.89 to 4.92],P=0.38).Further,the GRADE assessment demonstrated that the STAI-S was of moderate quality,the VAS of low quality and the HAMA of very low quality.Conclusion:Acupuncture therapy may be able to decrease anxiety in preoperative patients,but the results need to be further verified due to the small sample sizes and the low quality of evidence to date.Qiu-yu Tong Ran Liu Kun Zhang Yuan G ao Guang-wei Cui Wei-dong Shen 2021Journal of Integrative Medicine2021,19,1:15
7The herbal compound geniposide rescues formaldehyde-induced apoptosis in N2a neuroblastoma cells显示文摘The herbal medicine Tong Luo Jiu Nao(TLJN)contains geniposide(GP)and ginsenoside Rg1 at a molar ratio of 10:1.Rg1 is the major component of another herbal medicine,panax notoginseng saponin(PNS).TLJN has been shown to strengthen brain function in humans,and in animals it improves learning and memory.We have previously shown that TLJN reduces amyloidogenic processing in Alzheimer’s disease(AD)mouse models.Together this suggests TLJN may be a potential treatment for patients with dementia.Because chronic damage of the central nervous system by formaldehyde(FA)has been presented as a risk factor for age-associated cognitive dysfunction,in the present study we investigated the protective effect of both TLJN and GP in neuron-like cells exposed to FA.FA-exposed murine N2a neuroblastoma cells were incubated with TLJN,its main ingredient GP,as well as PNS,to measure cell viability and morphology,the rate of apoptosis and expression of genes encoding Akt,FOXO3,Bcl2 and p53.The CCK-8 assay,cytoskeletal staining and flow cytometry were used to test cell viability,morphology and apoptosis,respectively.Fluorescent quantitative real-time PCR(qRT-PCR)was used to monitor changes in gene expression,and HPLC to determine the rate of FA clearance.Treatment of N2a cells with 0.09 mmol L?1 FA for 24 h significantly reduced cell viability,changed cell morphology and promoted apoptosis.Both TLJN and GP conferred neuroprotection to FA-treated N2a cells,whereas PNS,which had to be used at lower concentrations because of its toxicity,did not.Our data demonstrate that TLJN can rescue neuronal damage caused by FA and that its main ingredient,GP,has a major role in this efficacy.This presents purified GP as a drug or lead compound for the treatment of AD.CHEN JinYan SUN MengRu WANG XingHua LU Jing WEI Yan TAN Yan LIU Ying GTZ Jürgen HE RongQiao HUA Qian 2014Science China(Life Sciences)2014,57,4:15

11Ent-11α-Hydroxy-15-oxo-kaur-16-en-19-oic-acid Inhibits Growth of Human Lung Cancer A549 Cells by Arresting Cell Cycle and Triggering Apoptosis显示文摘Objective: To examine the apoptotic effect of ent-11α-hydroxy-15-oxo-kaur-16-en-19-oic-acid (5F), a compound isolated from Pteris semipinnata L (PsL), in human lung cancer A549 cells. Methods: A549 cells were treated with 5F (0-80 μg/ml) for different time periods. Cytotoxicity was examined using a MTT method. Cell cycle was examined using propidium iodide staining. Apoptosis was examined using Hoechst 33258 staining, enzyme-linked immunosorbent assay (ELISA) and caspase-3 activity analysis. Expression of representative apoptosis-related proteins was evaluated by Western blot analysis. Reactive oxygen species (ROS) level was measured using standard protocols. Potential interaction of 5F with cisplatin was also examined. Results: 5F inhibited the proliferation of A549 cells in a concentration-and time-dependent manner. 5F increased the accumulation of cells in sub-G1 phase and arrested the cells in the G2 phase. Exposure to 5F induced morphological changes and DNA fragmentation that are characteristic of apoptosis. The expression of p21 was increased. 5F exposure also increased Bax expression, release of cytochrome c and apoptosis inducing factor (AIF), and activation of caspase-3. 5F significantly sensitized the cells to cisplatin toxicity. Interestingly, treatment with 5F did not increase ROS, but reduced ROS production induced by cisplatin. 12Spatial-temporal dynamics of desert vegetation and its responses to climatic variations over the last three decades:a case study of Hexi region in Northwest China显示文摘Analysis of spatial-temporal variations of desert vegetation under the background of climate changes can provide references for ecological restoration in arid and semi-arid areas.In this study,we used the Global Inventory Modeling and Mapping Studies(GIMMS)NDVI data from 1982 to 2006 and Moderate Resolution Imaging Spectroradiometer(MODIS)NDVI data from 2000 to 2013 to reveal the dynamics of desert vegetation in Hexi region of Northwest China over the past three decades.We also used the annual temperature and precipitation data acquired from the Chinese meteorological stations to analyze the response of desert vegetation to climatic variations.The average value of NDVImax(the maximum NDVI during the growing season)for desert vegetation in Hexi region increased at the rate of 0.65×10–3/a(P<0.05)from 1982 to 2013,and the significant increases of NDVImax mainly appeared in the typical desert vegetation areas.Vegetation was significantly improved in the lower reaches of Shule and Shiyang river basins,and the weighted mean center of desert vegetation mainly shifted toward the lower reaches of the two basins.Almost 95.32% of the total desert vegetation area showed positive correlation between NDVImax and annual precipitation,indicating that precipitation is the key factor for desert vegetation growth in the entire study area.Moreover,the areas with non-significant positive correlation between NDVImax and annual precipitation mainly located in the lower reaches of Shiyang and Shule river basins,this may be due to human activities.Only 7.64% of the desert vegetation showed significant positive correlation between NDVImax and annual precipitation in the Shule River Basin(an extremely arid area),indicating that precipitation is not the most important factor for vegetation growth in this basin,and further studies are needed to investigate the mechanism for this phenomenon.YANG Xuemei LIU Shizeng YANG Taibao XU Xianying KANG Caizhou TANG Jinnian WEI Huaidong Mihretab G GHEBREZGABHER LI Zhiqi 2016Journal of Arid Land2016,8,4:10

16High-throughput screening of mouse gene knockouts identifies established and novel skeletal phenotypes显示文摘Screening gene function in vivo is a powerful approach to discover novel drug targets. We present high-throughput screening(HTS) data for 3 762 distinct global gene knockout(KO) mouse lines with viable adult homozygous mice generated using either gene-trap or homologous recombination technologies. Bone mass was determined from DEXA scans of male and female mice at 14 weeks of age and by microCT analyses of bones from male mice at 16 weeks of age. Wild-type(WT) cagemates/littermates were examined for each gene KO. Lethality was observed in an additional 850 KO lines. Since primary HTS are susceptible to false positive findings, additional cohorts of mice from KO lines with intriguing HTS bone data were examined. Aging,ovariectomy, histomorphometry and bone strength studies were performed and possible non-skeletal phenotypes were explored.
