1A Simplified PCR-SSP Method for HLA-A2 Subtype in a Population of Wuhan,China显示文摘HLA-A2 is the most frequent HLA-A allele in all ethnic populations, and an important restriction element for peptide presentation to T cells in infectious disease and cancer. However, the HLA-A2 supertype consisting of up to 75 subtypes, mutation studies and analyses using cytotoxic T lymphocytes suggest the functional relevance of subtype-specific differences in HLA-A2 molecules for peptide binding and T-cell recognition. Therefore, it is necessary for T-cell response study to discriminate the HLA-A2 subtypes and to understand the profile of HLA-A2 allelic distribution in a given population. In this study, we developed a simple, robust approach based on the nested polymerase chain reaction using sequence-specific primers (PCR-SSP) to discriminate 17 HLA-A2 subtypes which cover the most HLA-A2 alleles (> 99% allele frequency) reported in Chinese, using 15 combinations of 19 allelic specific primers. In the first round of PCR, 3 combinations of 5 primers were used to determine whether the tested sample was HLA-A2 positive, meanwhile the subtypes of HLA-A*0209 and HLA-A*0215N were determined for the variant position of these two subtypes is in exon 4 instead of exon 2, 3. Samples of HLA-A2 positive were subtyped in the second round of PCR, using PCR products of the first round as templates. This strategy was applied to test the samples of 78 random HLA-A2 positive individuals for their HLA-A2 subtypes. Those samples were screened for HLA-A2 positive by the first round PCR-SSP from 154 healthy blood donors in Wuhan, China. The subtyping results were verified by using flow cytometric analysis (FCM) with HLA-A2 specific monoclonal antibody BB7.2 and DNA sequencing. The typing results of the samples show 50.7% random individuals in the population carry HLA-A2, HLA-A*0201 ranks the first (allele frequency = 15.5%), followed by A*0207 (5.8%), A*0206 (4.7%), A*0203 (2.6%), A*0210 (0.7%), and these 5 alleles account for 99.0% HLA-A2 subtypes of allele frequency. 3Effect of monovalent anions on cationic Gemini micro-emulsion显示文摘Micro-emulsion usually consists of water, oil, surfactants and co-surfactants, and each component has an effect on the phase behavior and solubilization of the micro-emulsion. When the surfactant in the micro-emulsion system is quaternary ammonium cationic Gemini surfactant, the surfactant mainly combines with the anions in the salt. With the increase of salt concentration, the phase transformation of Winsor I → Winsor III → Winsor II occurred, but the optimum salinity and salt width are different because of the type of salt. The effects of 5 different kinds of monovalent anions, including C_6H_5SO_3^-, I-, Br-, NO_3^- and Cl-, on the phase behavior and solubilization of quaternary ammonium cationic Gemini micro-emulsion are researched by Winsor phase diagram. It is found that the effects of organic anions C_6H_5SO_3-and I-on the phase behavior and solubilization of quaternary ammonium cationic Gemini micro-emulsion are most significant, and the effects of Br-, NO_3^- and Cl-are less significant. 4Distribution of aluminum species and the characteristics of structure of poly-aluminum-chloride-sulfate(PACS)显示文摘A series of poly\|aluminum\|chloride\|sulfate (PACS), which has different basicities (γ) and Al 3+ /SO 2- 4 molar ratio, has been prepared and dried at 105℃ and 65℃, respectively. The distribution of aluminum species of PACS was examined, and the effect of γ value, Al 3+ /SO 2- 4 molar ratio, dilution on the distribution of aluminum species of PACS was also investigated by using Al\|ferron timed complex colorimetric method. The IR spectroscopy and X \|ray diffraction were used to study the effect of γ value, Al 3+ /SO 2- 4 molar ratio and the drying temperature on the structure of PACS. The experimental results show that Al 3+ /SO 2- 4 molar ratio has a great effect on the distribution of aluminum species, but the dilution has a little effect on the distribution of aluminum species. The lower the Al 3+ /SO 2- 4 molar ratio, the higher the proportions of the polymer and colloidal species in PACS. The polymeric degree of PACS was related to γ value and Al 3+ /SO 2- 4 molar ratio. 5A Novel Intrusion Detection Method Based on Improved SVM by Combining PCA and PSO显示文摘The paper presents an improved support vector machine (SVM) by combining principal component analysis (PCA) and particle swarm optimization (PSO).Then,the improved SVM is applied to the intrusion detection system (IDS) to improve the detection rate.First,PCA is used to reduce the dimension of feature vectors.Second,we use the PSO algorithm to optimize the punishment factor C and kernel parameters in SVM.The experimental results indicate that the intrusion detection rate (97.752 8%) of improved SVM by combining PCA and PSO is higher than those (95.635 5%) of PSO-SVM and those (90.476 2%) of standard SVM with KDD Cup 1999 data set.WANG Hui ZHANG Guilingi E Mingjie SUN Na 2011Wuhan University Journal of Natural Sciences2011,16,5:5
