1Ablation of Akt2 and AMPKα2 rescues high fat diet-induced obesity and hepatic steatosis through Parkin-mediated mitophagy显示文摘Given the opposing effects of Akt and AMP-activated protein kinase(AMPK)on metabolic homeostasis,this study examined the effects of deletion of Akt2 and AMPKα2 on fat diet-induced hepatic steatosis.Akt2–Ampkα2 double knockout(DKO)mice were placed on high fat diet for 5 months.Glucose metabolism,energy homeostasis,cardiac function,lipid accumulation,and hepatic steatosis were examined.DKO mice were lean without anthropometric defects.High fat intake led to adiposity and decreased respiratory exchange ratio(RER)in wild-type(WT)mice,which were ablated in DKO but not Akt2^(-/-) and Ampkα2^(-/-) mice.High fat intake increased blood and hepatic triglycerides and cholesterol,promoted hepatic steatosis and injury in WT mice.These effects were eliminated in DKO but not Akt2^(-/-) and Ampkα2^(-/-) mice.Fat diet promoted fat accumulation,and enlarged adipocyte size,the effect was negated in DKO mice.Fat intake elevated fatty acid synthase(FAS),carbohydrate-responsive element-binding protein(CHREBP),sterol regulatory element-binding protein 1(SREBP1),peroxisome proliferator-activated receptor gamma coactivator 1-alpha(PGC-1α),peroxisome proliferator-activated receptor-α(PPARα),PPARγ,stearoyl-CoA desaturase 1(SCD-1),phosphoenolpyruvate carboxykinase(PEPCK),glucose 6-phosphatase(G6Pase),and diglyceride O-acyltransferase 1(DGAT1),the effect was absent in DKO but not Akt2^(-/-) and Ampkα2^(-/-) mice.Fat diet dampened mitophagy,promoted inflammation and phosphorylation of forkhead box protein O1(FoxO1)and AMPKα1(Ser^(485)),the effects were eradicated by DKO.Deletion of Parkin effectively nullified DKO-induced metabolic benefits against high fat intake.Liver samples from obese humans displayed lowered microtubule-associated proteins 1A/1B light chain 3B(LC3B),Pink1,Parkin,as well as enhanced phosphorylation of Akt,AMPK(Ser^(485)),and FoxO1,which were consolidated by RNA sequencing(RNAseq)and mass spectrometry analyses from rodent and human livers.These data suggest that concurrent deletion of Akt2 and AMPKα2 offers resilience to fat diet-induced obesity and hepatic steatosis,possibly through preservation of Parkin-mediated mitophagy and lipid metabolism.Shuyi Wang Jun Tao Huaguo Chen Machender R.Kandadi Mingming Sun Haixia Xu Gary D.Lopaschuk Yan Lu Junmeng Zheng Hu Peng Jun Ren 2021Acta Pharmaceutica Sinica B2021,11,11:4
2Analysis of genes differentially expressed during initial cellular dedifferentiation in cotton显示文摘The early phase of phytohormone induction is a vital stage of somatic embryogenesis. This phase includes a key process for acquiring cellular totipotency through cellular dedifferentiation. To unravel the molecular mechanism of cellular dedifferentiation in cotton, we constructed a cDNA library using the suppression subtractive hybridization method. A total of 286 differential cDNA clones were sequenced and identified. Among these clones, 112 unique ESTs were significantly up-regulated during the early phase of phytohormone induction, and 40.2% of the ESTs were first identified. GST was highly expressed from 6 to 24 h after induction with phytohormone treatment. PRPs were predominantly expressed and exhibited distinct expression patterns in different treatments, suggesting that they are closely related to cellular dedifferentiation in cotton. Putative GhSAMS, GhSAMDC, GhSAHH and GhACO3 involvement in SAM metabolism was identified in this library. The analysis of qRT-PCR showed that two remarkable increased expressions of the four SAM-related genes happened during the early phase of phytohormone induction, and that a highly positive correlation existed between GhSAMS and GhSAHH. The highest expression level of GhSAMS might be associated with its reentry into the cell cycle. The histological observations further showed that some cells accomplished cellular dedifferentiation and division within 72 h in 2,4-D treatment, and that cellular dedifferentiation might be regulated through two alterations in SAM-dependent transmethylation activity in cotton. 3Optimized Mid-bond Order for 3D-Stacked ICs Considering Failed Bonding显示文摘One notable difference between 3D test flow and 2D test flow mainly lies in the mid-bond test, in which the stacking yield can be further enhanced through optimized bonding arrangement. In contrast to the existing sequential stacking, this paper proposes a novel rearranged stacking scheme which estimates the probability and cost of failed bonding in each stacking step and optimizes the mid-bond order to screen out the failed component as early as possible. The effect of the rearranged stacking has been extensively analyzed using the yield model and cost model of 3D-SICs considering different process parameters such as die yield, stacking size, failure rate and redundancy degree of TSVs. 4Identification of African swine fever virus MGF505-2R as a potent inhibitor of innate immunity in vitro显示文摘African swine fever(ASF)is etiologically an acute,highly contagious and hemorrhagic disease caused by African swine fever virus(ASFV).Due to its genetic variation and phenotypic diversity,until now,no efficient commercial vaccines or therapeutic options are available.The ASFV genome contains a conserved middle region and two flexible ends that code for five multigene families(MGFs),while the biological functions of the MGFs are not fully characterized.Here,ASFV MGF505-2R-deficient mutant ASFV-Δ2R was constructed based on a highly virulent genotype II field isolate ASFV CN/GS/2018 currently circulating in China.Transcriptomic profiling demonstrated that ASFV-Δ2R was capable of inducing a larger number of differentially expressed genes(DEGs)compared with ASFV CN/GS/2018.Hierarchical clustering of up-regulated DEGs revealed that ASFV-Δ2R induced the most dramatic expression of interferon-related genes and inflammatory and innate immune genes,as further validated by RT-qPCR.The GO and KEGG pathway analysis identified significantly enriched pathways involved in pathogen recognition and innate antiviral immunity.Conversely,pharmacological activation of those antiviral immune responses by exogenous cytokines,including type I/II IFNs,TNF-αand IL-1β,exerted combinatory effects and synergized in antiviral capacity against ASFV replication.Collectively,MGF505-2R is a newly identified inhibitor of innate immunity potentially implicated in immune evasion.Huaguo Huang Wen Dang Zhengwang Shi Mingyang Ding Fan Xu Tao Li Tao Feng Haixue Zheng Shuqi Xiao 2023Virologica Sinica2023,38,1:0

