| 1 | Human RBCs blood group conversion from A to O using a novel α-N-acetylgalactosaminidase of high specific activity显示文摘α-N-acetylgalactosaminidase (αNAGA) can convert group A human red blood cells (RBCs) to group O. One novel αNAGA gene was cloned by PCR from Elizabethkingia meningosepticum isolated from a domestic clinical sample. Pure recombinant αNAGA was obtained by genetic engineering and protein purification with a calculated molecule of 49.6 kD. αNAGA was selective for terminal α-N-acetylgalacto-samine residue with a high specific activity. αNAGA could completely remove A antigens of 1 U (about 100 mL) group A1 or A2 RBCs in 1 h at pH 6.8 and 25℃ with a consumption of 1.5 or 0.4 mg recombinant enzyme. Enzyme-converted group A RBCs did not agglutinate after being mixed with monoclonal anti-A or sera of groups A,B,AB and O. Other blood group antigens except ABO had no change. FCM analy-sis showed that A antigens and A1 antigens disappeared while H antigens increased. It indicated that αNAGA successfully converted human blood group A RBCs to universally transfusable group O RBCs without the risk of ABO-incompatible transfusion reactions. | 2 | A novel method for preparation of tissue microarray显示文摘AIM:To improve the technique of tissue microarray (tissue chip).METHODS: A new tissue microarraying method was invented with a common microscope installed with a special holing needle, a sampling needle, and a special box fixing paraffin blocks on the microscope slide carrier. | 3 | Tumor-specific gene expression patterns with gene expression profiles显示文摘Gene expression profiles of 14 common tumors and their counterpart normal tissues were analyzed with machine learning methods to address the problem of selection of tumor-specific genes and analysis of their differential expressions in tumor tissues. First, a variation of the Relief algorithm, 'RFE_Relief algorithm' was proposed to learn the relations between genes and tissue types. Then, a support vector machine was employed to find the gene subset with the best classification performance for distinguishing cancerous tissues and their counterparts. After tissue-specific genes were removed, cross validation experiments were employed to demonstrate the common deregulated expressions of the selected gene in tumor tissues. | 12 | Threshold-Based Adaptive Gaussian Mixture Model Integration(TA-GMMI)Algorithm for Mapping Snow Cover in Mountainous Terrain显示文摘Snow cover is an important parameter in the fields of computer modeling,engineering technology and energy development.With the extensive growth of novel hardware and software compositions creating smart,cyber physical systems’(CPS)efficient end-to-end workflows.In order to provide accurate snow detection results for the CPS’s terminal,this paper proposed a snow cover detection algorithm based on the unsupervised Gaussian mixture model(GMM)for the FY-4A satellite data.At present,most snow cover detection algorithms mainly utilize the characteristics of the optical spectrum,which is based on the normalized difference snow index(NDSI)with thresholds in different wavebands.These algorithms require a large amount of manually labeled data for statistical analysis to obtain the appropriate thresholds for the study area.Consideration must be given to both the high and low elevations in the study area.It is difficult to extract all snow by a fixed threshold in mountainous and rugged terrains.In this research,we avoid relying on a manual analysis for different elevations.Therefore,an algorithm based on the GMM is proposed,integrating the threshold-based algorithm and the GMM.First,the threshold-based algorithm with transferred thresholds from other satellites’analysis results are used to coarsely classify the surface objects.These results are then used to initialize the parameters of the GMM.Finally,the parameters of that model are updated by an expectation-maximum(EM)iteration algorithm,and the final results are outputted when the iterative conditions end.The results show that this algorithm can adjust itself to mountainous terrain with different elevations,and exhibits a better performance than the threshold-based algorithm.Compared with orbit satellites’snow products,the accuracy of the algorithm used for FY-4A is improved by nearly 2%,and the snow detection rate is increased by nearly 6%.Moreover,compared with microwave sensors’snow products,the accuracy is increased by nearly 3%.The validation results show that the proposed algorithm can be adapted to a complex terrain environment in mountainous areas and exhibits good performance under a transferred threshold without manually assigned labels. | Yonghong Zhang Guangyi Ma Wei Tian Jiangeng Wang Shiwei Chen | 2020 | Computer Modeling in Engineering & Sciences2020,,9: | 0 |
| 13 | Snow Cover Mapping for Mountainous Areas by Fusion of MODIS L1B and Geographic Data Based on Stacked Denoising Auto-Encoders显示文摘Snow cover plays an important role in meteorological and hydrological researches.However,the accuracies of currently available snow cover products are significantly lower in mountainous areas than in plains,due to the serious snow/cloud confusion problem caused by high altitude and complex topography.Aiming at this problem,an improved snow cover mapping approach for mountainous areas was proposed and applied in Qinghai-Tibetan Plateau.In this work,a deep learning framework named Stacked Denoising Auto-Encoders(SDAE)was employed to fuse the MODIS multispectral images and various geographic datasets,which are then classified into three categories:Snow,cloud and snow-free land.Moreover,two independent SDAE models were trained for snow mapping in snow and snow-free seasons respectively in response to the seasonal variations of meteorological conditions.The proposed approach was verified using in-situ snow depth records,and compared to the most widely used snow products MOD10A1 and MYD10A1.The comparison results show that our method got the best performance:Overall accuracy of 98.95%and F-measure of 73.84%.The results indicated that our method can effectively improve the snow recognition accuracy,and it can be further extended to other multi-source remote sensing image classification issues. | Xi Kan Yonghong Zhang Linglong Zhu Liming Xiao Jiangeng Wang Wei Tian Haowen Tan | 2018 | Computers, Materials & Continua2018,,10: | 0 |