6A comparison of surgical efficacy between a 1.8-mm microincision and 3.2-mm and 5.5-mm incisions for phacoemulsification显示文摘Phacoemulsification is a commonly used surgical method in cataract surgery. This paper observes and compares the surgical efficacy of three incisions of different length for phacoemulsification to identify the optimal method for cataract surgery. Ninety patients were enrolled in the present study and divided into three groups. The 1.8-mm group received Bausch & Lomb MI60 foldable intraocular lens(IOL) implantation(n=30), 3.2-mm group received Bausch & Lomb Akreos AO foldable lens implantation(n=30), and 5.5-mm group received Alcon TYPE 05 rigid IOL implantation(n=30). Visual acuity, Oculyzer-based anterior segment analysis, and corneal endothelial cell count before surgery, and 3, 7, 30, and 90 d after surgery were recorded and compared. Pseudophakic accommodation three days, one week, one month, and three months after surgery was determined. Intraoperative ultrasound time and ultrasonic energy were recorded. 8Celiac disease serology in patients with different pretest probabilities: Is biopsy avoidable?显示文摘AIM: To establish the diagnostic performance of sev-eral serological tests, individually and in combination, for diagnosing celiac disease (CD) in patients with different pretest probabilities, and to explore potential se- rological algorithms to reduce the necessity for biopsy. METHODS: We prospectively performed duodenal biopsy and serology in 679 adults who had either high risk (n = 161) or low risk (n = 518) for CD. Blood samples were tested using six assays (enzyme-linked immunosorbent assay) that detected antibodies to tissue transglutaminase (tTG) and deamidated gliadin peptide (DGP). RESULTS: CD prevalence was 39.1% in the high-risk population and 3.3% in the low-risk group. In high-risk patients, all individual assays had a high diagnostic efficacy [area under receiving operator characteristic curves (AU ROC): 0.968 to 0.999]. In contrast, assays had a lower diagnostic efficacy (AU ROC: 0.835 to 0.972) in the low-risk group. Using assay combinations, it would be possible to reach or rule out diagnosis of CD without biopsy in 92% of cases in both pretest populations. We observed that the new DGP/tTG Screen assay resulted in a surplus compared to more conventional assays in any clinical situation. CONCLUSION: The DGP/tTG Screen assay could be considered as the best initial test for CD. 9OsbZIP76 interacts with OsNF-YBs and regulates endosperm cellularization in rice (Oryza sativa)显示文摘Following double fertilization,plant endosperm nuclei undergo syncytial divisions,followed by synchro-nous cellularization.Cellularization is a key event during endosperm development,but our understanding of its regulation is limited to Arabidopsis.In this study we show that OsbZIP76 regulates cellularization in rice(Oryza sativa).Activation of OsbZIP76 coincided with the initiation of cellularization,and its knockdown or knockout mutants exhibited precocious cellularization.Genes involved in en-dosperm development or starch biosynthesis were pre-maturely activated in the osbzip76 caryopsis.As a putative transcription factor,OsbZIP76 alone lacked transcriptional activation activity;however,it interacted with the nuclear factor Y(NF-Y)family transcription factors OsNF-YB9 and OsNF-YB1 in yeast and in planta.OsbZIP76 and OsNF-YB9 were predominantly expressed in the endosperm and the proteins colocalized.Seeds of osnf-yb1 and osbzip76 mu-tants showed reduced size and reduced apparent amylose content.The parent-of-origin-dependent expression of OsbZIP76 is variable in different rice accessions.In sum-mary,OsbZIP76 is an endosperm-expressed imprinted gene that regulates endosperm development in rice.Baixiao Niu Hui Deng Tingting Li Sandeep Sharma Qianbin Yun Qianru Li Zhiguo E Chen Chen 2020Journal of Integrative Plant Biology2020,62,12:4