7Influences of different alkaline and acidic diagenetic environments on diagenetic evolution and reservoir quality of alkaline lake shales显示文摘Thin section and argon-ion polishing scanning electron microscope observations were used to analyze the sedimentary and diagenetic environments and main diagenesis of the Permian Fengcheng Formation shales in different depositional zones of Mahu Sag in the Junggar Basin,and to reconstruct their differential diagenetic evolutional processes.The diagenetic environment of shales in the lake-central zone kept alkaline,which mainly underwent the early stage(Ro<0.5%)dominated by the authigenesis of Na-carbonates and K-feldspar and the late stage(Ro>0.5%)dominated by the replacement of Na-carbonates by reedmergnerite.The shales from the marginal zone underwent a transition from weak alkaline to acidic diagenetic environments,with the early stage dominated by the authigenesis of Mg-bearing clay and silica and the late stage dominated by the dissolution of feldspar and carbonate minerals.The shales from the transitional zone also underwent a transition from an early alkaline diagenetic environment,evidenced by the formation of dolomite and zeolite,to a late acidic diagenetic environment,represented by the reedmergnerite replacement and silicification of feldspar and carbonate minerals.The differences in formation of authigenic minerals during early diagenetic stage determine the fracability of shales.The differences in dissolution of minerals during late diagenetic stage control the content of free shale oil.Dolomitic shale in the transitional zone and siltstone in the marginal zone have relatively high content of free shale oil and strong fracability,and are favorable“sweet spots”for shale oil exploitation and development.LI Changzhi GUO Pei XU Jinghong ZHONG Kai WEN Huaguo 2024Petroleum Exploration and Development2024,51,1:0
8Analysis of ship wake features and extraction of ship motion parameters from SAR images in the Yellow Sea显示文摘The identifying features of ship wakes in synthetic aperture radar (SAR) remote sensing images are of great importance for detecting ships and for extracting ship motion parameters. A statistical analysis was conducted on the identifying features of ship wakes in SAR images in the Yellow Sea. In this study, 1091 ship wake sub-images were selected from 327 SAR images in the Yellow Sea near Qingdao. Analysis of the identifying features of ship wakes in SAR images revealed that both turbulent wakes and Kelvin wakes account for the majority of ship wakes, with turbulent wakes occurring approximately four times as frequently as Kelvin wakes. Narrow-V wakes and internal wave wakes were comparatively rare, which is due to the peculiarities of the radar system parameters and marine environments required to observe these wakes. Additionally, we extracted ship motion parameters from four types of ship wakes in the SAR images. Specifically, internal wave wakes in SAR images in the Yellow Sea were also used to extract ship motion parameters. Validation of the extracted parameters indicated that the extraction of these parameters from ship wakes is a viable and accurate approach for the acquisition of ship motion parameters. 11A comprehensive construction of the domain ontology for stratigraphy显示文摘Stratigraphic knowledge,the cornerstone of geoscience,needs to be represented by the Knowledge Graph based upon ontology,in order to apply the state-of-the-art big-data techniques.This study aims to comprehensively construct the ontologies for the stratigraphic domain.This has been achieved by a federated,crowd intelligence-based collaboration among domain experts of major stratigraphic subdisciplines.The initial step is to enumerate key terms from authoritative references and incorporate them into the Geoscience Professional Knowledge Graphs(GPKGs)of Deep-time Digital Earth Project.During this process,semantic heterogeneities were meticulously addressed by professional judgement aided by an automatic detection of Homonyms at the GPKGs platform.Afterwards,these terms were further differentiated as either classes or properties and arranged in a hierarchical framework in a top-down process.Consequently,seven ontologies are constructed for major stratigraphic branches,i.e.,Lithostratigraphy,Biostratigraphy,Chronostratigraphy,Chemostratigraphy,Magnetostratigraphy,Cyclostratigraphy and Sequence Stratigraphy.The ontology of Biostratigraphy,among them,is elaborated here,as no biostratigraphic ontology has been attempted before to our knowledge.The constructed biostratigraphic ontology comprises following major root classes:Fossil,Biostratigraphic unit,Biostratigraphic horizon.Altogether,they contribute to the eventual dating and correlating of strata in another root class:Biostratigraphic correlation.In summary,the achievements of this study are probably heretofore the most comprehensive ontologies for the stratigraphic domain.Moreover,a proto model of semantic search engine was conceived to discuss potential application of our work for better querying stratigraphic references,utilizing the semantic liaison of the classes in the constructed ontologies.Huiqing Xu Yingying Zhao Hao Huang Shaochun Dong Yukun Shi Chunju Huang Huaichun Wu Zhiqi Qian Qiang Fang Huaguo Wen Zhongtang Su Shuang Dai Ronghua Wang Chao Li Chao Sun Junxuan Fan 2023Geoscience Frontiers2023,14,5:0
