| 3 | Indirect printing of hierarchical patient-specific scaffolds for meniscus tissue engineering显示文摘The complex meniscus tissue plays a critical role in the knee. The high susceptibility to injury has led to an intense pursuit for better tissue engineering regenerative strategies, where scaffolds play a major role. In this study, indirect printed hierarchical multilayered sca ffolds composed by a silk fibroin (SF) upper layer and an 80/20 (w/w) ratio of SF/ionic-doped β-tricalcium phosphate (TCP) bottom layer were developed. Furthermore, a comparative analysis between two types of sca ffolds pro- duced using di fferent SF concentrations, i.e., 8% (w/v) (Hi8) and 16% (w/v) (Hi16) was performed. In terms of architecture and morphology, the produced sca ffolds presented homogeneous porosity in both layers and no di fferences were observed when comparing both sca ffolds. A decrease in terms of mechanical performance of the sca ffolds was observed when SF concentration decreased from 16 to 8% (w/v). Hi16 revealed a static compressive modulus of 0.66 ± 0.05 MPa and dynamical mechanical properties ranging from 2.17 ± 0.25 to 3.19 ± 0.38 MPa. By its turn, Hi8 presented a compressive modulus of 0.27 ± 0.08 MPa and dynamical mechanical properties ranging from 1.03 ± 0.08 MPa to 1.56 ± 0.13 MPa. In vitro bioactivity studies showed formation of apatite crystals onto the surface of Hi8 and Hi16 bottom layers. Human meniscus cells (hMCs) and human primary osteoblasts were cultured separately onto the top layer (SF8 and SF16) and bottom layer (SF8/TCP and SF16/TCP) of the hierarchical sca ffolds Hi8 and Hi16, respectively. Both cell types showed good adhesion and proliferation as denoted by the live/dead staining, Alamar Blue assay and DNA quanti fication analysis. Subcutaneous implantation in mice revealed weak in flammation and sca ffold’s integrity. | 10 | A multi-scale modelling framework to guide management of plant invasions in a transboundary context显示文摘Background:Attention has recently been drawn to the issue of transboundary invasions,where species introduced and naturalized in one country cross international borders and become problematic in neighbouring countries.Robust modelling frameworks,able to identify the environmental drivers of invasion and forecast the current and future potential distribution of invasive species,are needed to study and manage invasions.Limitations due to the lack of species distribution and environmental data,or assumptions of modelling tools,often constrain the reliability of model predictions.Methods:We present a multiscale spatial modelling framework for transboundary invasions,incorporating robust modelling frameworks(Multimodel Inference and Ensemble Modelling) to overcome some of the limitations.The framework is illustrated using Hakea sericea Schrad.(Proteaceae),a shrub or small tree native to Australia and invasive in several regions of the world,including the Iberian Peninsula.Two study scales were considered:regional scale(western Iberia,including mainland Portugal and Galicia) and local scale(northwest Portugal).At the regional scale,the relative importance of environmental predictors sets was evaluated and ranked to determine the main general drivers for the species distribution,while the importance of each environmental predictor was assessed at the local scale.The potential distribution of H.sericea was spatially projected for both scale areas.Results:Model projections for western Iberia suggest that a large area is environmentally suitable in both Portugal and Spain.Climate and landscape composition sets were the most important determinants of this regional distribution of the species.Conversely,a geological predictor(schist lithology) was more important in explaining its local-scale distribution.Conclusions:After being introduced to Portugal,H.sericea has become a transboundary invader by expanding in parts of Galicia(Spain).The fact that a larger area is predicted as environmentally suitable in Spain raises concerns regarding its potential continued expansion.This highlights the importance of transboundary cooperation in the early management of invasions.By reliably identifying drivers and providing spatial projections of invasion at multiple scales,this framework provides insights for the study and management of biological invasions,including the assessment of transboundary invasion risk. | Joao Martins David M.Richardson Renato Henriques Elizabete Marchante Hélia Marchante Paulo Alves Mirijam Gaertner Joao P.Honrado Joana R.Vicente | 2016 | Forest Ecosystems2016,3,4: | 0 |

| 12 | Endoscopic balloon dilation of crohn's disease stricturessafety,efficacy and clinical impact显示文摘AIM To evaluate the incidence of anastomotic strictures after intestinal resection in Crohn's disease(CD), demonstrate long-term efficacy and safety of endoscopic balloon dilation(EBD) in CD strictures and its impact on the diagnosis of subclinical postoperative endoscopic recurrence. METHODS Retrospective single tertiary center study based on prospectively collected data between 2010 and 2015including anastomotic and non-anastomotic strictures. RESULTS29% of 162 CD patients included developed an anastomotic stricture. 43 patients with anastomotic strictures and 37 with non-anastomotic strictures underwent EBD; technical success was 97.7% and 100%, respectively, however, 63% and 41% needed repeat dilation during the 4.4-year follow-up. Longer periods between surgery and index colonoscopy and higher lactoferrin levels were associated with the presence of stricture after surgery. Calprotectin levels > 83.35 μg/g and current or past history of smoking were associated with a shorter time until need for dilation(HR = 3.877, 95%CI: 1.480-10.152 and HR = 3.041, 95%CI: 1.213-7.627). Anastomotic strictures had a greater need for repeat dilation(63% vs 41%, P = 0.047). No differences were found between asymptomatic and symptomatic cohorts. Disease recurrence diagnosis was only possible after EBD in a third of patients. CONCLUSION EBD is an effective and safe alternative to surgery, with a good short and long-term outcome, postponing or even avoiding further surgery.