10Development and calibration of the Moon-based EUV camera for Chang'e-3显示文摘The process of development and calibration for the first Moon-based extreme ultraviolet(EUV) camera to observe Earth's plasmasphere is introduced and the design, test and calibration results are presented. The EUV camera is composed of a multilayer film mirror, a thin film filter, a photon-counting imaging detector, a mechanism that can adjust the direction in two dimensions, a protective cover, an electronic unit and a thermal control unit. The center wavelength of the EUV camera is 30.2 nm with a bandwidth of 4.6 nm. The field of view is 14.7° with an angular resolution of 0.08°, and the sensitivity of the camera is 0.11 count s-1Rayleigh-1. 13Simulating global soil-CO_2 flux and its response to climate change显示文摘It has been argued that increased soil respiration would become a major atmospheric source of CO\-2 in the event of global warming. The simple statistical models were developed based on a georeferenced database with 0 5°×0 5° longitude/latitude resolution to simulate global soil\|CO\-2 fluxes, to investigate climatic effects on these fluxes using sensitivity experiments, and to assess possible responses of soil\|CO\-2 fluxes to various climate change scenarios. The statistical models yield a value of 69 PgC/a of global soil\|CO\-2 fluxes for current condition. Sensitivity experiments confirm that the fluxes are responsive to changes in temperature, precipitation and actual evapotranspiration, but increases in temperature and actual evapotranspiration affect soil\|CO 2 fluxes more than increases in precipitation. Using climatic change projections from four global circulation models, each corresponding to an equilibrium doubling of CO 2, it can be found that the largest increases in soil\|CO 2 fluxes were associated with the boreal and tundra regions. The globally averaged soil\|CO 2 fluxes were estimated to increase by about 35% above current values, providing a positive feedback to the greenhouse effect.PENG Chang\|hui 1* , Michael J. 14Experimental study on dispersive soil in western Jilin显示文摘In order to research engineering geological properties of the soil in Zhenlai of western Jilin,especially the dispersivity of soil,the authors carried out the basic physicochemical test and dispersivity identification test.The results show that the dispersivity of the soil increases with the increase of depth within 0-30 cm (surface soil);it decreases as the depth increases within in 30-100 cm.Furthermore,the statistical analysis of the dispersivity indexes and physicochemical propertity indexes show that the DP is positively linear correlated with total soluble salt content,sodium ion content,ESP,pH and organic matter content.Meanwhile,it is negatively linear correlated with clay content,and the linear relationship is better.Through the study of the dispersion mechanism of soil samples,it can be concluded that sodium montmorillonite,higher percentage of exchangeable sodium and high pH are the main reasons for the dispersion of soils in western Jilin.ZHANG Jing WANG Qing WANG Wenhua CHEN Hui'e WANG Ying 2010Global Geology2010,13,1:2
15Effect of endothelial progenitor cells derived from human umbilical cord blood on oxygen-induced retinopathy in mice by intravitreal transplantation显示文摘·AIM: To investigate the effect of endothelial progenitor cells(EPCs) labeled by carboxy fluorescein diacetate succinimidyl ester(CFSE) on murine oxygen-induced retinopathy(OIR) by intravitreal transplantation.·METHODS: After isolated from human umbilical cord blood mononuclear cells, EPCs were cultivated and then labeled with CFSE in vitro. C57BL/6J mice were placed to75% hyperoxia chamber from P7 to P12 to establish OIR model. At P12, OIR mice were intravitreally injected with1 μL suspension contained 2×105EPCs(EPCs group) or isometric phosphate buffered saline(PBS group). The contralateral eye of each mice received no injection(OIR group). Evans blue angiography and frozen section were examined to track the labeled cells in OIR group at P15 and P19. Using retina paraffin sections and adenosinediphos phatase staining at P12 and P19, the effect of EPCs on OIR mice was evaluated quantitatively and qualitatively.· RESULTS: The retinas from EPCs group with less non-perfusion area and fewer peripheral tufts wereobserved at P19, comparing with that from PBS or OIR group. The retinopathy in EPCs group receded earlier with less non-ganglion cells and neovascular nuclei,together with relatively regular distribution. The counts of the neovascular nuclei at P19 were reduced by 44% or45%, compared with those of OIR group or PBS group respectively. Three days after EPCs injection, a large number of EPCs appeared in the vitreous cavity and adhered to the retinal surface.
