| 1 | Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. | 2 | A PIF7-CONST ANS-Centered Molecular Regulatory Network Underlying Shade-Accelerated Flowering显示文摘To compete with their neighbors for light and escape shaded environments,sun-loving plants have devel-oped the shade-avoidance syndrome(SAS),a set of responses including alteration of plant architecture and initiation of early flowering and seed set.Previous studies on SAS mainly focused on dissecting molec-ular basis of hypocotyl elongation in seedlings under shade light;however,the molecular mechanisms underlying shade-accelerated flowering in adult plants remain unknown.In this study,we found that CONSTANS(CO)and PHYTOCHROME-INTERACTING FACTOR 7(PIF7)have an additive effect on shade-induced flowering,but that LONG HYPOCOTYL IN FAR-RED1(HFR1)represses early flowering by binding to CO and PIF7 and preventing the binding of CO to the promoter of FLOWERING LOCUS T(FT)and the binding of PIF7 to the promoter of pri-MIR156E/F.Under shade,de-phosphorylated PIF7 and accumulated CO,balanced by HFR1,upregulate the expression of FT,TSF,SOC1,and SPLs to accelerate flowering.Moreover,we found that the function of PIF7 in flowering time is independent of phyA.Collectively,these regulatory interactions establish a crucial link between the light signal and genetic network that regulates flowering transition under shade. | Renshan Zhang Chuanwei Yang Yupei Jiang Lin Li | 2019 | Molecular Plant2019,12,12: | 5 |
| 3 | Effects of Community Community Communication Technology(ICT) Usage on Community Satisfaction: Case Study in Nanjing, China显示文摘Future or smart community, which mainly refers to the development of community information and communication technology(ICT) platforms or devices, has received considerable attention from urban governments and scholars. However, only a few studies have been conducted to test the actual effects of using these community ICT platforms or devices on the community satisfaction of residents. Therefore, the present study conducts a survey in 40 communities in Nanjing, China and uses a mixed linear regression model to determine the relationship between community ICT usage and community satisfaction. Results indicate that residents with high-level community ICT usage are more satisfied with their community than those with low-level community ICT usage. Moreover, evident differences are observed regarding the influence of new commodity, old commodity and affordable housing communities in Nanjing. | 4 | Evolution Process of Urban Spatial Pattern in Hubei Province Based on DMSP/OLS Nighttime Light Data显示文摘This paper intends to investigate the urban spatial patterns of Hubei Province and its evolution from three different perspectives: urban nodes, urban connections and urban clusters. The research adopts nighttime light imagery of cities in Hubei Province, the viewpoint of ′point-axis-area′ in the ′point-axis system′ theory, and employs light index model, gravity model and social network analysis. The findings are as follows: 1) In terms of urban nodes, the urbanization process of Hubei has been carried out mainly on the basis of external expansion rather than internal increasing. The polarization trend of urban connection network is strengthening. 2) As for urban connections, the estimation of urban connections using light index model is capable of containing various actual flow, and the connections are getting increasingly closer. 3) In regard to urban groups, seven urban groups of varying sizes have formed. On that basis, three stable and relatively independent urban groups as the centers, namely Wuchang, Yichang and Xiangyang emerge as well. | 8 | Notch signaling inhibitor and anti-PD-L1 antibody combination therapies decelerate tumor progression in pancreatic cancer显示文摘Objective:Pancreatic cancer(PC)is a highly lethal malignancy with an immunosuppressive environment.Yet,current immune checkpoint inhibitor monotherapies have shown limited efficacy in PC,prompting the need for combination therapies.Herein,we hypothesized that combinations of Notch signaling inhibitor and anti-ligand programmed death-ligand 1(PD-L1)antibody immunotherapy would show synergistic efficacy.Methods:The baseline expression of PD-L1 and HES1 was measured in PC cell lines,single-cell RNA-seq data of PC(GSA:CRA001160),and cBioPortal databases.In an in vitro study,MIA PaCa2 and SW1990 were used to explore the mechanism between Notch signaling and PD-L1.To study the effects in vivo,a subcutaneous tumor model was established using Pan02 cells treated with either anti-PD-L1 monoclonal antibody and/or Notch inhibitor DAPT.The study performed involving human samples was approved by the Ethics Committee of Peking Union Medical College Hospital(approval No.S-K460,approval date:April 23,2018).Animal studies were approved by the Animal Research Ethics Committee of Peking Union Medical College Hospital(approval No.XHDW-2019-049,approval date:November 28,2019).Results:The Notch signaling inhibitor upregulated PD-L1 expression in PC tumor cells both in vitro and in vivo.Notch effector HES1 knockdown produced PD-L1 upregulation in both MIA PaCa2 and SW1990 cells.Combined DAPT and anti-PD-L1 antibody treatment of Pan02 subcutaneous tumor model resulted in significantly reduced tumor weights compared to that with monotherapy,as well as significantly reduced Ki67 than that in the monotherapy group and control group.Flow cytometry analysis revealed significantly increased CD8+T cell infiltration in tumors of the combination group compared with those of the monotherapy group.Conclusion:Notch signaling blockade might enhance the antitumor effect of anti-PD-L1 therapy in PC. | Hongmei Dai Xiafei Hong Dan Huang Huanwen Wu Xianze Wang Hao Chen Rui Jiang Wenyan Chen Yupei Zhao Wenming Wu | 2021 | Journal of Pancreatology2021,4,2: | 0 |
