| 1 | 呼吸道纤毛运动状况的高速数字化显微成像显示文摘目的 通过对呼吸道纤毛摆动的频率和形式进行精确测量,以建立客观评价呼吸道上皮纤毛传输功能的指标。方法 采用高速数字化显微成像技术,对体外培养的兔和鼠气管黏膜以及人鼻腔黏膜的纤毛摆动状况进行观察。结果 ①数字化图像中测量点灰度的变化反映纤毛运动的状况,通过分析图像灰度的变化过程,可评价纤毛运动周期中各个时段的特征变化;②图像采集系统的采样速率为240帧/s时,可清晰显示纤毛运动状况,而当采样速率下降为30帧/s时,则无法获得正确的纤毛摆动频率(ciliary beat frequency,CBF)数据;③30℃时,鼠呼吸道CBF基值(x±sx)为(15.6±0.7)Hz(n=5);兔呼吸道CBF基值为(13.2±0.9)Hz(n=7);人鼻腔CBF基值为(13.4±1.1)Hz(n=5),3者间差异无显著性(P>0.05)。当向培养液中加入ATP后,CBF将在数秒内快速升高。结论 高速数字化显微成像和精确的数据处理技术,能够准确反映呼吸道纤毛运动状况,为客观评价呼吸道黏液纤毛传输功能提供临床实用指标。 | 张罗 韩德民 王鸿 周兵 Michael J.Sanderson | 2004 | 中华耳鼻咽喉科杂志2004,39,7: | 15 |
| 2 | Evolutionary dynamics of leucine‐rich repeat receptor‐like kinases and related genes in plants:A phylogenomic approach显示文摘Leucine‐rich repeat(LRR) receptor‐like kinases(RLKs), evolutionarily related LRR receptor‐like proteins(RLPs) and receptor‐like cytoplasmic kinases(RLCKs) have important roles in plant signaling, and their gene subfamilies are large with a complicated history of gene duplication and loss. In three pairs of closely related lineages, including Arabidopsis thaliana and A. lyrata(Arabidopsis), Lotus japonicus,and Medicago truncatula(Legumes), Oryza sativa ssp. japonica,and O. sativa ssp. indica(Rice), we find that LRR RLKs comprise the largest group of these LRR‐related subfamilies, while the related RLCKs represent the smallest group. In addition,comparison of orthologs indicates a high frequency of reciprocal gene loss of the LRR RLK/LRR RLP/RLCK subfamilies.Furthermore, pairwise comparisons show that reciprocal gene loss is often associated with lineage‐specific duplication(s) in the alternative lineage. Last, analysis of genes in A. thaliana involved in development revealed that most are highly conserved orthologs without species‐specific duplication in the two Arabidopsis species and originated from older Arabidopsis‐specific or rosid‐specific duplications. | 3 | Bioinformatic analysis of chromatin organization and biased expression of duplicated genes between two poplars with a common whole-genome duplication显示文摘The nonrandom three-dimensional organization of chromatin plays an important role in the regulation of gene expression.However,it remains unclear whether this organization is conserved and whether it is involved in regulating gene expression during speciation after whole-genome duplication(WGD)in plants.In this study,high-resolution interaction maps were generated using high-throughput chromatin conformation capture(Hi-C)techniques for two poplar species,Populus euphratica and Populus alba var.pyramidalis,which diverged~14 Mya after a common WGD.We examined the similarities and differences in the hierarchical chromatin organization between the two species,including A/B compartment regions and topologically associating domains(TADs),as well as in their DNA methylation and gene expression patterns.We found that chromatin status was strongly associated with epigenetic modifications and gene transcriptional activity,yet the conservation of hierarchical chromatin organization across the two species was low.The divergence of gene expression between WGD-derived paralogs was associated with the strength of chromatin interactions,and colocalized paralogs exhibited strong similarities in epigenetic modifications and expression levels.Thus,the spatial localization of duplicated genes is highly correlated with biased expression during the diploidization process.This study provides new insights into the evolution of chromatin organization and transcriptional regulation during the speciation process of poplars after WGD. | Le Zhang Jingtian Zhao Hao Bi Xiangyu Yang Zhiyang Zhang Yutao Su Zhenghao Li Lei Zhang Brian J.Sanderson Jianquan Liu Tao Ma | 2021 | Horticulture Research2021,8,1: | 2 |
