| 1 | Platelet thromboxane(11-dehydro-Thromboxane B_2) and aspirin response in patients with diabetes and coronary artery disease显示文摘Aspirin(ASA) irreversibly inhibits platelet cyclooxygenase-1(COX-1) leading to decreased thromboxane-mediated platelet activation. The effect of ASA ingestion on thromboxane generation was evaluated in patients with diabetes(DM) and cardiovascular disease. Thromboxane inhibition was assessed by measuring the urinary excretion of 11-dehydro-thromboxane B2(11dhTxB2), a stable metabolite of thromboxane A2. The mean baseline urinary 11dhTxB2 of DM was 69.6% higher than healthy controls(P = 0.024): female subjects(DM and controls) had 50.9% higher baseline 11dhTxB2 than males(P = 0.0004), while age or disease duration had no influence. Daily ASA ingestion inhibited urinary 11dhTxB2 in both DM(71.7%) and controls(75.1%, P < 0.0001). Using a pre-established cut-off of 1500 pg/mg of urinary 11dhTxB2, there were twice as many ASA poor responders(ASA 'resistant') in DM than in controls(14.8% and 8.4%, respectively). The rate of ASA poor responders in two populations of acute coronary syndrome(ACS) patients was 28.6 and 28.7%, in spite of a significant(81.6%) inhibition of urinary 11dhTxB2(P < 0.0001). Both baseline 11dhTxB2 levels and rate of poor ASA responders were significantly higher in DM and ACS compared to controls. | 5 | A single synonymous mutation determines the phosphorylation and stability of the nascent protein显示文摘p53 is an intrinsically disordered protein with a large number of post-translational modifications and interacting partners.The hierarchical order and subcellular location of these events are still poorly understood.The activation of p53 during the DNA damage response(DDR)requires a switch in the activity of the E3 ubiquitin ligase MDM2 from a negative to a positive regulator of p53.This is mediated by the ATM kinase that regulates the binding of MDM2 to the p53 mRNA facilitating an increase in p53 synthesis.Here we show that the binding of MDM2 to the p53 mRNA brings ATM to the p53 polysome where it phosphorylates the nascent p53 at serine 15 and prevents MDM2-mediated degradation of p53.A single synonymous mutation in p53 codon 22(L22L)prevents the phosphorylation of the nascent p53 protein and the stabilization of p53 following genotoxic stress.The ATM trafficking from the nucleus to the p53 polysome is mediated by MDM2,which requires its interaction with the ribosomal proteins RPL5 and RPL11.These results show how the ATM kinase phosphorylates the p53 protein while it is bang synthesized and offer a novel mechanism whereby a single synonymous mutation controls the stability and activity of the encoded protein. | Konstantinos Karakostis Sivakumar Vadivel Gnanasundram Ignacio Lopez Aikaterini Thermou Lixiao Wang Karin Nylander Vanesa Ollvares-Iliana Robin Fahraeus | 2019 | Journal of Molecular Cell Biology2019,11,3: | 1 |

| 11 | Assessing stakeholders'perceptions and values towards social-ecological systems using participatory methods显示文摘Introduction:Many conservation initiatives for managing social-ecological landscapes factor in the best available biophysical information.However,insufficient consideration of social aspects can render such initiatives ineffective.By incorporating stakeholders'perceptions and values,and by involving stakeholders(transparently)in decision-making processes,conservation plans and efforts can better achieve desired goals and targets.Results:In this paper,we present and analyze three case studies within different ecological and land use contexts.Each case study uses different participatory approaches to elicit stakeholders'perceptions and values,while seeking to involve stakeholders in the decision-making process.The case studies are the following:(1)participatory scenario planning for a protected area in Doñana coastal wetland in Southwestern Spain;(2)assessment of ecosystem services trade-offs and social responses on rewards for agro-biodiversity in the rubber agroforest landscape in Jambi,Indonesia;and(3)socio-cultural evaluation,through visual stimuli,of ecosystem services provided by transhumance cultural landscapes in Central Spain.We discuss how stakeholders perceive and value their environments and to what extent participatory approaches are useful for capturing information relating to land use and ecological processes.Conclusions:Social-ecological systems are inherently complex,having a variety of interacting actors with different types of ecological knowledge,interests,and values.Different participatory tools or approaches are appropriate for various and specific contexts and objectives.Determining and integrating the various types of knowledge and values of different actors can contribute to a more comprehensive understanding of social-ecological systems. | Grace B Villamor Ignacio Palomo Cesar A Lopez Santiago Elisa Oteros-Rozas Joe Hill | 2014 | Ecological Processes2014,3,1: | 0 |
